Dit artikel beschrijft twee hardnekkige murine wond modellen die het mogelijk maken voor de beoordeling van de acute wondgenezing reactie. De eerste methode omvat de chirurgische implantatie van PVA sponzen in de dorsale onderhuidse ruimte. Deze aanpak biedt een duidelijk voordeel ten opzichte van biopsie-gebaseerde wondmodellen voor het bestuderen van de cellulaire wondgenezingsrespons vanwege het grote aantal cellen en de hoeveelheid wondvloeistoffen verkregen uit de geïsoleerde sponzen. Er zijn veel murine model systemen beschikbaar om wondgenezing processen te onderzoeken, elk met specifieke voordelen en beperkingen1,2. De volgende methoden presenteren twee murinewondmodellen, die elk een bepaald aspect van de wondgenezingsrespons aandeorde, en die kunnen worden gebruikt om de oorzaak en het effect van verstoringen in de reactie op letsel te identificeren. Het proces van wondgenezing vindt plaats in verschillende fasen. De eerste fase is inflammatoir, gekenmerkt door de snelle instroom van bloedplaatjes, neutrofielen en monocyten/macrofagen, evenals de productie van ontstekingscytokines en chemokines. Na de oplossing van ontsteking, de omgeving overgangen naar een meer herstellende toestand met de inductie van profibrotische en proangiogene cytokines en groeifactoren. Granulatieweefsel wordt afgezet en neovessels vormen zich met de migratie van myofibroblasten, fibroblasten, epitheelcellen en endotheelcellen. In de laatste fase wordt de voorlopige extracellulaire matrix gerenoveerd en verloopt littekenvorming en wondsluiting2,3,4,5,6,7,8.
Geen enkel murinemodel biedt een systeem om alle stadia van wondgenezing te bestuderen2. Hier worden twee chirurgische wondmodellen beschreven: de ene verduidelijkt acute cellulaire en cytokinewondgenezingsreacties, en de andere maakt de beoordeling van wondsluiting en histologische analyses mogelijk. Deze twee methoden kunnen op complementaire wijze worden gebruikt om de effecten van een verstoring of comorbiditeit op verschillende aspecten van de wondgenezingsrespons te beoordelen. De dorsale onderhuidse implantatie van polyvinyl alcohol (PVA) sponzen is een systeem dat is gebruikt in knaagdiermodellen voor decennia tot tal van aspecten van cellulaire en granulatie weefsel reacties9,,10,11,12,14,,15,16,17,18,19,20,1420 ,21,22,23,24. Deze aanpak maakt het ophalen van cytokine-rijke wondvloeistoffen en cellulaire infiltreert. In dit model worden 1 cm x 1 cm x 0,5 cm stukken PVA-spons in onderhuidse zakken geplaatst door middel van een incisie van 2 cm gemaakt aan de achterste ruglijn. De incisie wordt gesloten met chirurgische clips, en de sponzen kunnen worden opgehaald op latere tijd punten voor cel-en vloeistofisolatie. Het cellulaire en cytokine-milieu van geïsoleerde sponzen weerspiegelt de normale stadia van acute wondgenezing tot ongeveer 14 dagen na implantatie. Op latere tijdpunten is het model voordeliger voor het bestuderen van granulatieweefselvorming en de vreemde lichaamsrespons1. Met dit systeem is het mogelijk om >106 cellen te isoleren, wat een duidelijk voordeel biedt voor fenotypische en functionele testen en RNA-isolatie, over het isoleren van cellen van andere biopsie-gebaseerde methoden1,22,23,25,26.
De snelheid van wondsluiting wordt bepaald met behulp van het staarthuidexcisiemodel. In dit model, zoals in eerste instantie beschreven door Falanga et al. en gemeld door anderen27,28,29,30, een 1 cm x 0,3 cm volledige dikte sectie van de staart huid wordt verwijderd in de buurt van de basis van de staart. Het wondgebied wordt gemakkelijk gevisualiseerd en kan na verloop van tijd worden gemeten. Als alternatief kan staartweefsel worden geïsoleerd voor histologische analyse. Deze aanpak kan worden gebruikt als een alternatief voor of in combinatie met de gevestigde dorsale punch biopsie methode. Het primaire onderscheid tussen deze twee modellen zijn de snelheid van wondsluiting, de aan- of afwezigheid van bont en de huidstructuur2,31,32. Staarthuidwonden bieden een langer tijdsbestek om de wondsluiting te beoordelen, omdat het ongeveer 21 dagen duurt voordat de volledige sluiting optreedt. Dit is in tegenstelling tot ongespkal dorsale punch biopten, die veel sneller genezen (~ 7-10 dagen), voornamelijk door contractie als gevolg van de werking van de panniculus carnosus. Spalkd rugstompbiopten genezen langzamer en verminderen de effecten van contractiele genezing, maar vertrouwen op de aanwezigheid van een vreemd lichaam om contractiele mechanismen te beperken1,2,27,30,31,33.
De beschreven wondmodellen zijn informatief voor het begrijpen van normale wondgenezingsprocessen bij afwezigheid van verstoring. Terwijl de genezing van knaagdierhuid op zeer significante manieren verschilt van de menselijke huid, waaronder losse structuur, afhankelijkheid van contractiele genezing en andere anatomische verschillen, biedt het murinesysteem bepaalde voordelen voor mechanistische en screeningsstudies. Belangrijkste onder deze is de beschikbaarheid van inteelt stammen en genetische mutanten, genetische traktaat, en lagere kosten. Mechanistisch inzicht dat is opgedaan met murinestudies kan worden vertaald naar complexe diermodellen die de genezing van de menselijke huid, zoals het varkenssysteem2,,31,beter nabootsen.
Naast het onderzoeken van wondgenezingsreacties in de steady state, kunnen deze modellen worden gecombineerd met comorbide aandoeningen om de basis van wondgenezingsdefecten op cellulair, cytokine en brutoweefselniveau te begrijpen. Het is in deze specifieke setting dat de twee modellen in overleg kunnen worden gebruikt om de effecten van een bepaalde comorbide aandoening, zoals postoperatieve longontsteking, te beoordelen op zowel de acute cellulaire wondgenezingsrespons als de snelheid van wondsluiting30.

Wondgenezing is een complex proces dat de ordelijke progressie van ontsteking, granulatieweefselvorming, fibrose en resolutie vereist. Murinemodellen bieden waardevol mechanistisch inzicht in deze processen; echter, geen enkel model volledig adressen alle aspecten van de wondgenezing reactie. In plaats daarvan is het ideaal om meerdere modellen te gebruiken om de verschillende aspecten van wondgenezing aan te pakken. Hier worden twee verschillende methoden beschreven die betrekking hebben op diverse aspecten van de wondgenezingsrespons. In het eerste model worden polyvinyl alcoholsponzen onderhuids geïmplanteerd langs de muisdorsum. Na het ophalen van spons, cellen kunnen worden geïsoleerd door mechanische verstoring, en vloeistoffen kunnen worden geëxtraheerd door centrifugering, waardoor een gedetailleerde karakterisering van cellulaire en cytokine reacties in de acute wond omgeving. Een beperking van dit model is het onvermogen om de snelheid van wondsluiting te beoordelen. Hiervoor wordt gebruik gemaakt van een staarthuidexcisiemodel. In dit model wordt een rechthoekig stuk staarthuid van 10 mm x 3 mm uitgesneden langs het rugoppervlak, in de buurt van de basis van de staart. Dit model kan gemakkelijk worden gefotografeerd voor planimetrische analyse om genezingtarieven te bepalen en kan worden weggesneden voor histologische analyse. Beide beschreven methoden kunnen worden gebruikt in genetisch veranderde muizenstammen, of in combinatie met modellen van comorbide aandoeningen, zoals diabetes, veroudering of secundaire infectie, om wondgenezingsmechanismen op te helderen.

Systemische ontstekingsreactie na PVA sponsimplantatie De PVA spons implantatie operatie genereerde een systemische ontstekingsreactie, zoals blijkt uit de inductie van IL-6 in het plasma 1 dag na het verwonden (Figuur 2A). Deze modellen kunnen worden geïmplementeerd om verschillende aspecten van de wondgenezingsrespons te beoordelen, waaronder cellulaire en cytokinetiek en wondsluiting. De PVA spons model maakt het herstel van miljoenen wond leukocyten voor fenotypische en functionele analyses, terwijl de staart huid excisie model maakt de gemakkelijke visualisatie van langzaam genezende wonden. De PVA spons en excisional staart wond modellen kunnen worden gebruikt in combinatie met comorbide voorwaarden, zoals diabetes of longontsteking, om hun impact op wondgenezing te begrijpen.Het aantonen van de procedures met Meredith Crane zal Bill Henry zijn, een onderzoeksassistent van mijn lab. Gebruik voor het begin van het experiment een schaar om vellen PVA-spons in stukken van acht bij acht bij vier millimeter te snijden. Rehydrateer de spons stukken in steriele PBS in een bekerglas gedurende 10 minuten voor autoclave steriliseren van de sponzen in de PBS.Wanneer de sponzen zijn afgekoeld, bewaar de sponzen ondergedompeld in PBS op vier graden Celsius. Op de dag van de procedure, plaats zes sponzen per proefdier in een steriele cultuur schotel in een steriele laminaire flow kap, en bevestigen een gebrek aan reactie op teen knijpen in een verdoofde, acht-tot 12-week-oude, man, C57-Black/6 muis. Laat een assistent clippers gebruiken om het haar langs de dorsum en steriel gaas te verwijderen om povidone-jodium oplossing toe te passen op het geschoren gebied twee keer, gevolgd door een enkele 70% ethanol toepassing.De assistent moet de muis vervolgens overbrengen op steriele chirurgische gordijnen geplaatst over een verwarmd pad. Het dragen van steriele handschoenen, gebruik tangen om de rughuid weg te trekken van het onderliggende weefsel, en gebruik steriele chirurgische schaar om een twee centimeter incisie langs de dorsale middellijn ongeveer twee centimeter voorste naar de basis van de staart te maken. Houd de incisie open met steriele tangen, gebruik steriele, gebogen, stompe kant een chirurgische schaar om een onderhuidse zak langs de dorsum te vormen in een van de posities zoals aangegeven in de figuur.Zodra de schaar zijn ingevoegd, open en sluit de tips twee keer om een zak groot genoeg om een ingevoegde spons te houden vormen. Wanneer de zak is gemaakt, gebruik steriele chirurgische schaar om voorzichtig knijp een PVA spons in de cultuur schotel om overtollige PBS te verwijderen. Til vervolgens de spons door een hoek, en, leidt met de hoek gehouden door de schaar, plaats de spons in de onderhuidse zak.Wanneer alle zes sponzen zijn geplaatst zoals aangetoond, gebruik steriele tangen om de ingesneden rughuid samen te knijpen, en sluit de incisie met twee roestvrijstalen wondclips. Voor PVA spons vloeistof isolatie, een tot 14 dagen na implantatie, verwijder de chirurgische nietjes, en open de incisie met getande tangen. Gebruik een schaar om de incisie uit te breiden langs de dorsale middellijn, en gebruik tangen om een spons te halen uit zijn onderhuidse zak.Plaats de spons in het vat van een vijf-milliliter spuit genesteld in een 16-milliliter cultuur buis op ijs, en gebruik schaar om elk bindweefsel dat blijft aanhangen aan het oppervlak van de spons, indien nodig los te koppelen. Wanneer alle sponzen zijn geëxtraheerd en overgebracht zoals aangetoond, centrifugeren de cultuur buis om de wond vloeistof te verzamelen. Om de cellen te isoleren van de PVA sponzen, na het verzamelen van de sponzen zoals net aangetoond, plaats ze in een 15-milliliter conische buis met vijf milliliter hbss collectie medium.Wanneer alle sponzen zijn verzameld, decanteren de spons schorsing in een 80-milliliter blender zak, en hang de zak van het luik van de peddel blender, zodat de peddels zal de sponzen en medium staking. Stel de peddelblender in om 60 seconden op hoog te draaien en druk op start. Wanneer de blender is gestopt, knijp de sponzen in de zak om het medium volledig los te laten, en gebruik een pipet om het medium van de blender zak terug te brengen in de 15-milliliter buis.Pas de instellingen van de peddelblender aan om 30 seconden op hoog te lopen en voeg vijf milliliter medium toe aan de blenderzak. Herhaal het maagproces nog twee keer voordat u de buis van verzameld mediumcentrifugeert. Een rode pellet van cellen en rode bloedcellen moet zichtbaar zijn aan de onderkant van de conische buis.Meng 900 microliter gedestilleerd water met de cellen gedurende drie tot vijf seconden voordat u de lyse neutraliseert met 100 microliter van 10x PBS en vier milliliter 1x PBS. Verzamel de cellen door centrifugatie, en resuspend de witte cel pellet in het juiste medium voor downstream-analyse. Om een staarthuidwond te creëren, gebruik steriel gaas na bevestiging van een gebrek aan respons op teensnuifje in de verdoofde, acht-tot twaalf weken oude, man, C57-Black/6 muis, steriel gaas om povidone-jodiumoplossing twee keer toe te passen op de chirurgische site, gevolgd door één toepassing van 70%ethanol.Met behulp van een permanente marker en een vooraf gemaakte sjabloon, traceer een 10-bij-drie-millimeter sectie op het dorsale oppervlak van de staart, 10 millimeter van de staart basis, en plaats de muis op steriele chirurgische gordijnen. Het dragen van steriele chirurgische handschoenen, gebruik maken van een steriele scalpel mes om een volledige dikte incisie langs de rechter-, onderkant en linkerranden van de wond gebied te maken. Met behulp van steriele tangen, schil de afgeslepen huid uit de buurt van de staart, en gebruik steriele chirurgische schaar weg te snijden de bovenste rand van het wondgebied.Gebruik steriel gaas om druk uit te oefenen op de wond om het bloeden te stoppen en breng een spray barrièrefilm aan op het wondbed. Fotografeer vervolgens de wonden van een vaste afstand op regelmatige tijdstippen en analyseer de foto's op planimetrische analyse om de metingen van het wondgebied te bepalen. PVA spons implantatie chirurgie genereert een systemische ontstekingsreactie, zoals blijkt uit de inductie van IL-6 in het plasma een dag na het verwonden.Het aantal cellen dat kan worden teruggewonnen uit PVA spons wonden toeneemt na verloop van tijd, met neutrofielen, monocyten, en macrofagen bestaande uit de primaire cellulaire infiltreren populaties in de sponzen. Neutrofielen worden geïdentificeerd als Ly6G-positieve, Siglec-F-negatieve cellen. Siglec-F-positieve cellen zijn voornamelijk eosinofielen.Gating op de Ly6G-negatieve, Siglec-F-negatieve cellen maakt de identificatie van F4/80-positieve monocyten en macrofagen. F4/80-positieve cellen kunnen verder worden onderscheiden door hun Ly6C expressie om Ly6C hoge ontstekingsmonocyten en Ly6C lage monocyten-afgeleide macrofagen te onderscheiden. De excisional staart huid model biedt een alternatief voor de dorsale huid punch biopsie methode om wond sluiting in stevige huid ontbreekt dichte vacht te bestuderen.Wondsluiting kan worden gekwantificeerd door het gebied van het wondbed na verloop van tijd te meten. De staart huid wond kan ook worden waargenomen in dwarsdoorsnede door histologische analyse via H en E en Masson's trichrome kleuring. In deze beelden worden de zijdelingse marges van de uitgesneden huid aangegeven door de pijlpunten op het rugoppervlak van de staart.Om te begrijpen steady-state wond genezing, therapeutische verbindingen of interventies kunnen ook worden getest door hun injectie rechtstreeks in de PVA spons of door levering aan de staart wond bed.
