Geprogrammeerde celdood in erytrocyten, ook bekend als eryptosis, is gebruikelijk in vele klinische aandoeningen en hematologische aandoeningen. Eryptosis wordt geassocieerd met celkrimp en het verlies van fosfolipide asymmetrie in de cel plasma membraan1,2. Verlies van asymmetrie resultaten in de translocatie van fosfatidylserine (PS), een lipide normaal gelokaliseerd in de binnenste bijsluiter3,4, naar de buitenste bijsluiter van de cel, die signalen naar macrofagen aan fagocytose en verwijderen defecte erytrocyten5,6,7,8. Aan het einde van de normale levensduur van erytrocyten zorgt het verwijderen van eryptotische cellen door macrofagen voor het evenwicht van erytrocyten in omloop. Echter, in zieke omstandigheden, zoals sikkelcelziekte en thalassemie9,10,11, verbeterde eryptosis kan resulteren in ernstige anemie2. Vanwege het belang in hematologische ziekten, is er belangrijke belangstelling voor het onderzoeken van de factoren inducerende of remmende eryptosis en de moleculaire mechanismen die aan dit proces ten grondslag liggen.
Het plasma membraan van gezonde erytrocyten is asymmetrisch, met verschillende fosfolipiden lokaliseren op de buitenste en binnenste folders. Membraan asymmetrie wordt voornamelijk gereguleerd door de werking van membraan enzymen. Aminopfosfolipide translocase vergemakkelijkt het transport van aminophospholipiden, PS en fosfatidylethanolamine (PE), door deze lipiden naar de binnenste folder van de cel te sturen. Aan de andere kant, floppase vervoert de choline met fosfolipiden, dunlaag (PC) en sphingomyelinase (SM), van de binnenste naar de buitenste bijsluiter van de cel membraan12. Echter, in tegenstelling tot gezonde cellen, het membraan van eryptotic erytrocyten is vervormd. Dit komt door de werking van een derde enzym, scramblase, dat de fosfolipide asymmetrie verstoort door het bidirectionele transport van aminophospholipiden13,14,15,16te vergemakkelijken. Scramblase wordt geactiveerd door verhoogde intracellulaire niveaus van CA2 +. Daarom zijn calcium-ionoforen, die het transport van CA2 + over het celmembraan12faciliteren, efficiënte inductoren van eryptosis.
Ionomycine, een calcium-ionophore, is op grote schaal gebruikt voor het opwekken van eryptosis in erytrocyten12,17,18,19,20,21,22,23,24,25,26. Ionomycine heeft zowel hydrofiele als hydrofobe groepen, die nodig zijn om ca2 + Ion te binden en te vangen, en deze te transporteren naar de cytosolische ruimte27,28,29. Dit leidt tot de activering van de scramblase en translocatie van PS naar de buitenste bijsluiter, die gemakkelijk kan worden gedetecteerd met behulp van annexin-V, een cellulair eiwit met een hoge affiniteit voor PS12. Hoewel het triggeren van eryptosis door ionomycine vaak wordt gerapporteerd, is er een aanzienlijke methode discrepantie in de literatuur (tabel 1). De populatie van erytrocyten die eryptosis ondergaan, hangt af van verschillende factoren zoals ionofiele concentratie, behandelingstijd met ionofoor, en het suikergehalte van extracellulaire omgeving (glucose depletie activeert catie kanalen en vergemakkelijkt de intrede van CA2 + in de cytosolische ruimte)30,31. Er is echter weinig consistentie in deze factoren in de literatuur, waardoor het moeilijk is om eryptosis reproductief in vitrouit te voeren.
In dit protocol presenteren we een stapsgewijze procedure voor het opwekken van eryptosis in menselijke erytrocyten. Factoren die van invloed zijn op succesvolle eryptosis, waaronder CA2 + concentratie, ionofiele concentratie, behandelingstijd, en pre-incubatie in glucose-verarmd buffer worden onderzocht en optimale waarden worden gerapporteerd. Deze procedure toont aan dat de pre-incubatie van erytrocyten in een glucose vrije buffer het percentage van de eryptosis aanzienlijk verhoogt in vergelijking met de glucose-bevattende buffer. Dit protocol kan in het laboratorium worden gebruikt om eryptotische erytrocyten te produceren voor verschillende toepassingen.

Eryptosis, erytrocyten geprogrammeerde celdood, treedt op in een aantal hematologische ziekten en tijdens letsel aan erytrocyten. Een kenmerk van de eryptotische cellen is het verlies van compositorische asymmetrie van het celmembraan, wat leidt tot de translocatie van fosfatidylserine naar de buitenste bijsluiter van het membraan. Dit proces wordt veroorzaakt door verhoogde intracellulaire concentratie van CA2 +, die activeert scramblase, een enzym dat bidirectionele beweging van fosfolipiden tussen membraan folders vergemakkelijkt. Gezien het belang van eryptosis in verschillende zieke omstandigheden, zijn er inspanningen gedaan om de eryptosis in vitrote induceren. Dergelijke inspanningen hebben zich over het algemeen gebaseerd op het calcium-ionophore, ionomycine, om de intracellulaire CA2 + -concentratie te verbeteren en eryptosis te induceren. Echter, veel discrepanties zijn gemeld in de literatuur met betrekking tot de procedure voor het induceren van eryptosis met behulp van ionomycine. Hierin rapporteren we een stap-voor-stap protocol voor ionomycine-geïnduceerde eryptosis in menselijke erytrocyten. We richten ons op belangrijke stappen in de procedure, waaronder de ionofiele concentratie, incubatietijd en glucose depletie, en geven representatief resultaat. Dit protocol kan worden gebruikt voor het reproduceren van eryptosis in het laboratorium.

Optimalisatie van de ionomycine concentratie

Er zijn veel inspanningen geweest om eryptose te induceren met behulp van ionomycine. De exacte procedure voor dit proces wordt echter niet duidelijk beschreven in de literatuur. Wij bieden een stapsgewijze procedure om eryptose te induceren met behulp van ionomycine.Het belangrijkste voordeel van deze procedure is het op een betrouwbare en reproduceerbare manier induceren van eryptose bij menselijke erytrocyten. Om te beginnen, voeg 500 microliters koud vol bloed in zuurcitraat dextrose toe aan een microcentrifugebuis. Centrifugeer het hele bloed op 700 keer g gedurende vijf minuten bij kamertemperatuur en verwijder het heldere plasma en de dunne buffy jas met behulp van een pipet om de rode erythrocyte laag te verlaten.Bereid een liter Ringer oplossing met 125 millimolar natriumchloride, vijf millimolar kaliumchloride, een millimolar magnesiumsulfaat, 32 millimolar HEPES, vijf millimolar glucose, en een millimolar calciumchloride. Pas de pH aan op 7,4 door twee microliterdruppels van één molaire natriumhydroxide toe te voegen. Om glucosevrije Ringer-oplossing voor te bereiden, volgt u hetzelfde protocol, maar neem geen glucose op in de oplossing.Was de erytrocyten tweemaal in Ringer oplossing door het opschorten van de cel pellet in 1,5 milliliter ringer oplossing, centrifugeren op 700 keer g gedurende vijf minuten bij kamertemperatuur en het verwijderen van de supernatant. Maak vervolgens een 0,4%hematocriet door 40 microliter van de erythrocytenpellet in 9, 960 microliter glucosevrije Ringer-oplossing opnieuw in te stellen om een eindvolume van 10 milliliter te bereiken. Incubeer de celsuspensie op 37 graden Celsius gedurende zeven dagen.Pre-incubatie van erythrocyten in een glucosevrije buffer activeert aanzienlijk eryptose. Hoge eryptose werd verkregen na zeven dagen pre-incubatie in de suikervrije buffer. Los een milligram ionomycine calciumzout op in 630 microliter DMSO om een uiteindelijke concentratie van twee millimolar te bereiken.Aliquot in 20 milliliter en op te slaan bij min 20 graden Celsius. Neem een milliliter van de bereide 0,4%hematocriet en voeg 0,5 microliter van twee millimolar ionomycine toe om een uiteindelijke concentratie van één micromolar te bereiken. Twee uur lang uitbroeden bij 37 graden Celsius.Gebruik een milliliter van de hematocriet zonder ionomycine behandeling als een negatieve controle. Centrifuge de ionomycine behandeld en onbehandelde hematokriten op 700 keer g gedurende vijf minuten bij kamertemperatuur en verwijder hun supernatants om de cel pellets te verlaten aan de onderkant van de buizen. Was de cellen drie keer met Ringer-oplossing door de celpellets op te schorten in 1,5 milliliter Ringer-oplossing, vijf minuten lang te centrifugeren op 700 g bij kamertemperatuur en de supernatants weg te gooien.Om hemolyse te meten, voeg een milliliter van de onbehandelde 0,4%hematocriet toe aan een microcentrifugebuis en incubeer gedurende twee uur bij 37 graden Celsius als negatieve controle van 0%hemolyse. Om de positieve controle van 100% hemolyse voor te bereiden, voeg een milliliter van de onbehandelde 0,4% hematocaan toe aan een microcentrifugebuis en centrifuge bij 700 keer g gedurende vijf minuten bij kamertemperatuur. Verwijder de supernatant en voeg een milliliter gedestilleerd water aan de cel pellet te resuspend en uitbroeden voor twee uur op 37 graden Celsius.Voeg vervolgens een milliliter van de met 0,4% behandelde ionomycine toe aan een microcentrifugebuis. Centrifugeren de onbehandelde cellen, behandelde cellen, en de cellen in gedestilleerd water op 700 keer g gedurende vijf minuten bij kamertemperatuur. Breng 200 microliter van de supernatants over in elke put van een 96-put plaat.Meet de absorptie op 541 nanometer met behulp van een microplaatlezer. Bereken de hemolyse met behulp van de vergelijking waarbij A0, A100 en AT de absorptie van erytrocyten in Ringer-oplossing in water en de absorptie van behandelde erthyrocyten door ionomycine zijn. Verdun twee milliliter van de 5X Annexin-V binding buffer in acht milliliter PBS om 1X bindende buffer te verkrijgen.Resuspend de ionomycine-behandelde en onbehandelde celpellets in één milliliter van de 1X bindingsbuffer. Neem 235 microliter van de celsuspensies in de bindingsbuffer en voeg 15 microliter van Annexin-V Alexa Fluor 488 conjugaat toe in een microcentrifugebuis. Incubeer de cellen bij kamertemperatuur gedurende 20 minuten op een donkere plaats.Centrifuge op 700 keer g gedurende vijf minuten bij kamertemperatuur. Verwijder de supernatant. Was de cellen twee maal met 1X binding buffer door het opschorten van de celpellet in 1,5 milliliter van de binding buffer en centrifugeren op 700 keer g gedurende vijf minuten bij kamertemperatuur.Verwijder de supernatant en resuspend de celpellets in 250 microliters van 1X bindende buffer voor flow cytometrie meting. Breng nu 200 microliter van de met bijlage in-V gekleurde erytrocyten over naar één milliliter ronde polystyreenbuizen die compatibel zijn met stroomcytometrie. Log in op de flow cytometriesoftware en klik op de nieuwe experimentknop.Klik op de nieuwe tube knop. Selecteer de globale plaat en kies de toepassingsanalyse om de fluorescentieintensiteit te meten met een excitatiegolflengte van 488 nanometer en een emissiegolflengte van 530 nanometer. Stel het aantal cellen in op 20.000 dat moet worden verzameld voor fluorescentie geactiveerde celsorteeranalyse.Selecteer de gewenste buis en klik op de laadknop. Klik op de recordknop voor forward scatter en side scatter meting. Herhaal dit voor alle monsters.Klik met de rechtermuisknop op de knop specimen en klik op batchanalyse toepassen om het resultaatbestand te genereren. Klik met de rechtermuisknop op de knop specimen en klik op het genereren van FSC-bestanden. In dit protocol was de behandeling van erytrocyten met één micromolarionomycine- en Ringeroplossing gedurende twee uur voldoende om eryptose te veroorzaken, zoals blijkt uit succesvolle etikettering met annexin-V Fluor 488 conjugaat en kwantificering door facs-analyse.Hogere concentraties ionomycine bij vijf en 10 micromolar resulteerden in een lichte toename van eryptose. Echter, dergelijke concentraties ook versterkt hemolyse. Incubatie van erytrocyten in ionomycine en Ringer oplossing voor zo weinig als 30 minuten was genoeg om eryptose te induceren.Verhoogde incubatietijd verhoogde het niveau van eryptose voor maximaal twee uur. Verdere incubatietijd resulteerde echter in een lichte daling van het eryptoseniveau. De hogere waarde van hemolyse na 180 minuten verklaart de vermindering van eryptose na dezelfde hoeveelheid incubatie als minder levensvatbare cellen bestonden na 180 minuten behandeling met ionomycine.Erytrocyten met ionomycinebehandeling vertoonden een helder fluorescentiesignaal voor de binding van annexin-V aan fosphatidyl serine in de bijsluiter. Cellen zonder behandeling vertoonden daarentegen een zeer zwak fluorescentiesignaal dat wijst op een zeer lage eryptose. Pre-incubatie in een glucosevrije buffer is een belangrijke trigger voor inductie van eryptose.Aangezien eryptose wordt waargenomen bij een breed scala aan ziekten, zijn de experimenten die dit protocol volgen veldspecifiek. In ons lab zijn we geïnteresseerd in het onderzoeken van de effecten van eryptose op membraanbiofysische eigenschappen en membraaninteracties met nanomaterialen. Eryptose wordt geassocieerd met een groot aantal ziekten, waaronder diabetes, sikkelcelarmoede en thalassemie.Het hebben van een reproduceerbaar protocol voor het opwekken van eryptose kan onderzoekers op een van deze gebieden helpen om verdere wetenschappelijke vragen te onderzoeken die specifiek zijn voor elk gebied.
