末梢神経系では、感覚神経(知覚神経)は受容体から中枢神経系(CNS)への感覚シグナル伝達を行い、運動神経(神経)はCNSから筋肉への信号を行う。これまでの研究では、SCは、末梢神経再生,8,9を支持する明確な運動および感覚表現型および分泌神経栄養因子を発現することが示されている。最近の研究によると、線維芽細胞はまた、異なる運動および感覚表現型を発現し、SCs10の移動に影響を与える。このように、運動神経線維芽細胞と感覚神経線維芽細胞/SCの違いを探ると、末梢神経特異的再生の複雑な分子機構を研究することができます。
現在のところ、SCおよび線維芽細胞を精製する方法は多数あり、抗ミトキ薬の適用、抗体媒介性細胞分解11、12、,12シーケンシャルイムノパンニング13およびラミニン皮層14を含む14。しかし、上記の方法はすべて線維芽細胞を除去し、SCのみを保存する。高度に精製されたSCおよび線維芽細胞は、フローサイトメトリー選別技術15によって得ることができるが、それは時間とコストのかかる技術である。そこで本研究では、多数の線維芽細胞やSCをより迅速に得るために、感覚および運動神経線維芽細胞およびSCを精製・単離するための単純な微分消化および微分付着法が開発された。
