골절 수리는 배아 골격 발달 및 리모델링과 구별되는 동적 다세포 과정입니다. 이 과정에서 손상된 조직으로부터의 부상 신호 유도는 골격 줄기/전구 세포의 신속한 모집, 증식 및 후속 분화로 이어지며, 이는 골절의 전반적인 안정성 과 고정에 모두 중요합니다1. 특히, 골절 치유의 초기 단계는 주로 periosteal 상주 세포2에 기인하는 부드러운 callus 형성을 필요로한다2. 뼈가 다치면, periosteal 세포의 부분 집합은 급속하게 반응하고 periosteum 내의 명백한 골격 줄기/선조 세포 집단의 존재를 연루하는 callus3 내의 새로 분화한 연골 중간체 및 골에 기여합니다. 따라서, periosteum-거주자 골격 줄기 세포(SSC)의 식별 및 기능적 특성화는 퇴행성 뼈 질환 및 뼈 결함에 대한 유망한 치료 접근법을 구성한다4.

SSC의 기능적 특성 및 철새 역학은 종종 다른 세포 집단에서 이주를 유도하는 것으로 알려진 사이토카인 또는 성장 인자와 함께 트랜스웰 또는 스크래치 분석을 수행함으로써 시험관 내에서 평가됩니다. 따라서, 생체 내 시스템에서 해당 되는 생체 내 시스템에서 적용을 위한 이러한 체외 실험의 결과 해석은 도전적이다. 현재, 골격 줄기/전구 세포 이동의 생체 내 평가는 전형적으로 실시간으로 관찰되지 않는다; 오히려, 그것은 골절 후 고정 된 시점에서 측정5,7,14,15,16.

real-time imaging of ccl5-induced migration of periosteal skeletal stem cells in mice

Periosteal 골격 줄기 세포 (P-SSC)는 평생 뼈 유지 보수 및 수리에 필수적이며 골절 치유를 향상시키기 위한 치료법 개발에 이상적인 초점입니다.  Periosteal 세포는 골절 치유를 위한 새로운 연골 세포 및 골세포를 공급하기 위하여 상해로 급속하게 이동합니다. 전통적으로, 세포 이동을 유도 하는 사이토카인의 효능은 트랜스웰 또는 스크래치 분석 능력을 수행 하 여 체외에서 만 실시 되었습니다. 다광원 여기를 이용한 인트라바이탈 현미경법의 발전과 함께, 최근 1) P-SSC가 CCL5로 알려진 CCR5 리간드를 이용한 철새 유전자 CCR5 및 2) 치료를 발현하는 것으로 밝혀졌으며, CCL5에 대응하여 골절 치유및 P-SSC의 이동을 향상시킨다. 이러한 결과는 실시간으로 캡처되었습니다. 여기에 설명된 프로토콜은 CCL5로 치료 후 부상으로 칼변이 봉합사 골격 줄기 세포(SSC) 틈새 에서 P-SSC 마이그레이션을 시각화하는 프로토콜이다. 이 프로토콜은 마우스 구속 및 이미징 마운트의 구성, 마우스 칼바리아의 수술 준비, 칼바리아 결함 유도 및 시간 경과 이미징의 수집에 대해 자세히 설명합니다.
