O reparo da fratura é um processo dinâmico e multicelular distinto do desenvolvimento e remodelação esquelético embrionário. Durante esse processo, a indução de sinais de lesão do tecido danificado é seguida pelo rápido recrutamento, proliferação e subsequente diferenciação das células esqueléticas de tronco/progenitor, que são todas fundamentais para a estabilidade geral e fixação de fraturas1. Em particular, os estágios iniciais da cicatrização da fratura requerem a formação de calo macio, que é atribuída principalmente às células residentes periosteis2. Quando os ossos são feridos, um subconjunto de células periosteais responde rapidamente e contribui para intermediários de cartilagem e osteoblastos recém-diferenciados dentro do calo3, implicando a presença de uma distinta população de células-tronco/progenitoras esqueléticas dentro do peristeum. Portanto, a identificação e caracterização funcional das células-tronco esqueléticas residentes no periosteum (SSCs) constituem uma abordagem terapêutica promissora para doenças ósseas degenerativas e defeitos ósseos4.
Acredita-se que os SSCs residam em vários locais de tecido, incluindo a medula óssea. Semelhante à medula óssea, os SSCs osteogênicos/condrogênicos também foram identificados no periosteum4,5,6.  Estes SSCs periosteais (P-SSCs) podem ser rotulados com marcadores de linhagem mesenquimal precoces (ou seja, Prx1-Cre5, Ctsk-Cre7 e Axin2-CreER8) durante o desenvolvimento do osso fetal4,7,8,9,10. Uma limitação significativa desses modelos de rastreamento de linhagem genética única é que há heterogeneidade substancial dentro de populações celulares rotuladas. Além disso, eles não podem distinguir SSCs rotulados de seus descendentes in vivo. Para lidar com essa limitação, desenvolvemos recentemente um mouse repórter duplo (Mx1-Cre+Rosa26-Tomate+α SMA-GFP+) para visualizar distintamente P-SSCs de SSCs de medula óssea (BM-SSCs)11. Com este modelo, foi determinado que os P-SSCs são marcados pela rotulagem dupla Mx1+αSMA+, enquanto as células Mx1+ mais diferenciadas residem nas superfícies de medula óssea, endosteal e periosteal, e ossos cortical e trabecular11,12.
Várias citocinas e fatores de crescimento são conhecidos por regular a remodelação e reparação óssea e foram testados para o aprimoramento da reparação esquelética em defeitos críticos do segmento1,13. No entanto, devido à complexidade celular dos modelos de lesões gerados pela ruptura da barreira física do compartimento ósseo, os efeitos diretos dessas moléculas na migração e ativação endógena P-SSC durante a cicatrização não são claros. As características funcionais e a dinâmica migratória dos SSCs são frequentemente avaliadas in vitro pela realização de um ensaio transwell ou scratch, em combinação com citocinas ou fatores de crescimento conhecidos por induzir migração em outras populações celulares. Assim, a interpretação dos resultados desses experimentos in vitro para aplicação em seus sistemas in vivo correspondentes é desafiadora. Atualmente, a avaliação in vivo da migração de células-tronco/progenitor esqueléticos não é normalmente observada em tempo real; em vez disso, é medido em pontos de tempo fixos após a fratura5,7,14,15,16.
Uma limitação desse método é que a migração não é avaliada em um nível unicelular; em vez disso, é medido através de mudanças nas populações celulares. Devido aos recentes avanços na microscopia intravital animal vivo e à geração de ratos repórteres adicionais, o rastreamento in vivo de células individuais agora é possível. Utilizando microscopia intravital animal viva, observamos a migração distinta de P-SSCs do nicho de sutura da calvaria para uma lesão óssea dentro de 24-48 h pós-lesão em camundongos Mx1/Tomate/αSMA-GFP.
O CCL5/CCR5 foi recentemente identificado como um mecanismo regulatório que influenciou os P-SSCs de recrutamento e ativação durante a resposta precoce de lesões. Curiosamente, não houve detecção de migração P-SSC substancial em resposta a uma lesão em tempo real. No entanto, o tratamento de uma lesão com CCL5 produz uma migração robusta e direcionalmente distinta de P-SSCs, que pode ser capturada em tempo real. Portanto, o objetivo deste protocolo é fornecer uma metodologia detalhada para o registro da migração in vivo de P-SSCs em tempo real após o tratamento com CCL5.

As células-tronco esqueléticas periosteais (P-SSCs) são essenciais para a manutenção e reparação óssea ao longo da vida, tornando-as um foco ideal para o desenvolvimento de terapias para melhorar a cicatrização da fratura.  As células periosteais migram rapidamente para uma lesão para fornecer novos condrócitos e osteoblastos para cicatrização de fraturas. Tradicionalmente, a eficácia de uma citocina para induzir a migração celular só tem sido conduzida in vitro realizando um ensaio transwell ou scratch. Com os avanços na microscopia intravital utilizando excitação multifotífica, descobriu-se recentemente que 1) P-SSCs expressam o gene migratório CCR5 e 2) tratamento com o ligante CCR5 conhecido como CCL5 melhora a cicatrização da fratura e a migração de P-SSCs em resposta ao CCL5. Estes resultados foram capturados em tempo real. Descrito aqui é um protocolo para visualizar a migração P-SSC do nicho de células-tronco esqueléticas de sutura calvarial (SSC) para uma lesão após o tratamento com CCL5. O protocolo detalha a construção de um suporte de contenção e imagem do rato, preparação cirúrgica do rato calvaria, indução de um defeito de calvaria e aquisição de imagens de lapso de tempo.
