この手順により、研究者は大型動物および臨床試験における研究に最適なナノ粒子を評価し、選択することができます。この簡単な方法は、ナノ粒子が生体内のヒト癌細胞を標的とすることができるかどうか、またそうであれば、標的化がどれほど効率的であるかを判断するのに役立ちます。手順のデモンストレーションは、ポスドクのXiodan Qin博士と、私たちの研究室の研究技術者であるアンドリュー・ラム氏です。夕方に魚の水で2つのチャンバー交配タンクを充填し、仕切りを使用して上部タンクを分離します。雄のゼブラフィッシュをチャンバーの片側に、メスのゼブラフィッシュを1、2匹ずつ部屋の別の側に置きます。翌朝8:00に仕切りを取り外します。人工濃縮植物を追加し、水の浅い領域を作成するために、上部チャンバーをわずかに傾けます。ゼブラフィッシュが3〜4時間繁殖し、メッシュネットを通して水を注ぐことによって底の部屋から卵を集めます。卵を1皿あたり200個以下のペトリ皿に移します。死んだ卵や未受精卵を取り除きます。新鮮な魚の水で3分の2を満たし、保育器に入れます。24時間後、インキュベーターから皿を取り出し、皿からできるだけ多くの水を取り除きます。1ミリリットルのプロナーゼ溶液あたり1ミリグラムを数滴加え、皿をそっと渦巻きます。この場合、発痛の兆候が現れると、数回、胚を上下にピペットして、その後の子を分解して、胚を放出する。胚の大部分が骨元から出たら、すぐに新鮮な魚の水を加してプロセスを終了します。魚の水で胚を3回リンスし、浮遊するコリオンを取り除き、胚をインキュベーターに戻します。100ミリリットルのオートクレーブ魚の水に3グラムの電気泳動グレードのアガロースを加えることによって、3%アガロース溶液を作ります。アガロースと水を完全に溶解するまで電子レンジで水を入れます。注入プレートを作成するには、熱いアガロース溶液をペトリ皿に注ぎ、4分の3がいっぱいになるまで、マイクロ射出成形金型をアガロースに置きます。アガロースが固まった後、慎重に金型を取り外します。得られた注入プレートに魚の水をオートクレーブして充填し、摂氏4度で保存します。注射針を作るために、ホウケイ酸ガラスの毛細血管をピペットの引き手で0.78ミリメートルの直径1ミリメートル引っ張ります。注射針をパテに入れて、エタノールタオルで拭いた大きなペトリ皿に入れます。HeLa細胞を収穫する前に、インキュベーターの注入プレートと魚の水を摂氏35.5度で温め直してください。移植の30分から1時間前に、HeLa細胞を収穫する。組織培養フードで働いて、吸引によってフラスコから培地を取り除く。滅菌PBSで細胞をすすいで、培地の痕跡をすべて除去します。フラスコに3ミリリットルの無菌トリプシンEDTAを加え、37°Cで3〜5分間インキュベートします。顕微鏡下でフラスコを観察し、酵素消化の進行を監視します。約80%の細胞が懸濁されたら、フラスコに6~8ミリリットルの増殖培地を加える。穏やかにピペットで細胞を収集し、15ミリリットルの滅菌チューブに転送します。チューブを135回Gで5分間遠心分離する。上清を吸引し、完全な成長培地の3ミリリットルでHeLa細胞を再懸濁します.洗浄工程を2回繰り返した後、完全増殖培地の1ミリリットルで細胞を再懸濁する。次に、ヘモサイトメーターを用いて顕微鏡下の細胞を数える。再度遠心した後、上清を取り除きます。1ミリリットルのマイクロ遠心分離管の中で、1ミリリットル当たり7番目の細胞に5倍の10倍の濃度で細胞を再懸濁する。魚の施設に運ぶときに片手で管を握って細胞を暖かく保つ。プラスチック製のピペットを使用して、ゼブラフィッシュ胚を注射プレートに置きます。プレート上の溝に合わせて前方を向いた前向きの胚を横に置きます。空気源とマイクロインジェクタをオンにします。培養器からHeLa細胞を取り出した後、P200を用いて細胞を上下に20~30回ピペットします。カットエンドを備えたゲルローディングチップを使用して、すぐに注射針の1つにHeLa細胞サスペンションの3マイクロリットルをロードします。針ホルダーに針を挿入し、注射板を顕微鏡の下に置きます。フットペダルを押して、膜腔の下の血管化領域で胚に細胞混合物を注入する。その後、胚に滅菌魚の水の数滴をピペット。注入プロセスを続けるには、非支配的な手を使用して射出プレートを次の胚に移動し、その後、足のペダルを同時に押しながら注入器を伸ばして引っ込めるために支配的な手を使用します。プレート上のすべての胚の注入を終了し、滅菌ペトリ皿に滅菌魚の水で胚を洗います。すぐに35.5°Cで料理をインキュベートします。3時間後、注入された胚を調べ、死んだ胚を取り除く。翌日、ナノ粒子の注射針を作り、大きなペトリ皿にパテに保管します。蛍光顕微鏡の下で働いて、慎重に尾転移で胚を拾い、滅菌魚の水で新しい注入プレートに置きます。尾転移を持つゼブラフィッシュごとに、ナノ粒子溶液または目の後ろに水を注入します。また、HeLa移植を受けていない対照群のゼブラフィッシュの目の後ろにナノ粒子溶液を注入し、注入されたすべての胚を摂氏35.5度でインキュベートする。注射直後と30〜60分間隔で再び、2〜3個の胚をペトリ皿に移し、希釈したMS222溶液を5滴加えて固定化する。胚が泳ぐのを止めたら、ほとんどの水を取り除いて、胚が横たわっているのを許します。端に薄い柔らかいブラシを取り付けたピペットを使用して、胚を前方に向けて前方に横たわり、片方の目だけが見えるように胚を整列させます。2X倍率を使用して、赤、青、およびブライトフィールドチャンネルで胚全体の画像をキャプチャします。尾部を捕獲するために、胚を6.4倍の倍率で画像化する。ナノ粒子の出血を避けるために、赤いチャネルの下に胚を集中させます。イメージングの直後に、新鮮な魚の水を胚に加え、インキュベーターに戻します。注入および対照ゼブラフィッシュ胚を蛍光顕微鏡を用いて画像化した。赤い点は転移性ヒト子宮頸癌細胞であり、青色の点はナノ粒子を表す。ある対照群では、癌細胞を注射したがナノ粒子を注射しなかったゼブラフィッシュは、蛍光HeLa細胞が赤色チャネルに見え、青色チャネルで特異的な蛍光シグナルは検出されなかった。他の対照群では、ゼブラフィッシュにはナノ粒子を注射したが、癌細胞では注射されなかった。青色蛍光ナノ粒子が循環系全体に分布した。特定の赤い花序は見えませんでした。尾転移を有するゼブラフィッシュにナノ粒子を注入すると、赤と青のチャンネルからの重ね合った画像は、HeLa細胞とナノ粒子の共局在化を示す。この方法を行った後、我々は、癌細胞を殺す能力を調整するためにナノ粒子を注入する有無にかかわらず、腫瘍の負担および癌細胞生存を分析することができる。この技術は、がんの早期発見や癌転移につながる生体内の癌細胞を特異的に認識できる最適なナノ粒子の選択を促進し得る。
