この方法論は、肥料ドライブ窒素が土壌作物システムに及ぼす影響を定量化し、窒素の使用効率に関する質問に答えるのに役立ちます。この技術の主な利点は、2つの連続した成長期にわたって季節の土壌と植物のサンプリングイベントで複数を可能にすることです。この方法論は、窒素肥料管理ガイドラインを改善するための鉱物化と動員の窒素サイクルプロセスをよりよく理解するのに役立ちます。フィールドプロットを設定するには、76センチメートルの間隔を持つ6つのトウモロコシを植え、最終的な15.2 x 4.6メートルのプロット次元を使用します。長さ寸法の両端から1.5メートルの境界領域と、サンプリング領域と収穫面積に隣接する長さ1.5メートルの境界領域を追加で設定します。2列目と3列目を季節の植物と土壌サンプリングエリア、4列目と5列目をトウモロコシの収穫量の収穫面積として指定します。窒素濃縮プラントおよび土壌サンプルのコレクションの幅寸法を中心とした2.4 x 3.8メートルのマイクロプロットエリアを確立し、エッジ効果を最小限に抑えるために長さと幅寸法に38メートルの未サンプリングの境界線を残します。次に、異なる色のフラグを持つ処理プロットとマイクロプロットコーナーを線分します。マイクロプロットにアクセスする際に靴カバーを着用し、マイクロプロットのフットトラフィックを最小限に抑え、富のないサンプリングエリアの汚染を防ぎ、マイクロプロットエリアを出るときに足のカバーを取り除きます。窒素15濃縮肥料を適用するには、従来の尿素中の窒素-15富化尿素10原子%を5原子%の窒素濃縮尿素に希釈し、尿素を2リットルの脱イオン水に溶かし、尿素肥料の均一な濃縮を確保します。キャリブレーション済みのバックパック二酸化炭素噴霧器を使用して、窒素-15濃縮尿素溶液をマイクロプロットに均等に適用します。次に、軽い耕作、手の熊手または64センチメートルの灌漑を含む肥料を含む尿素を適用の24時間以内に組み込み、揮発損失の可能性を最小限に抑えます。各サンプリング段階で、サンプリングエリア内から地上6個の窒素-15アンリッチトウモロコシ植物複合サンプル、窒素-15濃縮マイクロプロットから6つの地上トウモロコシ植物複合サンプルを収集します。VHとR1を地上のバイオマスの上に刻み、刻んだバイオマスをラベル付けされた袋または乾燥に入れ、一定の質量まで摂氏60度で強制空気オーブンを入れます。バイオマス乾燥重量を記録し、2ミリメートルのふるいを通過できるまで乾燥植物材料の100〜200グラムを十分に混合し、粉砕します。その後、十分に地面材料を混合し、さらなる処理のためにラベル付けされたコインエンベロープにサブサンプルを保存します。土壌サンプル処理の場合、肥料塗布から8日以内に、ハンドプローブを使用して、VHおよびR1のアンエンリッチサンプリング領域から4コア1.8センチメートルの複合土壌サンプルを植物サンプリングと並行して収集し、別のハンドプローブを使用してマイクロプロットから15コア1.8センチメートルの複合土壌サンプルを収集します。各複合土壌サンプルをバケットに均質化し、サンプルをあらかじめラベル付けされた紙袋に入れます。その後、2ミリメートルのふるいを通過できるまで各サンプルを粉砕する前に、一定の質量まで強制空気オーブンで摂氏35度で土壌サンプルを乾燥させます。実験室でのサンプル処理のために乾燥60°Cのオーブンで一晩地盤計画サンプル、サンプルが一貫性のような微細な小麦粉を得るまで、ローラージャー内の乾燥した植物および土壌サンプルを4倍のgで6〜24時間粉砕する前に。その後、細かく粉砕された材料を清潔なラベル20ミリリットルのシリル化バイアルに移します。ニトリル手袋を着用した各サンプルの合計濃度と窒素-15濃度を決定するには、まず実験室のワイプとエタノールを使用して、マイクロスケール、作業面、へら、鉗子を洗浄します。ラボベンチのラボワイプに清潔な調理器具を置き、鉗子を使用してサンプルカプセルの開口部を静かにフレアアウトします。オーブンとマイクロスケール重量を量る鍋の上に2ミリメートル上に変更されたカプセルを解放し、カプセルを引き裂きます.鉗子を使用してカプセルをクリーンな作業面に戻し、スパチュラを使用して必要な細かく粉砕されたサンプル材料の質量をカプセルに慎重に追加します。鉗子を使用して、ロードされたカプセルの上の3分の1をゆっくりと圧着し、折り畳んで密封します。カプセルを折りたたんで圧縮し続け、球状になるまでスズを穿刺または引き裂かないように注意する。鉗子を使用して、包まれたカプセルを1センチメートルの高さからきれいな暗い表面に数回落とし、漏れをチェックします。ほこりが出ない場合は、サンプルの重量を量り、96ウェルプレートの1つのウェルにカプセルを入れ、井戸の配置を記録します。各サンプルカプセル化の間に、エタノールと実験室のワイプで各調理器具と表面をきれいにし、へらと鉗子の端に特別な注意を払います。この代表的な分析では、上の粉砕トウモロコシバイオマスサンプル中の肥料由来窒素濃度は、成長期の早い時期に最も大きく、連続したサンプリング期間ごとに減少した。しかし、土壌由来窒素は、一貫して地上バイオマス窒素の最大の割合であり、最適なトウモロコシの成長のための土壌窒素供給の重要性を示した。初年度の生理的成熟時には、地上バイオマス窒素の約27%が、穀物、ストーバーおよびコブ画分で観察された同様の割合で肥料由来であった。2年目の生理的成熟期には、1年目の肥料由来窒素のわずか2%が地上バイオマスで回収された。1年の肥料由来の窒素の1ヘクタール当たり約1.6キログラムで穀物に輸出。肥料の適用から8日以内に、肥料由来窒素の大部分は、予想通り土壌プロファイルのトップ15センチメートルにあった。しかし、1ヘクタール当たり約22キログラムの窒素はすでに深部に移動していましたが、肥料由来の窒素の4〜10%は不明でした。実際、1年と2年目の終わりまでに、肥料由来の窒素の50%未満が土壌トウモロコシシステム内で占め、残りは環境に失われたか、90センチメートルの土壌サンプル深度の下に浸出した。この手順を試みる場合は、結果を無効にする可能性のある交差汚染を避けるために細心の注意を払う必要があります。窒素循環力学の理解を深めるために、土壌の無機分量および有機分画をさらに分析することができる。植物部品で分析した植物サンプルは、時間の経過とともに窒素の取り込みと転位の理解を知らせるために使用することができます。
