Este protocolo describe un modelo de roedor de DAI. La lesión cerebral traumática (TBI, por sus) es una de las principales causas de muerte y discapacidad en los Estados Unidos. Los TMI contribuyen a aproximadamente el 30% de todas las muertes relacionadas con lesiones1,2. Las principales causas de TBI difieren entre grupos de edad e incluyen caídas, colisiones de alta velocidad durante los deportes, autolesiones intencionales, accidentes automovilísticos y asaltos1,2,3.
La lesión axonal difusa cerebral (DAI) es un tipo específico de TBI inducida por la aceleración rotacional, temblores o lesiones por explosión del cerebro resultantes del movimiento sin restricciones de la cabeza en el instante después de la lesión4,5,6,7,8. DAI implica un daño axonal generalizado que conduce a un deterioro neurológico duradero que se asocia con un mal resultado, costos de atención de la salud gravosos, y una tasa de mortalidad del 33-64%1,2,4,5,9,10,11. A pesar de importantes investigaciones recientes sobre la patogénesis de DAI, no ha habido un consenso sobre las mejores opciones de tratamiento11,12,13,14.
En las últimas décadas, numerosos modelos experimentales han intentado replicar con precisión diferentes aspectos de DAI11,,12,15,16. Sin embargo, estos modelos tienen limitaciones dada la presentación única de DAI en comparación con otras lesiones focales. Estos modelos anteriores no sólo causan lesiones axonales en regiones de materia blanca, sino que también resultan en lesiones cerebrales focales. Clínicamente, DAI va acompañado de micro hemorragias, que pueden constituir una causa importante de daño a la materia blanca.
Sólo se ha demostrado que dos modelos animales replican las características clínicas clave de DAI. Gennarelli y sus colegas produjeron el primer dispositivo de rotación de cabeza lateral en 1982, utilizando la aceleración rotacional de la cabeza sin impacto para inducir coma con DAI en un primate no humano modelo15. Este modelo de primate empleó una sola rotación controlada para la aceleración y desaceleración para desplazar la cabeza a través de 60o dentro de 10-20 ms. Esta técnica fue capaz de emular la conciencia deteriorada y el daño axonal generalizado que se asemejaba a los efectos de la TBI grave observada en el cerebro humano. Sin embargo, los modelos de primates son muy caros4,11,16. Basado en parte en el modelo anterior, un modelo porcino de lesión cerebral de aceleración rotacional fue diseñado en 1994 (Ross et al.) con resultados similares14.
Estos dos modelos animales, aunque produjeron diferentes presentaciones de patología típica, han añadido en gran medida a los conceptos de patogénesis DAI. La rotación rápida de la cabeza es generalmente aceptada como el mejor método para inducir DAI, y los roedores proporcionan un modelo menos costoso para los estudios de rotación rápida de la cabeza11,16. Aquí, validamos un modelo de roedor simple, reproducible y confiable de DAI que causa daños generalizados en la materia blanca sin fracturas de cráneo o contusiones. Este modelo actual permitirá una mejor comprensión de la fisiopatología de la DAI y el desarrollo de tratamientos más eficaces.

La lesión cerebral traumática (TBI) es una de las principales causas de muerte y discapacidad. La lesión axonal difusa (DAI) es el mecanismo predominante de lesión en un gran porcentaje de pacientes con TBI que requieren hospitalización. DAI implica daño axonal generalizado por sacudida, rotación o lesiones por explosión, lo que conduce a lesiones de estiramiento axonal rápido y cambios axonales secundarios que se asocian con un impacto duradero en la recuperación funcional. Históricamente, los modelos experimentales de DAI sin lesiones focales han sido difíciles de diseñar. Aquí validamos un modelo de roedor simple, reproducible y confiable de DAI que causa daños generalizados en la materia blanca sin fracturas de cráneo o contusiones.

La Tabla 1 ilustra la línea de tiempo del protocolo. La tasa de mortalidad en este modelo de DAI fue del 0%. En esta investigación, me gustaría presentar nuestro modelo sobre la inducción difusa lesión cerebral axonal basada en el resultado del mecanismo de aceleración rotacional TBI.Seleccione ratas Sprague-Dawley machos adultos que pesen de 300 a 350 gramos.
