Les données génétiques structurelles peuvent être transcrites à un produit fonctionnel par l’expression des gènes. L’analyse de l’ARN est utilisée pour découvrir des différences dans l’expression des gènes dans différentes conditions. Il existe un certain nombre de méthodes pour extraire les acides nucléiques comme suit : guanidinium thiocyanate, extraction par phénol-chloroforme, chromatographie à base de cellulose, extraction par matrices de silice, et anion-échange1,2.
Une détection adéquate de l’expression génique est influencée par l’intégrité de l’ARN isolé des tissus; par conséquent, il est essentiel d’évaluer l’intégrité de l’ARN isolé des tissus avant que d’autres tests ne soient effectués parce que des tests moléculaires complémentaires sur un ARN de mauvaise qualité peuvent compromettre les résultats des applications diagnostiques. Ainsi, l’ARN à haute intégrité est nécessaire pour les tests biologiques moléculaires avec différentes applications diagnostiques : RT-PCR quantitatif, micro-réseaux, test de protection ribonucléase, analyse de tache nordique, cartographie de l’ARN et construction de bibliothèque cDNA3,4.
L’ARN devient plutôt instable après avoir été longtemps conservé. Les longs fragments d’ARNm de plus de 10 ko sont particulièrement sensibles à la dégradation5,6. Ainsi, les chercheurs doivent tenir compte de divers facteurs qui influencent l’intégrité de l’ARN purifié. La pureté de l’ARN doit être protégée contre les RNases, les protéines, l’ADN génomique et la contamination par les inhibiteurs enzymatiques. En outre, le meilleur et acceptable rapport d’absorption de l’ARN aux UV (260/280) doit être dans la gamme de 1,8-2.0 avec une fragmentation minimale sur l’électrophorèse. Des techniques de laboratoire récemment développées ont permis aux scientifiques d’évaluer l’intégrité de l’échantillon d’analyse moléculaire plus pratiquement7,8.
Il est beaucoup plus difficile d’extraire l’ARN intact du tissu pancréatique que d’autres types de tissus en raison de la grande quantité de ribonucléases (RNases). Cependant, les méthodes d’extraction existantes, à savoir l’éjection rapide du tissu pancréatique de la cavité abdominale et l’homogénéisation à basse température pour entraver RNases, se sont avérées inefficaces7,8,9,10,11,12,13,14.
Le but de la présente étude expérimentale comparative est de modifier et de comparer les méthodes existantes pour déterminer les méthodes les plus efficaces. À cette fin, divers protocoles d’extraction de l’ARN ont été modifiés et comparés. Il visait spécifiquement à déterminer la méthode la moins coûteuse nécessitant une quantité minimale de tissu pancréatique.

Indépendamment de la méthode d’extraction, l’extraction optimisée de l’ARN des tissus et des lignées cellulaires se fait en quatre étapes : 1) homogénéisation, 2) dénaturation efficace des protéines de l’ARN, 3) l’inactivation de la ribonucléase, et 4) l’élimination de la contamination de l’ADN, des protéines et des glucides. Cependant, il est très laborieux de maintenir l’intégrité de l’ARN quand il ya des niveaux élevés de RNase dans le tissu. L’autolyse spontanée rend très difficile d’extraire l’ARN du tissu pancréatique sans l’endommager. Ainsi, une méthode pratique d’extraction d’ARN est nécessaire pour maintenir l’intégrité des tissus pancréatiques pendant le processus d’extraction. Une étude expérimentale et comparative des protocoles existants a été réalisée en obtenant 20-30 mg de tissus pancréatiques de rat en moins de 2 minutes et en extrayant l’ARN. Les résultats ont été évalués par électrophorèse. Les expériences ont été réalisées trois fois pour la généralisation des résultats. L’immersion du tissu pancréatique dans le réactif de stabilisation de l’ARN à -80 °C pendant 24 h a donné un ARN à haute intégrité, lorsque le réactif d’extraction de l’ARN a été utilisé comme réactif. Les résultats obtenus étaient comparables aux résultats obtenus à partir de kits commerciaux avec fixations de colonne de spin.

Salut tout le monde. Nous avons conçu, une méthode descriptive simple et rapide en classe pour l’extraction efficace d’ARN du tissu pancréatique. Le principal avantage de cette méthode est une procédure optimisée pour extraire l’ARN de la quantité minimale de tissu due à l’autolyse. Salut encore. Démonstration de la procédure sera faite par Sanaz Dastghaib, c’est mon étudiant au doctorat dans notre laboratoire au département de biochimie de l’Université shiraz des sciences médicales.First préparer le lieu de la chirurgie, mettre toutes les exigences sous le capot. Pour inactiver RNase stériliser tous les instruments de chirurgie dans un four. Injecter de la kétamine-xylazine intraperitoneally 80/8 milligramme par kilogramme.
