Neste artigo, descreve-se uma metodologia otimizada para a produção e purificação do mbtBDNF em um procedimento baseado em cromatografia de afinidade, a partir do trabalho de Sung e colaboradores17. As otimizações incluem o uso de um reagente de transfecção econômico (PEI) mantendo a eficiência de métodos de transfecção mais caros, como a lipofectamina. Os neurônios são as unidades funcionais do sistema nervoso que possuem uma morfologia complexa e especializada que permite a comunicação sináptica e, portanto, a geração de comportamento coordenado e complexo em resposta a diversos estímulos. Projeções neuronais como dendritos e axônios são características estruturais críticas envolvidas na comunicação neuronal, e neurotrophins são atores cruciais na determinação de sua morfologia e função1. Neurotrophins são uma família de fatores de crescimento secretos que incluem NGF, NT-3, NT-4 e fator neurotrófico derivado do cérebro (BDNF)2. No sistema nervoso central (SNC), o BDNF participa de diversos processos biológicos, incluindo neurotransmissão, arborização dendrítica, maturação de colunas dendríticas, potencialização a longo prazo, entre outros3,,4. Portanto, o BDNF desempenha um papel crítico na regulação da função neuronal.
Diversos processos celulares regulam a dinâmica e a função do BDNF. Na superfície neuronal, o BDNF liga o receptor de tropomyosina quinase B (TrkB) e/ou o receptor de neurotropfina p75 (p75). Os complexos BDNF-TrkB e BDNF-p75 são endocitados e classificados em diferentes organelas endocíticas5,,6,,7,,8. O correto tráfico intracelular do complexo BDNF/TrkB é necessário para sinalização BDNF adequada em diferentes circuitos neuronais9,,10,,11. Por essa razão, uma compreensão profunda da dinâmica do tráfico de BDNF e suas alterações encontradas nos processos fisiofisiológicos é essencial para compreender a sinalização de BDNF em saúde e doença. O desenvolvimento de novas e específicas ferramentas moleculares para monitorar esse processo ajudará a impulsionar este campo e permitirá uma melhor compreensão dos mecanismos regulatórios envolvidos.
Existem várias ferramentas disponíveis para o estudo do tráfico de BDNF em neurônios. Uma metodologia comumente utilizada envolve a transfecção de BDNF recombinante marcado com moléculas fluorescentes como proteína fluorescente verde (GFP) ou a variante monomérica fluorescente de mudança vermelha do GFP mCherry12,13. No entanto, uma grande deficiência da superexpressão do BDNF é que elimina a possibilidade de fornecer concentrações conhecidas dessa neurotropina. Além disso, pode resultar em toxicidade celular, obscurecendo a interpretação dos resultados14. Uma estratégia alternativa é a transfecção de um TrkB marcado por epitope, como Flag-TrkB. Essa metodologia permite o estudo da dinâmica de internalização TrkB15,mas também envolve transfecção, o que pode resultar em alteração da função TrkB e toxicidade celular. Para superar esses obstáculos metodológicos, foram desenvolvidas variantes recombinantes de NGF e BDNF contendo uma sequência avi (BDNFAvi), que pode ser mono-biointinalada pela enzima biotin-ligase BirA,16,,17. BDNF recombinante biotinína pode ser acoplado a diferentes ferramentas ligadas a streptavidin, que incluem fluoroforos, contas, nanopartículas paramagnéticas, entre outras para detecção. Em termos de imagem de células vivas, os pontos quânticos (QD) tornaram-se fluoroforos frequentemente utilizados, pois possuem características desejáveis para o rastreamento de partículas únicas, como aumento do brilho e resistência ao fotobleaching quando comparados com fluoroforos de molécula pequena18.
A produção de BDNF mono-biotinilado (mbtBDNF) usando BDNFAvi foi alcançada pela co-transfecção de plasmídeos que conduzem a expressão de BDNFAvi e BirA, seguido pela purificação da proteína recombinante por cromatografia de afinidade com um rendimento de 1-2 μg de BDNF por 20 mL de mídia cultural hek293-condicionada17. Aqui, propomos uma modificação deste protocolo que permite a purificação do BDNFAvi a partir de 500 mL de mídia hek293, que busca maximizar a recuperação de proteínas em um protocolo baseado em coluna cromatografia para facilitar a manipulação. O agente de transfecção usado, polietileneimina (PEI), garante um método econômico sem sacrificar o rendimento da transfecção. A etapa de mono-biotintilação foi adaptada a uma reação in vitro para evitar as complicações associadas às co-transfecções e garantir a rotulagem homogênea de BDNF. A atividade biológica do mbtBDNF foi demonstrada por experimentos de microscopia de bolhas ocidentais e fluorescência, incluindo ativação de pCREB e imagem de células vivas para estudar o transporte axonal retrógrado de BDNF em câmaras microfluidas. O uso deste protocolo permite uma produção otimizada e de alto rendimento de BDNF mono-biotinilado e biologicamente ativo.

CREB é um fator de transcrição que é alvo de ERK12 ativado em neurônios corticais. Estimular neurônios com concentrações crescentes de pontos quânticos BDNF resultou em um aumento dependente de dose de fosforilação do CREB e presença de partículas de pontos quânticos ao redor do núcleo indicando que as partículas de pontos quânticos BDNF foram endocitadas e desencadearam a ativação de vias de sinalização associadas à ativação TrkB mediada pelo BDNF.Um aumento de duas vezes no sinal de fosfo-CREB foi detectado ao estimular neurônios com 200 pontos quânticos BDNF picomolares, enquanto estimular com dois pontos quânticos BDNF nanomolar resultou em um aumento de 3,5 vezes no sinal phospho-CREB. Portanto, o oviduto BDNF biotinilado é biologicamente ativo e não perde sua atividade quando acoplado a pontos quânticos streptavidin tornando-o adequado para imunofluorescência e imagem de células vivas. Finalmente, o potencial de imagem dos pontos quânticos BDNF foi avaliado em culturas compartimentadas utilizando câmaras microfluidas.Os neurônios corticais foram semeados em câmaras microfluidas para separar os compartimentos axonal e somatodendritic e foram estimulados com dois pontos quânticos BDNF nanomolar por 3,5 horas. A microscopia de células vivas foi realizada e os kymógrafos resultantes foram usados para quantificar a velocidade do ponto quântico BDNF contendo organelas. Foi detectada uma velocidade móvel média de 0,91 micrômetros por segundo, em consonância com a análise prévia do transporte mediado por timina citoplasmática.Câmaras microfluidas tratadas com dois pontos quânticos nanomolar streptavidin não mostraram pontos quânticos em movimento nos micro grupos, como mostrado pelo kymograph.
