主要な組織適合性複合体(MHC)は、すべての脊椎動物に存在し、感染性病原体に対する細胞介在免疫を決定する遺伝子のセットである。MHCクラスIは、ウイルス感染時に産生されるウイルス成分などの内因性ペプチドを、細胞性免疫を媒介し免疫調節1に関与するCD8+T細胞の表面上のT細胞受容体(TCR)に提示する。ペプチドに結合するMHC Iの構造研究は、CD8+ T細胞免疫応答の評価およびワクチン開発において重要な役割を果たすMHC I分子によるペプチド結合モチーフおよび提示特徴に関する情報を提供する。
ビョルクマンら2によるMHC I分子の最初の結晶化と構造決定以来、MHCI分子の結晶構造解析は、ペプチドがMHC I分子に結合する方法の理解を大きく促進し、重鎖およびペプチドとの軽鎖の相互作用を理解するのに役立つ。一連のフォローアップ研究は、軽鎖をコードする遺伝子がMHCと関連していないが、軽鎖はMHC I分子3,4の集合体にとって重要なタンパク質であることを示した。これは、複数の表面上のMHCクラスI分子の3つのドメインと相互作用する。軽鎖が存在しない場合、MHCクラスI分子は抗原提示細胞の表面上で正しく発現できず、TCRと相互作用して免疫学的機能を発揮することができません。
MHCIは重鎖(H鎖)と軽鎖(すなわち、β 2-ミクログロブリン(β 2m))から構成され、適当なペプチド5に結合して組み立てられる。H鎖の細胞外セグメントは、α1、α2およびα3ドメイン6から成る。α1およびα2ドメインは、ペプチド結合溝(PBG)を形成する。β 2m鎖は、MHC Iにおけるアセンブリ複合体の構造サブユニットとして機能し、複合体の立体構造を安定化させ、かつMHC IH鎖折りたたみ7、8、9に対する分子シャペロンである。一連の研究は、コウモリ(カイロプテラ)(Ptal-N*01:01)10、アカゲザル(霊長類)(マムB*17)11(Mamu-A*01)12(Mamu-A*02)13、マウス(Rodentia)(H-14)などの様々な哺乳類からのMHC I H鎖が示されています(H-14) 15,犬 (カルニボラ) (DLA-88*50801)16, 牛 (アルチオダクチラ) (BoLA-A11)17と馬 (ペリソダクチラ) (Eqca-N*00602 および Eqca-N*00601)18はヘテロβ 2m ) と組み合わせることができる。 これらのハイブリッド分子は、構造および機能研究でしばしば使用されます。.しかしながら、2mの異種βを有するハイブリッドMHCIの機能・構造研究のための方法論は、まだ要約されていない。一方、異なるタキサ間の2mの交換βの構造的根拠は不明のままである。

Stability and Structure of Bat Major Histocompatibility Complex Class I with Heterologous ÃƒÅ½Ã‚Â²2-Microglobulin

本研究では、方法論を要約し、異種β-2ミクログロブリンと複合体化したハイブリッドMHCクラスの実現可能性を評価するモデルとして、Bat MHCクラスIを用いた。これまでの研究では、哺乳動物のB2M置換がペプチドの提示に有意に影響を与えないことを示している。実際、ハイブリッドMHCクラスIは、元の分子と同様の構造と機能を活性化することができる。これらの技術は、感染症および腫瘍免疫療法中の組織応答評価のためのMHC Iの機能的および構造化された研究を容易にするために使用することができる。大腸菌培養変換のために、バットまたはヒトMHCクラスIを含むプラスミドの10ナノグラムを2マイクログロブリン水素鎖を100マイクログロブリン水素鎖を100マイクログロブリン水素鎖に加え、30分間氷で懸濁液を浴びる。インキュベーションの終わりに、42°Cで90秒間細菌に熱ショックを与え、培養物を氷浴に2分間戻す。次に、培養液に800マイクロリットルのリソジニースープを加え、ロッキングプラットフォーム上で37°Cで20分間、毎分200回転でサスペンションを振ります。インキュベーションの終わりに、適切な抗生物質耐性培養ブレードに100マイクロリットルの細菌を広げる。翌朝、最近変換された単一の細菌クローンを選び、抗生物質培地で3ミリリットルのLBでクローンを接種する。12〜16時間摂氏37度のロッキングプラットフォームに文化を置きます。翌朝、活性化細菌ストックの500マイクロリットルを抗生物質培地で50ミリリットルのLBに移し、一晩ロッキングインキュベーターに細菌を戻す。残りの活性細菌ストックを500マイクロリットルのアリコートに分割し、各アリコートを40%グリセロールの500マイクロリットル量に加え、マイナス80°Cの貯蔵を行います。大量の組換えタンパク質を生成するために、翌朝、抗生物質培地を用いて2リットルのLBに細菌懸濁液を1〜100比で移し、600ナノメートルで0.6の吸光度が達成されるまで、ロッキングインキュベーターに培養液を戻す。次に、培養物に1ミリモルIPTGを加え、トランス遺伝子の発現を誘導する。4〜6時間の誘導後、細菌培養物を瓶に移して遠心分離し、細菌細胞ペレットをボトルあたり60ミリリットルのPBSで再懸濁する。次に超音波細胞破壊器で発現した組換えタンパク質を99回、超音波処理当たり300ワットで12秒間隔で解放する。封入体の精製のために遠心分離によって超音波処理された細菌を収集し、2つの追加の遠心分離のための洗浄バッファーの適切な量でペレットを再中断します。2回目の洗浄後、封入体と再懸濁液バッファーを再中断し、20マイクロリットルのサンプルをSDSページ用に取っておき、封入体純度を試験する。残りのサンプルを遠心分離し、ペレットを含む封入体を秤量する。ペレットに溶解バッファーを追加し、バッファーのミリリットル当たり 30 ミリグラムの包み込み体の最終濃度にします。.そして、封入体が溶解緩衝液に溶解するまで、溶液をゆっくりと摂氏4度で攪拌するために磁気スターラーを使用してください。沈殿物を廃棄した後、封入体をマイナス20度またはマイナス80°Cで保管します。MHC複合リフォールディングの場合は、グルタチオンを5ミリモル減らし、250~300ミリリットル(摂氏4度リフォールディングバッファー)に0.5ミリモル酸化グルタチオンを加えます。そして、ゆっくりと10〜20分間摂氏4度で磁気攪拌機で溶液をかき混ぜます。MHC水素鎖およびベータ2ミクログロブリン希釈液については、1ミリリットルのシリンジに封入体をロードし、1ミリリットルの包込み体溶液全体を攪拌棒の近くのリフォールディングバッファーの1リットルに注入し、迅速かつ効率的な希釈を得る。次に、ジメチルスルホキシド中のペプチド1ミリリットル当たり5ミリグラムを溶解し、ちょうど実証したように、200マイクロリットルのペプチドを再折り返し溶液に素早く注入する。10~20分のゆっくり撹拌の後、H鎖包包体の3ミリリットルを新しい1リットルのリフォールディングバッファに注入し、再折を摂氏4度で8~10時間進めます。再折したタンパク質濃度を決定するために、10キロダルトン多銅オキシダーゼ膜を有する加圧チャンバに交換バッファーを加え、バッファーを30〜50ミリリットルの体積に濃縮する。リフォールディング溶液を遠心分離管に移し、遠心分離により沈殿物を除去する。慎重にチャンバーにスーパーネートを移し、さらに約1ミリリットルの最終体積にバッファーを濃縮します。遠心分離によって最終的な汚染物質を除去し、末面を滅菌チューブに移します。タンパク質を精製し、ピーク時にサンプルを収集し、SDSページを使用して分析するために、10 300GLサイズの除外カラムを使用します。MHC複合体のピークを収集し、1ミリリットルあたり15ミリグラムの最終的な濃度にタンパク質を濃縮.その後、1ミリリットル当たり7.5ミリグラムに複合体を希釈します。シッティングドロップ蒸気拡散技術を用いて複合MHCとペプチドの結晶化を行うために、0.8マイクロリットルのサンプルを市販のクリスタルボードに加え、基板をシールします。100マイクロリットルのリザーバー溶液に対してサンプル溶液を4~18°Cで平衡化し、顕微鏡を使用して今後6ヶ月間の結晶成長を定期的に観察します。極低温保護のために、実験の終わりに、100度ケルビン気体窒素流でサンプルを急速に冷却する前に、20%グリセロールを含む貯蔵溶液に結晶を移す。次に、標準プロトコルに従ってX線回折データを収集します。結晶複合体の構造を決定するには、HK L 2000ソフトウェアパッケージ内のDenzoプログラムで高解像度の構造X線回折データを開き、タンパク質データバンク内の初期モデルを選択します。タンパク質の構造を決定するには、CCP4のフェイザーMRプログラムでデータを開き、初期関節絞り込みには精製されたX線モデルを使用します。次に、フェニックスの洗練とジョイントモードを使用してX線単独の改良を行い、関節中性子をX線の改良に使用します。各絞り込みラウンドの後、Fo-Fcに対してモデルを手動でチェックし、クートの2Fo-Fc正の核密度マップを確認します。これらの代表的な実験では、ヘンドラウイルス由来ヘンドラウイルス1ペプチドの結合能力を、相同のコウモリβ2ミクログロブリンおよび異種ヒトβ2ミクログロブリンを用いたMHCクラスI水素鎖を打つペプチドを評価した。どちらの解析でも、β2マイクログロブリン軽鎖で、レナールを介して高分解能の結晶が形成された。ベータ2ミクログロブリンの存在下では、これらの複合体は形成しない。MHCクラスIの水素鎖ヘンドラウイルス1では、 ヒトβ2ミクログロブリン構造、保存残基H31、D53、W60、およびヒトβ2ミクログロブリンのY63は、2つのマイクログロブリンのコウモリベータに相当し、ペプチド結合溝の底部に接触し、保存されたQ8、Y10、R12、および24残基を3つのβに対応する。全体のコウモリとヒトの構造において、水素鎖の1〜184個の残基の平均二乗平均平方導出量は、炭素α原子の重ね合いの全てにおいて0.248であり、これら2つの複合体間に差がないことを示している。ペプチドアライメントの構造は、これら2つの複合体におけるヘンドラウイルス1個のペプチドの確認が非常に類似していることを示している。配列アライメントはまた、異なる種由来のβ2ミクログロブリンのアミノ酸が高度に保存されていることを示している。高い報告効率で敵対者の複合体を提供することが重要です。したがって、適切なβ2マイクログロブリンを選択し、高度に精製された封入体を使用することが重要です。このプロトコルは、他の種における2つのミクログロブリン置換の可能性を通じて安定したMTC I複合体を得るために使用することができ、そしてそれらが存在することはMTC Iの構造および機能である。これらの研究から得られたデータは、感染症および腫瘍免疫療法中のT細胞応答を評価するために使用することができる。
