カルス産生用の外植植物としての若い花序は、遺伝子機能を明らかにするとともに作物の改善のために、迅速かつ効率的な遺伝子変換と再生方法を確立するために重要である。カルス産生用の外植植物としての若い花序は、遺伝子機能を明らかにする迅速かつ効率的な遺伝子変換および再生方法を確立するために重要である。この技術のビデオデモンストレーションは、時間と労力を無駄にすることなく、希望するトランスジェニックアライメントを生成するために材料を維持するプロセスを理解するのに役立ちます。まず、水田や煙草の段階で温室から若い米の花序を集め、葉の鞘で覆われていることを確認することから始めます。70%アルコール綿棒で各花序を拭き、切断する前に乾燥させます。無菌ラボベンチに花序を持ってきて、滅菌したはさみで小さく切り、挿し木をNBD2培地を含むペトリプレートに移します。カルスを誘発するために10〜14日間摂氏26度で暗闇の中でプレートをインキュベートします。変態を行うために、選択的抗生物質を用いた白色EBプレートから、50ミリリットルの円錐型滅菌試験管に同じ抗生物質を含む5ミリリットルの液体YEB培地に単一コロニーを移管する。細菌が0.5のOD 600に成長するまで、250倍gと摂氏25〜28度で軌道シェーカーでチューブを振ります。250ミリリットルの円錐フラスコに同じ選択的抗生物質を含むYEB培地の100ミリリットルに細菌懸濁液1ミリリットルを加え、250倍gと摂氏28度で軌道シェーカーでフラスコを4時間振ります。培養物を4,000gで室温で10分間遠心し、菌を採取する。上清を捨てて、AAM AS培地でペレットを再懸濁し、その後、0.4のOD 600に希釈します。インキュベーション後、約150の健康な淡黄色の胚発生カリを150ミリリットルの無菌フラスコに集める。フラスコに50〜75ミリリットルの細菌細胞懸濁液を加え、新鮮なAAM AS培地を10〜25ミリリットル加えて10〜20分間カリを浸し、時折揺れます。フラスコから細菌懸濁液を慎重に注ぎ、滅菌濾紙でカリを乾燥させ、NBD AS培地でペトリ皿に置き、ろ紙で覆います。暗闇の中で25~28度のカリを3日間インキュベートし、細菌の過剰増殖をチェックします。3日間の共同栽培の後、フィルターペーパーを使用して無菌ペトリ皿にカリを移し、きれいなベンチで2時間空気乾燥します。カリがフィルタ用紙に付着していないことを確認し、それらを一次選択媒体NBD2に転送します。2週間後、新しい選択媒体を含む新しいプレートにカリを均等に移し、その後、分化のための新鮮なMS培地にカリを移動し、より多くの芽を増殖させるために1リットルハイグロマイシンあたり10ミリグラムのMS培地に芽を撃つ。新しい芽を半分のMS培地に移して根管誘導し、25〜28度の光の下で培養します。形質転換された植物は2週間以内に根を作り出すはずです。生殖表現型観察を行うために、フローリから古い点とレンマを取り出し、ステレオ顕微鏡でアンサー全体を画像化します。花粉の生存率を観察するには、花の無菌の前に成熟した米の小穂を選び、古い花とレンマを取り除いてアンサーを放出します。6つのアンサーを取り、ガラスのスライドの上に置きます。蒸留水を1滴加え、ピンセットでアンサーを粉砕して花粉粒を放出し、2~3滴のヨウ素溶液を加え、カバースリップで覆います。低倍率の顕微鏡でスライドを観察します。黒く染めた花粉粒はより活発な生存率を示し、色のない穀物や黄色い茶色に染められた穀物はスタントまたは退化する。横断的切除顕微鏡を行うには、0.5%トルイジンブルー溶液を使用してスライドを30分間染色し、水ですすいでフュームフードで乾燥させます。乾いたら、スライドをニュートラルツリーの接着剤で密封し、スライドにカバースリップをそっと置き、ヒュームフードで乾燥させ、顕微鏡でサンプルをイメージします。このプロトコルは、米中のアグロバクテリウム媒介性遺伝子変換によって男性の無菌線を作成するために使用された。胚原性のカリを、増殖のためにトランスフォーメーションまたはサブ培養のために直接使用し、さらなる感染のためにより多くのカリを得た。48時間の共同栽培後、2回目の選択培地にリを移した。10~14日後、形質転換された呼び出しは新たに形成されたマイクロカリを示し、未変換の呼び出しは茶色になって死んだ。その後、健康でクリーミーな色のカリを再生培地に移した。変身したカリは徐々に緑色の斑点を示した。これらの緑色のスポットは、芽の成長を可能にするために再生培地で培養し、その後、根を誘導するために半分のMS培地にシフトした。その後、よく発達した根を持つ健康で活発な成長芽が収穫されました。合計で21種の再生苗が得られた。ハイグロマイシン領域のPCR増幅は、トランスフォーム剤がsgRNA標的部位で変異を同定するために遺伝子型化された約85.71%の変換頻度であることを示した。18のトランスジェニック植物のうち、8つのヘテロ接合線と3つのホモ接合線がCRISPR CAS9陽性であった。ホモ接合ノックアウト突然変異体は、基本的な男性生殖器官観察によって検証された。osabcg15のアンサーは野生型のものよりも小さく、青ざめ、成熟した花粉粒を欠いていた。横断断面顕微鏡は、osabcg15におけるアンサー形態学的欠陥を調べるため行った。ステージ10では、野生型のマイクロスポアが丸い形になり、濃い青色の染色されたエキシンで空いた。対照的に、osabcg15マイクロスポは崩壊し、分解した。このプロトコルを試みるとき、収集された材料がmeiosis段階にあることを確認し、カリの空気乾燥時間を制御することが重要です。
