Dit manuscript beschrijft een stap-voor-stap protocol om te controleren op de aanwezigheid van heterologe interacties tussen Golgi-residente type III membraaneiwitten in tijdelijk getransfecteerde menselijke cellen met behulp van de meest recente variant van de split luciferase complementatie assay. De procedure is het meest uitgebreid getest tegen nucleotide suikertransporters (NST's), maar we konden ook positieve resultaten behalen voor andere Golgi-residente type III-membraaneiwitten waarvan de N- en / of C-termini tegenover het cytoplasma staan.
Onze onderzoeksgroep onderzoekt de rol van NST's bij glycosylering van macromoleculen. NST's zijn Golgi- en/of ER-residente type III membraaneiwitten met N- en C-termini naar de cytoplasmatische kant van het organellaire membraan1. Van NST's wordt gedacht dat ze nucleotide-geactiveerde suikers over organelmembranen dragen om glycosyltransferasen van hun substraten te voorzien. NST's vormen dimeren en/of hogere oligomeren2,3,4,5,6,7,8,9,10. Bovendien zijn er ook heterologe interacties tussen verschillende NST's gemeld6,11. NST's bleken ook complexen te vormen met functioneel gerelateerde glycosylatie-enzymen12,13,14. We zochten naar een alternatief voor de momenteel gebruikte techniek, fluorescence lifetime imaging (FLIM)-gebaseerde FRET-benadering, voor het bestuderen van interacties van NST's en functioneel gerelateerde Golgi-residente eiwitten, dus besloten we om de split luciferase complementatie assay te testen. Het stelde ons in staat om een nieuwe interactie te identificeren tussen een NST en een functioneel gerelateerd glycosylatie-enzym9.
De meest recente wijziging van de split luciferase complementatie assay, NanoBiT, wordt gebruikt in het protocol dat hier wordt gepresenteerd15. Het is afhankelijk van de reconstitutie van het luciferase-enzym (bijv. NanoLuc) uit de twee fragmenten - de grote, aangeduid als grote BiT of LgBiT, een 17,6 kDa-eiwit, en de kleine, samengesteld uit slechts 11 aminozuren, aangeduid als kleine BiT of SmBiT. De twee eiwitten van belang zijn versmolten met de complementaire fragmenten en tijdelijk tot expressie gebracht in een menselijke cellijn. Als de twee fusie-eiwitten op elkaar inwerken, wordt in situ een luminescentie geproduceerd na toevoeging van een celdoorlatend substraat. Deze twee fragmenten zijn geoptimaliseerd zodat ze zich associëren met minimale affiniteit, tenzij ze worden samengebracht door een interactie tussen de eiwitten van belang waarmee ze zijn gefuseerd.
Over het algemeen hebben op bioluminescentie gebaseerde methoden enkele voordelen ten opzichte van methoden die gebaseerd zijn op fluorescentie. Bioluminescente signalen hebben een hogere signaal-ruisverhouding omdat de achtergrondlumescentie verwaarloosbaar is in vergelijking met het luciferase-afgeleide signaal16. Fluorescentie-gebaseerde benaderingen daarentegen lijden meestal aan een relatief hoge achtergrond veroorzaakt door het fenomeen van autofluorescentie. Bovendien is bioluminescentie minder schadelijk voor de geanalyseerde cellen dan fluorescentie, omdat het in het eerste geval niet nodig is om het monster te prikkelen. Om die redenen concurreren bioluminescente benaderingen voor het bestuderen van PPI's in vivo met de veelgebruikte fluorescerende methoden zoals Förster Resonance Energy Transfer (FRET) of Bimolecular Fluorescence Complementation (BiFC).
Ons protocol is gebaseerd op verwijzende luminescentie verkregen voor de eiwitcombinatie die van belang is voor luminescentie verkregen voor de controlecombinatie. De laatste omvat een van de geteste eiwitten die is gefuseerd met een groter fragment en een controle-eiwit (bijv. HaloTag), gefuseerd met een kleiner fragment. De laatste is een eiwit van bacteriële oorsprong waarvan niet wordt verwacht dat het interageert met een van de eiwitten van zoogdieren. Het gebruik van dit eiwit als controle stelt beperkingen aan de topologie van de Golgi-residente paren van eiwitten die moeten worden geanalyseerd. Omdat in zoogdiercellen dit eiwit wordt gesynthetiseerd in het cytoplasma, moeten beide eiwitten van belang ten minste één cytoplasmatische staart hebben.
Deze aanpak kan met name nuttig zijn voor de eerste screening van PPI's. Het kan de voorkeursmethode worden wanneer de fusie-eiwitten van belang worden uitgedrukt op niveaus die eenvoudigweg onvoldoende zijn om andere benaderingen toe te passen. Evenzo kan de split luciferase-complementatietest de beste optie zijn als de eiwitten van belang op hoge niveaus tot expressie komen, maar dit heeft een negatieve invloed op hun subcellulaire lokalisatie of het is bekend dat het niet-specifieke interacties forceert. Omdat het kleinere fragment slechts 11 aminozuren bevat, kan de split luciferase complementatietest worden toegepast wanneer het gebruik van grotere tags onmogelijk is. Ten slotte kan het worden gebruikt om gegevens die zijn verkregen met behulp van andere technieken verder te bevestigen, zoals in het hier gepresenteerde geval.

Het doel van dit protocol is om de toepasbaarheid te onderzoeken van de meest recente variant van split luciferase complementatie voor het aantonen van heterologe complexen gevormd door nucleotide suikertransporters (NST's). Deze ER- en Golgi-residente multitransmembraaneiwitten dragen de cytoplasmatisch gesynthetiseerde nucleotidesuikers over organelmembranen om enzymen te leveren die glycosylering bemiddelen met hun substraten. NST's bestaan als dimeren en/of hogere oligomeren. Heterologe interacties tussen verschillende NST's zijn ook gemeld. Om te verifiëren of de techniek geschikt is voor het bestuderen van het fenomeen van NST-heteromerisatie, hebben we het getest tegen een combinatie van de twee Golgi-residente NST's waarvan eerder is aangetoond dat ze op verschillende andere manieren associëren. De luciferase-complementatietest lijkt bijzonder geschikt te zijn voor het bestuderen van interacties tussen Golgi-residente membraaneiwitten, omdat het geen hoge expressieniveaus vereist, die vaak eiwitmislokalisatie veroorzaken en het risico op valse positieven verhogen.

Om de meest betrouwbare gegevens in deze benadering te verkrijgen, moeten alle mogelijke combinaties worden getest (zie figuur 1). Tegelijkertijd moeten positieve en negatieve controles worden opgenomen. De positieve controle moet bestaan uit de twee eiwitten waarvan bekend is dat ze interageren, waarvan de ene is gefuseerd met het grotere fragment en de andere is gefuseerd met het kleinere fragment. Demonstration of Heterologous Complexes formed by Golgi-Resident Type III Membrane Proteins using Split Luciferase Complementation Assay

Our protocol allows to identify and analyze interactions between multi transmembrane proteins of the endoplasmic reticulum and golgi complex in living cells at low expression levels. The main advantage of this technique is it's to special sensitivity, which allows to avoid high over expression levels that are often detrimental to cellular health and may disturb protein subcellular localization. Harvest the adherent HEK293 T-cell culture by trypsinization and re-suspend the cells in a dedicated complete growth medium.Plate the cells onto a clear bottom, white side 96-well plate. Adjust the total number of wells to accommodate all tested combinations and controls, including replicates. Then, culture the cells overnight in standard conditions.On the next day, transfect the cells with the desired combinations of expression plasmids. Dilute the plasmids in a serum-free medium to 6.25 nanograms per microliter for each construct and add the lipid-based transfection reagent. Add an appropriate lipid to DNA ratio.Incubate the plasmids according to manufacturer's instruction. Then, add eight microliters of the lipid DNA mixture to the designated wells. Mix the content of the plate with gentle rotation and culture the cells for 20 24 hours in standard conditions.On the next day, replace the conditioned medium in each well with 100 microliters of a serum-free medium, making sure that the cells do not detach upon medium exchange. Just before measuring luminescence, mix one volume of furimazine with 19 volumes of a dilution buffer. Add 25 microliters of the furimazine working solution into each well, and gently mix the plate by hand or with a orbital shaker.Insert the plate into a luminescence micro plate reader and equilibrate the plate at 37 degrees Celsius for 10 15 minutes. Select the wells to be analyzed and read luminescence with an integration time of 0.3 seconds. Continue to monitor luminescence for up to two hours when required.This protocol was tested on two nucleotide sugar transporters, SLC35A2 and SLC35A3, which are golgi-resident type three membrane proteins with N and C termini facing the cytoplasm. There were eight possible tagging options and the resulting fusion proteins could be set together in eight possible ways. Mean relative luminescence units or RLU values corresponding to all the tested and control combinations are shown here.The RLU values obtained for three of the combinations were significantly higher than those obtained for the corresponding control combination. To obtain ratio values for the combinations of interest, the RLU values were divided by the RLU values obtained for the respective controls. Ratios between 10 and 1000 are highly indicative of specific interactions.There were two such combinations supporting the idea that SLC35A2 and SLC35A3 interact. To confirm this data, HA tagged SLC35A2 or SLC35A3 were co-expressed with the combination that resulted in the highest relative luminescence. Varying amounts of the plasmids and coding HA tagged NST variants were used for co-transfection, resulting in a statistically significant dose dependent decrease in RLU values.The most critical thing is to tackle your protein self interest in all possible ways and to assay all possible combination, as some tagging orientation may produce false negative results. Once the best performing combination has been selected, the dynamics of the corresponding interaction can be monitored to, for example identify agents or conditions that disrupt your complex of interest over time.
