Questo manoscritto descrive un protocollo passo-passo per verificare la presenza di interazioni eterologhe tra proteine di membrana di tipo III residenti nel Golgi in cellule umane trasfettate transitoriamente utilizzando la variante più recente del test di complementazione della luciferasi divisa. La procedura è stata ampiamente testata contro i trasportatori di zucchero nucleotidici (NST), ma siamo stati anche in grado di ottenere risultati positivi per altre proteine di membrana di tipo III residenti a Golgi i cui N- e / o C-termini sono rivolti verso il citoplasma.
Il nostro gruppo di ricerca esplora il ruolo degli NST nella glicosilazione delle macromolecole. Le NST sono proteine di membrana di tipo III residenti in Golgi e/o ER con N- e C-termini rivolti verso il lato citoplasmatico della membrana organellare1. Si pensa che gli NST trasportino zuccheri attivati da nucleotidi attraverso le membrane degli organelli per fornire glicosiltransferasi con i loro substrati. Gli NST formano dimeri e/o oligomeri superiori2,3,4,5,6,7,8,9,10. Inoltre, sono state riportate anche interazioni eterologhe tra diversi NST6,11. È stato anche dimostrato che gli NST formano complessi con enzimi di glicosilazione funzionalmente correlati12,13,14. Abbiamo cercato un'alternativa alla tecnica attualmente utilizzata, l'approccio FRET basato sull'imaging a fluorescenza (FLIM), per studiare le interazioni di NST e proteine residenti in Golgi funzionalmente correlate, quindi abbiamo deciso di testare il test di complementazione della luciferasi divisa. Ci ha permesso di identificare una nuova interazione tra un NST e un enzima glicosilazione funzionalmente correlato9.
La modifica più recente del test di complementazione della luciferasi divisa, NanoBiT, è utilizzata nel protocollo qui presentato15. Si basa sulla ricostituzione dell'enzima luciferasi (ad esempio, NanoLuc) dai due frammenti - quello grande, definito come grande BiT o LgBiT, una proteina 17,6 kDa, e quello piccolo, composto da soli 11 amminoacidi, definito come piccolo BiT o SmBiT. Le due proteine di interesse sono fuse con i frammenti complementari ed espresse transitoriamente in una linea cellulare umana. Se le due proteine di fusione interagiscono, una luminescenza viene prodotta in situ con l'aggiunta di un substrato permeabile alle cellule. Questi due frammenti sono stati ottimizzati in modo da associarsi a un'affinità minima a meno che non siano riuniti da un'interazione tra le proteine di interesse a cui sono fusi.
In generale, i metodi basati sulla bioluminescenza hanno alcuni vantaggi rispetto a quelli basati sulla fluorescenza. I segnali bioluminescenti hanno un rapporto segnale-rumore più elevato perché la luminescenza di fondo è trascurabile rispetto al segnale derivato dalla luciferasi16. Al contrario, gli approcci basati sulla fluorescenza di solito soffrono di uno sfondo relativamente alto causato dal fenomeno dell'autofluorescenza. Inoltre, la bioluminescenza è meno dannosa per le cellule analizzate rispetto alla fluorescenza, poiché nel primo caso non è necessario eccitare il campione. Per questi motivi gli approcci bioluminescenti allo studio degli IPP in vivo superano i metodi fluorescenti comunemente usati come Förster Resonance Energy Transfer (FRET) o Bimolecular Fluorescence Complementation (BiFC).
Il nostro protocollo si basa sul riferimento della luminescenza ottenuta per la combinazione proteica di interesse alla luminescenza ottenuta per la combinazione di controllo. Quest'ultimo include una delle proteine testate che viene fusa con un frammento più grande e una proteina di controllo (ad esempio, HaloTag), fusa con un frammento più piccolo. Quest'ultima è una proteina di origine batterica che non dovrebbe interagire con nessuna delle proteine dei mammiferi. L'uso di questa proteina come controllo pone limitazioni alla topologia delle coppie di proteine residenti a Golgi da analizzare. Poiché nelle cellule di mammifero questa proteina è sintetizzata nel citoplasma, entrambe le proteine di interesse dovrebbero avere almeno una coda citoplasmatica.
Questo approccio può essere particolarmente utile per lo screening iniziale degli IPP. Può diventare il metodo di scelta quando le proteine di fusione di interesse sono espresse a livelli che sono semplicemente insufficienti per l'applicazione di altri approcci. Allo stesso modo, il test di complementazione della luciferasi divisa può essere l'opzione migliore se le proteine di interesse sono espresse ad alti livelli, ma ciò influisce negativamente sulla loro localizzazione subcellulare o è noto per forzare interazioni non specifiche. Poiché il frammento più piccolo ha solo 11 amminoacidi, il test di complementazione della luciferasi divisa può essere applicato quando l'uso di tag più grandi è impossibile. Infine, può essere utilizzato per confermare ulteriormente i dati ottenuti utilizzando altre tecniche, come nel caso qui presentato.

L'obiettivo di questo protocollo è quello di esplorare l'applicabilità della variante più recente della complementazione della luciferasi divisa per dimostrare complessi eterologhi formati da trasportatori di zucchero nucleotidici (NST). Queste proteine multitransmembrana residenti in ER e Golgi trasportano gli zuccheri nucleotidici sintetizzati citoplasmaticamente attraverso le membrane degli organelli per fornire enzimi che mediano la glicosilazione con i loro substrati. Our protocol allows to identify and analyze interactions between multi transmembrane proteins of the endoplasmic reticulum and golgi complex in living cells at low expression levels. The main advantage of this technique is it's to special sensitivity, which allows to avoid high over expression levels that are often detrimental to cellular health and may disturb protein subcellular localization. Harvest the adherent HEK293 T-cell culture by trypsinization and re-suspend the cells in a dedicated complete growth medium.Plate the cells onto a clear bottom, white side 96-well plate. Adjust the total number of wells to accommodate all tested combinations and controls, including replicates. Then, culture the cells overnight in standard conditions.On the next day, transfect the cells with the desired combinations of expression plasmids. Dilute the plasmids in a serum-free medium to 6.25 nanograms per microliter for each construct and add the lipid-based transfection reagent. Add an appropriate lipid to DNA ratio.Incubate the plasmids according to manufacturer's instruction. Then, add eight microliters of the lipid DNA mixture to the designated wells. Mix the content of the plate with gentle rotation and culture the cells for 20 24 hours in standard conditions.On the next day, replace the conditioned medium in each well with 100 microliters of a serum-free medium, making sure that the cells do not detach upon medium exchange. Just before measuring luminescence, mix one volume of furimazine with 19 volumes of a dilution buffer. Add 25 microliters of the furimazine working solution into each well, and gently mix the plate by hand or with a orbital shaker.Insert the plate into a luminescence micro plate reader and equilibrate the plate at 37 degrees Celsius for 10 15 minutes. Select the wells to be analyzed and read luminescence with an integration time of 0.3 seconds. Continue to monitor luminescence for up to two hours when required.This protocol was tested on two nucleotide sugar transporters, SLC35A2 and SLC35A3, which are golgi-resident type three membrane proteins with N and C termini facing the cytoplasm. There were eight possible tagging options and the resulting fusion proteins could be set together in eight possible ways. Mean relative luminescence units or RLU values corresponding to all the tested and control combinations are shown here.The RLU values obtained for three of the combinations were significantly higher than those obtained for the corresponding control combination. To obtain ratio values for the combinations of interest, the RLU values were divided by the RLU values obtained for the respective controls. Ratios between 10 and 1000 are highly indicative of specific interactions.There were two such combinations supporting the idea that SLC35A2 and SLC35A3 interact. To confirm this data, HA tagged SLC35A2 or SLC35A3 were co-expressed with the combination that resulted in the highest relative luminescence. Varying amounts of the plasmids and coding HA tagged NST variants were used for co-transfection, resulting in a statistically significant dose dependent decrease in RLU values.The most critical thing is to tackle your protein self interest in all possible ways and to assay all possible combination, as some tagging orientation may produce false negative results. Once the best performing combination has been selected, the dynamics of the corresponding interaction can be monitored to, for example identify agents or conditions that disrupt your complex of interest over time.
