Este manuscrito describe un protocolo paso a paso para verificar la presencia de interacciones heterólogas entre las proteínas de membrana tipo III residentes en Golgi en células humanas transfectadas transitoriamente utilizando la variante más reciente del ensayo de complementación de luciferasa dividida. El procedimiento ha sido probado más extensamente contra transportadores de azúcar de nucleótidos (NST), pero también pudimos obtener resultados positivos para otras proteínas de membrana tipo III residentes en Golgi cuyos N- y / o C-termini se enfrentan al citoplasma.
Nuestro grupo de investigación explora el papel de los NST en la glicosilación de macromoléculas. Los NST son proteínas de membrana tipo III residentes en Golgi y/o ER con N- y C-termini frente al lado citoplasmático de la membrana organelástica1. Se cree que los NST transportan azúcares activados por nucleótidos a través de las membranas de orgánulos para suministrar glicosiltransferasas con sus sustratos. Buscamos una alternativa a la técnica utilizada actualmente, el enfoque FRET basado en imágenes de fluorescencia de por vida (FLIM), para estudiar las interacciones de los NST y las proteínas residentes de Golgi funcionalmente relacionadas, por lo que decidimos probar el ensayo de complementación de luciferasa dividida. Nos permitió identificar una nueva interacción entre un NST y una enzima de glicosilación funcionalmente relacionada9.
La modificación más reciente del ensayo de complementación de luciferasa dividida, NanoBiT, se utiliza en el protocolo presentado aquí15. Se basa en la reconstitución de la enzima luciferasa (por ejemplo, NanoLuc) de los dos fragmentos: el grande, denominado como BiT grande o LgBiT, una proteína de 17,6 kDa, y el pequeño, compuesto de solo 11 aminoácidos, denominado BiT pequeño o SmBiT. Las dos proteínas de interés se fusionan con los fragmentos complementarios y se expresan transitoriamente en una línea celular humana. Si las dos proteínas de fusión interactúan, se produce una luminiscencia in situ tras la adición de un sustrato permeable a la célula. Estos dos fragmentos han sido optimizados para que se asocien con una afinidad mínima a menos que se unan mediante una interacción entre las proteínas de interés a las que se fusionan.
En general, los métodos basados en bioluminiscencia tienen algunas ventajas sobre los basados en la fluorescencia. Las señales bioluminiscentes tienen una mayor relación señal-ruido porque la luminiscencia de fondo es insignificante en comparación con la señal derivada de la luciferasa16. En contraste, los enfoques basados en la fluorescencia generalmente sufren de un fondo relativamente alto causado por el fenómeno de la autofluorescencia. Además, la bioluminiscencia es menos perjudicial para las células analizadas que la fluorescencia, ya que en el primer caso no hay necesidad de excitar la muestra. Por esas razones, los enfoques bioluminiscentes para estudiar los IBP in vivo superan a los métodos fluorescentes comúnmente utilizados como la transferencia de energía por resonancia de Förster (FRET) o la complementación de fluorescencia bimolecular (BiFC).
Nuestro protocolo se basa en referir la luminiscencia obtenida para la combinación proteica de interés a la luminiscencia obtenida para la combinación de control. Este último incluye la de las proteínas probadas que se fusiona con un fragmento más grande y una proteína de control (por ejemplo, HaloTag), fusionada con un fragmento más pequeño. Esta última es una proteína de origen bacteriano que no se espera que interactúe con ninguna de las proteínas de mamíferos. El uso de esta proteína como control plantea limitaciones a la topología de los pares de proteínas residentes de Golgi a analizar. Dado que en las células de mamíferos esta proteína se sintetiza en el citoplasma, ambas proteínas de interés deben tener al menos una cola citoplasmática.
Este enfoque puede ser particularmente útil para el cribado inicial de IBP. Puede convertirse en el método de elección cuando las proteínas de fusión de interés se expresan a niveles que son simplemente insuficientes para que se apliquen otros enfoques. Del mismo modo, el ensayo de complementación de luciferasa dividida puede ser la mejor opción si las proteínas de interés se expresan en niveles altos, pero esto afecta negativamente su localización subcelular o se sabe que fuerza interacciones no específicas. Dado que el fragmento más pequeño tiene solo 11 aminoácidos, el ensayo de complementación de luciferasa dividida se puede aplicar cuando el uso de etiquetas más grandes es imposible. Finalmente, se puede emplear para confirmar aún más los datos obtenidos utilizando otras técnicas, como en el caso presentado aquí.

El objetivo de este protocolo es explorar la aplicabilidad de la variante más reciente de complementación de luciferasa dividida para demostrar complejos heterólogos formados por transportadores de azúcar de nucleótidos (NST). Our protocol allows to identify and analyze interactions between multi transmembrane proteins of the endoplasmic reticulum and golgi complex in living cells at low expression levels. The main advantage of this technique is it's to special sensitivity, which allows to avoid high over expression levels that are often detrimental to cellular health and may disturb protein subcellular localization. Harvest the adherent HEK293 T-cell culture by trypsinization and re-suspend the cells in a dedicated complete growth medium.Plate the cells onto a clear bottom, white side 96-well plate. Adjust the total number of wells to accommodate all tested combinations and controls, including replicates. Then, culture the cells overnight in standard conditions.On the next day, transfect the cells with the desired combinations of expression plasmids. Dilute the plasmids in a serum-free medium to 6.25 nanograms per microliter for each construct and add the lipid-based transfection reagent. Add an appropriate lipid to DNA ratio.Incubate the plasmids according to manufacturer's instruction. Then, add eight microliters of the lipid DNA mixture to the designated wells. Mix the content of the plate with gentle rotation and culture the cells for 20 24 hours in standard conditions.On the next day, replace the conditioned medium in each well with 100 microliters of a serum-free medium, making sure that the cells do not detach upon medium exchange. Just before measuring luminescence, mix one volume of furimazine with 19 volumes of a dilution buffer. Add 25 microliters of the furimazine working solution into each well, and gently mix the plate by hand or with a orbital shaker.Insert the plate into a luminescence micro plate reader and equilibrate the plate at 37 degrees Celsius for 10 15 minutes. Select the wells to be analyzed and read luminescence with an integration time of 0.3 seconds. Continue to monitor luminescence for up to two hours when required.This protocol was tested on two nucleotide sugar transporters, SLC35A2 and SLC35A3, which are golgi-resident type three membrane proteins with N and C termini facing the cytoplasm. There were eight possible tagging options and the resulting fusion proteins could be set together in eight possible ways. Mean relative luminescence units or RLU values corresponding to all the tested and control combinations are shown here.The RLU values obtained for three of the combinations were significantly higher than those obtained for the corresponding control combination. To obtain ratio values for the combinations of interest, the RLU values were divided by the RLU values obtained for the respective controls. Ratios between 10 and 1000 are highly indicative of specific interactions.There were two such combinations supporting the idea that SLC35A2 and SLC35A3 interact. To confirm this data, HA tagged SLC35A2 or SLC35A3 were co-expressed with the combination that resulted in the highest relative luminescence. Varying amounts of the plasmids and coding HA tagged NST variants were used for co-transfection, resulting in a statistically significant dose dependent decrease in RLU values.The most critical thing is to tackle your protein self interest in all possible ways and to assay all possible combination, as some tagging orientation may produce false negative results. Once the best performing combination has been selected, the dynamics of the corresponding interaction can be monitored to, for example identify agents or conditions that disrupt your complex of interest over time.
