このプロトコルは、AML細胞の表面上のNKG2DLを検出する2つの異なる方法を示しています。この技術は、既知のNKG2DLおよび未知のNKG2DLをすべて検出するための、迅速で使いやすい染色法を提供します。この方法により、バルクAML細胞から白血病幹細胞を分離することができ、これらの細胞をさらに特徴付けることを可能にする。ビオチンとNKG2D融合タンパク質チューブを室温で解凍します。NKG2D FC粉末を素早くスピンダウンし、500マイクロリットルのPBSを加え、粉末を再構成し、P-1000マイクロピペットを使用して十分に混合します。NKG2DL融合タンパク質100マイクロリットルをビオチンチューブに加えて、1ミリリットル当たり10マイクログラムの最終濃度を得て、溶液をP-100マイクロピペットと完全に混合します。AML細胞を解凍するには、液体窒素貯蔵から原発AMLを含む凍結を取り除き、すぐに37°Cの水浴に入れます。チューブを水の中でゆっくりと前後に動かし、小さな氷の結晶だけが残るまでバイアルの内容物を解凍させます。解凍した細胞をチューブを含む培地にすぐに移し、1ミリリットルの培地を使用してバイアルをすすいします。細胞を10分間Gの300倍に遠心し、ペレットを乱さずに上清を捨てる。10%FCSを含むRPMI培地を5ミリリットルで細胞を洗浄し、遠心分離を繰り返します。染色バッファーを用いて細胞ペレットを、1ミリリットル当たり7番目の細胞に0.5倍の最終濃度まで再懸濁する。次に細胞懸濁液の100マイクロリットルを細胞培養96ウェルU-底板に移し、プレートを300倍Gで10分間遠心した。ペレットを邪魔することなく上清を捨てます。ビオチン化されたNKG2D融合タンパク質のマスターミックスを準備し、細胞が井戸当たり50マイクロリットルの最終体積で再懸濁され、最終的なNKG2D濃度は1ミリリットル当たり10マイクログラムになるようにします。準備したマスターミックスを加え、200マイクロリットルのピペットで細胞ペレットを再中断します。インキュベートおよび遠心分離機は、テキスト原稿に記載されているようにプレートを培養する。細胞が50マイクロリットルの最終容積で再懸濁されるようにストレプトアビジンPEを使用してマスターミックスを準備します。マスターミックスをセルに加え、300マイクロリットルのマルチチャンネルピペットでペレットを再中断します。プレートを遠心分離し、上清を捨て、300マイクロリットルのマルチチャンネルマイクロピペットを使用して、200マイクロリットルの染色バッファープラス7-AADで細胞ペレットを再懸濁させます。次にフローサイトメトリー装置を用いて細胞を解析する。分析されたAMLサンプルはCD34とNKG2DLでは正であるが、負の亜集団も存在し、合計で4つの異なる集団がある。典型的な格言戦略は、FSCおよびSSCを介して細胞の主要な集団を選択することから始まります。ダウンストリーム解析では、ダブレットとデッド セルは除外されます。ゲートは、正の細胞を正しく識別するためにFMOコントロールを使用して調整されます。CD34対NKG2DLに陽性の細胞の蛍光強度をここで強調しています。CD34に陽性であるAML細胞は、NKG2DLの表面発現が低く、NKG2DL発現がステムの欠如と関連していることを示す。3つの一次AMLサンプルは、プールされた抗NKG2DL抗体染色に対して、融合タンパク質染色に対して19.8、49.8、および89.4%の陽性事象を実証しました。一方、単一のリガンド染色は、リガンドに基づいて変化する割合で92%までの陽性事象の範囲を示した。この方法を試みるとき、原細胞は壊れやすく、このステップの間に簡単に死ぬ可能性があるため、プライマリAML細胞を解凍する際に迅速であることが非常に重要です。このプロトコルを実行した後、NKG2DLシグナルに基づく細胞の並べ替えを行い、さらに集団を調査することができます。
