Ulike primære hepatocytisolasjonsprotokoller er utviklet. De har også blitt holdt optimalisert og tilpasset basert på ulike behov (tabell 1). Isolasjonsprotokoller består vanligvis av to deler: perfusjon (inkludert enzym fordøyelse) og rensing.

Leveren fungerer som en av de viktigste organene i virveldyrkroppen på grunn av den vitale rollen den spiller i mange livsstøttende funksjoner som mat fordøyelse, blodsirkulasjon og avgiftning. Bruk av musen primære hepatocyte in vitro kultur er stadig mer populært i studier av karbohydrat metabolisme og leverkarsinom. Derfor er det viktig å utvikle en praktisk metode for mus primær hepatocytisolasjon samtidig som den opprettholder sin medfødte fysiologiske funksjon. På grunn av sin funksjon som et knutepunkt for glukosemetabolisme, er leveren også sentral i glukoseproduksjon og lagring1. Eksperimenter med primære hepatocytter in vitro er et must for de fleste glukosemetabolismestudier. Derfor har ulike forskningsgrupper i årevis utviklet protokoller for mus primær hepatocytisolasjon.
Den generelle prosedyren for mus hepatocytisolasjon er å først skylle ut blod i leveren med en isosmotisk væske som fosfatbufret saltvann (PBS) eller Hanks ' Balansert saltoppløsning (HBSS) og deretter bruke kollagenaseholdig løsning for å dissosiere hepatocytter. Disse protokollene deler en generell prosedyre, men varierer i reagenser og trinn basert på forskjellige behov. Det tar imidlertid tid å forberede nødvendige reagenser og utføre isolasjonstrinn. I utviklingen av den nåværende protokollen ble effektivitet satt som en prioritet, med alle reagenser klare til bruk og tilgjengelig fra markedet, og så få trinn som mulig. an improved time- and labor- efficient protocol for mouse primary hepatocyte isolation

Primære hepatocytter brukes mye i leverin vitro forskning, spesielt i glukose metabolisme studier. En basisteknikk er tilpasset basert på ulike behov, som tid, arbeid, kostnader og primær hepatocyttbruk, noe som resulterer i ulike primære hepatocytisolasjonsprotokoller. Imidlertid er de mange trinnene og tidkrevende reagenspreparatene i primær hepatocytisolasjon store ulemper for effektivitet. Etter å ha sammenlignet forskjellige protokoller for fordeler og ulemper, ble fordelene ved hver av dem kombinert, og en rask og effektiv primær hepatocytisolasjonsprotokoll ble formulert. I løpet av bare ~ 35 min kan denne protokollen gi så mye, om ikke mer, sunne primære hepatocytter som andre protokoller. Videre validerte glukosemetabolismeforsøk utført ved hjelp av de isolerte primære hepatocyttene nytten av denne protokollen i in vitro levermetabolismestudier. Vi har også gjennomgått og analysert betydningen og formålet med hvert trinn i denne studien, slik at fremtidige forskere kan optimalisere denne protokollen ytterligere basert på behov.

Primære hepatocytter er et verdifullt verktøy for å studere leverrespons og metabolisme in vitro. Ved hjelp av kommersielt tilgjengelige reagenser ble det utviklet en forbedret tids- og arbeidseffektiv protokoll for mus primær hepatocytisolasjon.

Tidsmessig kan isolasjonsprosessen være tidkrevende og energien kostbar.Denne protokollen kan gi en effektiv måte å isolere primære hepatocytter for mus på. Det er tids- og arbeidsvennlig. Krever svært få trinn med alle regioner kommersielt tilgjengelige.Begynn med å bruke den peristaltiske pumpen til å begynne å pumpe oppvarmet dobbelt destillert vann med en hastighet på 4 ml per minutt, i fem minutter. Bytt deretter pumperøret fra vann til oppvarmet perfusjonsmedium. Overvåking av luftboblen mellom vann og perfusjonsmedium.Vil bidra til å spore strømmen av perfusjonsmediet gjennom systemet. Deretter desinfiserer magen til en bedøvet åtte uker gammel C57 svart 6J kvinnelig mus med 70% etanol. Fortsett å pumpe til alle 25 ml av kollagenalissedispasemediet er utarmet.Deretter isolerer hele leveren uten galleblæren. Og plasserte den i 30 ml vaskemedium i en Petri-tallerken på is. Bruk tang rive leveren i stykker for å frigjøre primære hepatocytter i løsningen.På dette stadiet blir vaskemediet til en overskyet løsning som inneholder frigjorte primære hepatocytter og små leverstykker. Filtrer denne overskyete løsningen gjennom en 70 mikron cellesil, i 20 ml 1X per kall HBSS i et 50 ml rør på is. Og bland løsningen ved å invertere røret 20 ganger.Deretter sentrifugerer løsningen. Deretter aspirerer du supernatanten i vevskulturhetten. Og vask pelletsen med 30 ml kaldt vaskemedium.Pellet cellene igjen med sentrifugering. Fjern supernatanten, og re-suspendere pellet i 25 ml kulturmedium. Tell cellene og plate dem på ønskede kulturplater.Ifølge den eksperimentelle designen. Etter plating ble isolerte primære hepatocytter fast festet med en time og fullstendig utvidet etter 12 timer. I de isolerte hepatocyttene ble mRNA-nivåene av hepatocyttmarkører, som transthyretin, CD95, ASGR1 og ASGR2 betydelig økt sammenlignet med hele leveren.I kontrast var tilstedeværelsen av immunceller, stellateceller og endotelceller lavere. Som det fremgår av en kraftig nedgang i CD45, kollagen type 1 alfa 1, og endotelcelle 2 kinase 2 mRNA nivåer. Antyder at denne protokollen i stor grad kan redusere forstyrrelser fra andre leverceller.Etter plating ASGR1 og ASGR2 nivåer redusert med tiden, men forbli sammenlignbar med hele leveren til 12 timers tidspunktet. Spesielt for ASGR1. Uttrykksnivået for membranbundet CD81 var konsistent i opptil 48 timer.Til sammenligning økte bompengelignende reseptor 4-uttrykk generelt og ble høyere enn in vivo-nivåer etter 48 timer. Insulinfølsomhetsanalyser viste at insulin signifikant fremmet fosforylering av begge AKT ved serin 473. Og gaffeltrhead boks 01 på serine 256, i hepatocyttene.Indikerer følsomheten til primære hepatocytter til insulin. Fosfoenolpyruvat karboksykinase proteinnivåer økte betydelig etter glukagonbehandling. Antyder at glukoseproduksjonsveien ble aktivert.Denne aktiveringen ble ytterligere bekreftet av økt glukoseproduksjon. Og med andre glukoseproduksjonstimulatorer som Forskolin+IBMX. Denne protokollen kan spare både tid og energi for studier ved hjelp av musen primære hepatocytter.Etter denne protokollen kan relaterte eksperimenter som hypotetisk glukosemetabolisme og farmasøytiske biomarkøranalyser utføres.
