VAT는 신진대사 조절과 염증에 중요한 역할을 합니다. 비만과 관련된 만성 염증에서 지방 세포와 면역 세포의 역할에 대한 관심이 증가함에 따라 AT에 존재하는 SVF와 지방 세포를 분리하는 다양한 기술이 개발되었습니다. 그러나 대부분의 기술에서는 단일 절차로 동일한 VAT 생검에서 다운스트림 응용 분야에 사용할 수 있는 이 두 가지 다른 세포 세트를 얻을 수 없으며, 이는 지방 세포와 SVF 세포 ?...

백색 지방 조직은 지방 세포 또는 지방 세포뿐만 아니라 대식세포, 조절 T 세포(Tregs)와 같은 기타 면역 세포, 호산구, 지방세포, 섬유아세포로 구성된 이질적인 세포 집단을 포함하는 기질 혈관 분획(SVF)으로 알려진 비지방 세포 분획으로 구성되어 있으며 혈관 및 결합 조직으로 둘러싸여 있습니다 1,2. 지방 조직(AT)은 이제 다른 세포에서 생성되어 조직으로 방출되는 아디포카인, 사이토카인 및 마이크로RNA를 통해 대사 및 염증과 관련된 생리적 과정을 조절하는 기관으로 간주되며 자가분비, 파라크린 및 내분비 효과 3,4. 인간의 경우 백색 지방 조직은 피하 지방 조직(SAT)과 내장 지방 조직(VAT)으로 구성되며 중요한 해부학적, 분자적, 세포적, 생리학적 차이가 있습니다 2,5. SAT는 인간 AT의 최대 80%를 차지하는 반면, VAT는 주로 장간막과 난막에 있는 복강 내에 위치하며 신진대사가 더 활발하다6. 또한 VAT는 체중, 인슐린 감수성, 지질 대사 및 염증에 상당한 영향을 미치는 매개체를 분비하는 내분비 기관입니다. 결과적으로, 부가가치세 누적은 복부 비만과 제2형 당뇨병, 대사 증후군, 고혈압 및 심혈관 질환 위험과 같은 비만 관련 질병을 유발하며, 이는 비만 관련 사망률을 더 잘 예측할 수 있는 지표이다 6,7,8,9.
항상성 상태에서는 지방세포, 대식세포 및 기타 면역세포가 협력하여 항염증 매개체의 분비를 통해 VAT 대사를 유지한다10. 그러나 과도한 VAT 확장은 활성화된 T 세포, NK 세포 및 대식세포의 모집을 촉진합니다. 실제로, 희박한 VAT에서 대식세포의 비율은 5%인 반면, 비만에서는 이 비율이 50%까지 상승하며, 대식세포가 항염증제에서 전염증성 표현형으로 분극되어 만성 염증 환경을 생성합니다10,11.
