La estrategia de detección rápida permite la detección temprana de floraciones de cianobacterias y cianotoxinas relacionadas en las muestras de agua, ya sea en esa matriz orgánica, como los mariscos y otros productos pesqueros. Las floraciones de cianobacterias se deben al crecimiento excesivo de las cianobacterias, que pueden vivir en cualquier tipo de ambiente. Y han surgido como un problema ambiental en todo el mundo en los últimos 15 años.
A medida que el número de floraciones de cianobacterias dañinas ha aumentado durante los últimos años, la necesidad de la detección temprana, que es clave para abordar la floración y la propagación tóxica, se ha vuelto más urgente. Nuestra estrategia de primera dirección combina la tecnología técnica de detección remota y proximal con el análisis químico y bioinformático de laboratorio en un flujo de trabajo integrado único. Todo el proceso es seguro.
Se toman las medidas de seguridad adecuadas para evitar la inhalación de aerosoles y el contacto con la piel durante el muestreo y el análisis de laboratorio. Para la recuperación de datos, primero localice el área de destino en un mapa mundial global y recupere los datos de varios conjuntos de datos de teledetección públicos y privados para la fecha de recopilación. Después de la recuperación, procese los datos sin procesar, calcule los índices multiespectrales y clasifique la información resultante.
A continuación, defina los sitios de muestreo en el mapa temático generado. Para la recolección de muestras, transporte el equipo al sitio de muestreo seleccionado en el laboratorio móvil y planifique la ruta de vuelo del dron para realizar un estudio de área macro. En el sitio, utilice varios drones equipados con diferentes cargas útiles para realizar misiones de vuelo.
Y utilice las imágenes adquiridas por el dron para validar la presencia y extensión de la floración y para identificar puntos de muestreo precisos. En el punto de muestreo identificado, póngase el equipo de protección personal apropiado y recoja tres muestras de agua de 500 mililitros de cada sitio. Mida varios parámetros ambientales como la temperatura del aire y del agua y el pH y la salinidad del sitio.
A continuación, almacene las muestras recogidas en el laboratorio móvil para su transporte al laboratorio universitario. Prepare las diapositivas y examine las muestras con el microscopio móvil del laboratorio equipado de una cámara fotográfica digital para permitir el análisis taxonómico microscópico y la identificación de la especie presente dentro de las muestras en base de su color azul-verde, forma de la célula, y pelotillas del tamaño. Una vez identificadas las especies de cianobacterias recogidas dentro de las muestras, en el laboratorio universitario, centrifuga las muestras y transfiera cada sobrenadante a un nuevo recipiente sin perturbar los pellets de la muestra.
Añadir 500 mililitros de butanol a cada sobrenadante de muestra y transferir cada solución que se extraerá en un embudo separador. Después de agitar y colocar los embudos en posición vertical en abrazaderas de anillo individuales, permita que las fases acuosas drenen en matraces erlenmeyer individuales. Después de repetir la separación de capas tres veces, concentre las fases orgánicas al vacío y sopesa.
Para la extracción de muestras con disolventes orgánicos, agregue 50 mililitros de metanol fresco a cada pellet de muestra y sonice las muestras en un baño de hielo. Después de cinco minutos, agregue 50 mililitros de metanol fresco a cada muestra y agite suavemente el matraz antes de filtrar las soluciones a través de trozos individuales de papel de filtro y recoger los filtrados en frascos redondos de fondo. Después de filtrar cada muestra dos veces más, como se acaba de demostrar, analizar los extractos de la muestra por cromatografía líquida y espectrometría de masas en tándem de alta resolución de acuerdo con los protocolos estándar.
Luego genere una red molecular utilizando la plataforma social global de productos naturales y use las herramientas apropiadas para analizar la red resultante y los datos de espectrometría de masas en tándem para identificar cualquier toxina que se determine que está presente dentro de las muestras recolectadas. La estrategia propuesta fue validada por los resultados obtenidos en el programa de monitoreo costero, activo en la región de Campania en el sur de Italia de 2015 a 2021. Se generó un flujo de trabajo visual que vincula las técnicas con los resultados producidos.
Durante las campañas de monitoreo posteriores, cada paso se optimizó con el objetivo de una detección rápida. La optimización del flujo de trabajo de teledetección permitió reducir el número de plataformas y misiones, al tiempo que mejoraba el nivel de producto generado. Por ejemplo, esta estrategia de detección rápida facilita una transición del uso de varias plataformas aéreas diferentes a plataformas de satélites y drones solamente.
Y desde el uso de varios índices especializados multiespectrales diferentes hasta la clorofila más informativa y los mapas temáticos de índice de vegetación de diferencia normalizada. En paralelo, el flujo de trabajo se redujo de requerir un análisis metogenómico de 16 S a usar la observación microscópica solo para determinar la comunidad de cianobacterias. Y el nuevo flujo de trabajo químico utiliza redes moleculares basadas en LCMS para una detección rápida y precisa de cianotoxinas.
Esta estrategia permite el estudio de las cianobacterias como bioindicadoras de la contaminación, particularmente dentro de áreas en las que la presencia de la floración está relacionada con procesos de eutrofización y utilizada por presión antropogénica. Esta estrategia multidisciplinaria requiere la integración combinada de diferentes técnicas, tecnología y experiencia en un flujo de trabajo único para su implementación exitosa. La estrategia de detección rápida es útil para prevenir problemas de salud comunitarios debido a floraciones dañinas de cianobacterias y para monitorear grandes áreas en poco tiempo.
