BC細胞株の2次元培養、膜挿入共培養、三次元スフェロイドおよびオルガノイドなどの現在のインビトロBCモデルは、いずれも在来乳房組織アーキテクチャを再現しないため、NIHのMPS基準を満たしていない。細胞外マトリックス(ECM)がこれらのシステムに添加されると、乳房ECMは使用されません。代わりに、コラーゲンゲルと原膜マトリックスが使用されます。
患者由来異種移植片(PDX)などの生体外システムの電流は、同様に、マウス乳腺組織がヒトの乳房と大きく異なるため、NIHのMPS基準を満たしていない。さらに、免疫系とBCの相互作用は腫瘍の発症の鍵としてますます認識されているが、PDX腫瘍を発生させる免疫不全マウスモデルは成熟したT細胞、B細胞、およびナチュラルキラー細胞を欠いている。さらに、PDXは原発性乳房腫瘍を維持および拡張することを可能にするが、得られたPDX腫瘍は原発性マウス間質細胞およびECM5とに浸潤される。
