以前に特徴付けられるHORCプロトコル6、7、8、9は、外科トレフィンを使用して、基礎となるRPE-脈絡膜および鎖膜からレチナを分離することに関与していた。しかし、これらの基礎構造によって提供されるサポートがなければ、半透明のretinaは薄っぺらになり、取り扱いにくく、鉗子などのツールは容易にその完全性を破壊し得る。さらに、RPEを含まない培養での分離性レチナは、神経節細胞アポトーシスおよび感光体変性10、11、12を引き起こすことが示されている。したがって、retinal完全性の損失を最小限に抑え、in vivo環境をよりよく模倣する代替HORCプロトコルが有用であろう。これは、外植の取り扱い中の身体的傷害が人工物を導入する可能性があるため、疾患のメカニズムを研究する際に特に重要です。そこで本研究の目的は、外植の取り扱いと培養の際のレチナル完全性を保護するために、RPE-脈絡膜と皮膜を含む新しいHORCモデルを開発することであった。
この目的を達成するために、残留性の植物と下層のRPE-脈絡膜と皮膜の間に「挟まれた」植物の再チナリを抽出した。サンドイッチの外植では、網膜剥離と折りたたみを防ぐために網膜の重さを量るのに対し、硬い線維性強膜は構造的支持のための足場と鉗子の接触点の両方として機能する。また、動物モデルは、培養中にRPEを保持することで、残存性の変性やグリア増殖を防ぐことができる、低酸素および炎症10、11、12などの危険信号に対するミュラー細胞の応答を示している。
モデルを特徴付けるために、サンドイッチの残膜外植体をヘマトキシリンとエオシン(H&E)で染色し、解剖学的構造を評価し、免疫組織化学(IHC)を実施し、 末端デオキシヌクレオチジルトランスファーーゼdUTPニックエンドラベリング(TUNEL、アポトーシス細胞マーカー)、グリア性フィブリリン酸タンパク質(GFAP、眼炎およびミュラー細胞活性化マーカー)、およびミュラー細胞完全性のマーカーであるビメンチンを用いた外植体を標識する。このモデルが分子疾患の徴候を発症するように誘導できるかどうかを判断するために、外植体は、炎症促進性サイトカイン(Cyt)、インターロイキン-1β(IL-1β)および腫瘍壊死因子α(TNF-α)を有する高グルコース(HG)に曝露された。ルミネックスアッセイは、培養培地中に放出されたサイトカインを測定するためにDRモデルで使用された。

Characterization of a Novel Human Organotypic Retinal Culture Technique

この技術は、すべてのレチン基層および細胞タイプをその現場に保存し、動物およびインビトロモデルと比較してより臨床的に関連する。このプロトコルの主な利点は、健康な、病気のretinaを比較する際に不可欠である組織の取り扱い中の、整体性の破壊を最小限に抑えるということです。特定の条件下で組織を培養することによって、疾患を模倣することができます。これは、新しい薬物療法のテストを可能にします。解剖とサンプル収集手順は複雑です。したがって、書かれたプロトコルを理解し、手順を正常に実行するためには、視覚的なデモンストレーションが重要です。この手順のデモンストレーションは、私の研究室の博士課程2年生のカリス・クオです。Dulbeccoの修飾イーグル培地/栄養混合物F-12と抗生物質と抗ミコティック混合物を1回含む培地を調製します。まず、24ウェルプレートに500マイクロリットルの培地を入れ、加湿インキュベーターで摂氏37度と二酸化炭素5%で平衡化します。外植抽出の前に培地を準備しておくことは、その後の添加時にレティナの外れを避ける。裂け目の外植を抽出するには、アイリスとレンズを上向きにして、ONHがペトリ皿に接触したペトリ皿にアイカップを置くことから始めます。鉗子を使用して手足でアイカップをしっかりと保持し、手足の外縁に沿って周回に小さなカットを行うことによって虹彩とレンズを取り外します。虹彩とレンズを慎重に取り外し、レティナを邪魔しないようにしてください。明るい白色の光源を使用してONHを識別し、より簡単に取り扱いのためにペトリ皿を回転させるONHに向かって4つの象限で切開します。眼を慎重に広げて平らにします。レチナの代わりに強膜に鉗子を適用して、レチナルの完全性を乱さないようにします。調製した培地を含むプレートをインキュベーターから取り出します。レチナルフォールドのない領域のレチナに外科的トレフィンを置き、強膜を貫通するために強く押すと、ペトリ皿を突破する際に割れる音が発生するはずです。トレフィンを回転させて、強膜が完全に貫通し、残りの部分から外植が分離されるようにします。強膜で鉗子を塗布し、サンドイッチの残りの外植を培養液に移す。採取したサンドイッチの残り葉植物を加湿した5%の二酸化炭素インキュベーターで最大72時間摂氏37度で培養する。ヘマトキシリンとエオシン染色は、サンドイッチの残膜外植が完全性を維持し、GCLからONLまでの明確なラメラ構造を内側および外側の核層にコンパクトな核で保存することを示した。網膜の完全性が破壊されたのは、網膜が基礎となるRPE脈絡膜および皮膜から剥離した同じサンプルの端で発見され、INLおよびONLにおける網膜の厚さと核の喪失が減少したことを示した。細胞アポトーシスを示すTUNEL陽性細胞核は、基底条件で培養されたレチナル外植体に見られ、72時間後も持続的な細胞活力を示した。GFAP標識は、グリア線維症を伴わないGCLおよびIPLに限定された、ONLへのGFAPの拡張発現として同定される病理の徴候である。Vimentinは神経節細胞層から核外層まで高く発現し、正常な残線組織に見られるように保存されたミューラー細胞の完全性を示した。Luminex磁気アッセイは、IL-18、VEGF、IL-6、およびIL-8レベルが24時間後に高グルコースおよび炎症促進サイトカインにおけるサンドイッチの外植培養物のベースラインを大幅に上回って増加したことを示した。72時間後、IL-18およびVEGFレベルは有意に増加したが、IL-6およびIL-8レベルでは有意な差は見つからなかった。この手順を行う場合、十分な残留のビトリアスを残し、これは、リチナを量り、RP脈絡膜から剥離して培養媒体に浮遊しないようにする。培養されたレチナは免疫神学的化学および細胞の細胞学を用いてさらに特徴づけることができ、一方で培養培地中に放出される炎症性サイトカインは、ルミネックス磁気アッセイを用いて測定することができる。このモデルは、動物およびインビトロモデルに比べて臨床的に翻訳可能であり、疾患に対する新しい治療薬の有効性をテストするのに適しています。
