L’application de la technologie d’édition du génome fournit un outil efficace pour atteindre des mutants de gènes cibles chez diverses espèces. L’émergence du système CRISPR (Clustered Regular Interspaced short palindromic repeats)/Associated Protein 9 (Cas9) fournit une nouvelle méthode pour manipuler les génomes1. Le système CRISPR/Cas9 se compose d’un ARN guide (aRNg) et de l’endonucléase Cas92,3, tandis que l’ARNg peut être divisé en deux parties, un ARN CRISPR complémentaire cible (ARN crnc) et un ARNnc transactivant (tracrRNA). L’ARNg s’intègre à l’endonucléase Cas9 et forme une ribonucléoprotéine (RNP). Avec l’ARNg, l’endonucléase Cas9 peut être dirigée vers un site spécifique du génome via la complémentation de base. Les domaines RuvC et HNH du Cas9 coupent le site cible du génome trois bases avant la séquence protospacer-adjacent motif (PAM) et créent une rupture double brin (DSB). Le clivage de l’ADN peut ensuite être réparé par deux mécanismes, la jonction terminale non homologue (NHEJ) ou la réparation dirigée par homologie (HDR)4. La réparation du DSB introduit des insertions ou des délétions comme moyen d’inactiver le gène ciblé, ce qui peut entraîner une perte complète de la fonction du gène. Par conséquent, la héréditaire et la spécificité du système CRISPR/Cas9 en font une méthode robuste pour caractériser les fonctions des gènes in vivo et analyser les interactions géniques5.
Avec de nombreux mérites, le système CRISPR/Cas9 a été appliqué à divers domaines, y compris la biomédecine6,7, la thérapie génique8,9 et l’agriculture10,11,12, et a été utilisé pour divers systèmes biologiques, y compris les micro-organismes13, les plantes14,15, les nématodes16 et les mammifères17 . Chez les invertébrés, de nombreuses espèces d’insectes ont été soumises à l’édition du génome CRISPR/Cas9, comme la mouche des fruits Drosophila melanogaster et au-delà18,19,20,21,22.
Helicoverpa armigera est l’un des ravageurs les plus destructeurs au monde23 et endommage de nombreuses cultures, notamment le coton, le soja et le sorgho24,25. Avec le développement de la technologie de séquençage, le génome de H. armigera, ainsi que celui d’une gamme d’espèces d’insectes lépidoptères, ont été séquencés complètement26,27,28,29. Un grand nombre de gènes de résistance et de récepteurs olfactifs ont été identifiés et caractérisés à partir de ces insectes au cours des dernières années19,27,28,29. Certains gènes liés à la résistance ont été identifiés chez H. armigera, tels que les gènes codant pour la cadhérine30, un transporteur de cassettes liant l’ATP31,32, ainsi que HaTSPAN133. L’élimination de ces gènes à l’aide de la technologie CRISPR/Cas9 entraîne un niveau élevé de résistance à la toxine Bacillus thuringiensis (BT) chez les souches sensibles. En outre, Chang et al. (2017) ont éliminé un récepteur de phéromones, ce qui a validé sa fonction significative dans la régulation du temps d’accouplement19. Ces rapports suggèrent que CRISPR/Cas9 peut agir comme un outil efficace pour étudier la fonction des gènes in vivo dans les systèmes d’insectes. Cependant, une procédure détaillée pour la modification CRISPR/Cas9 dans les systèmes d’insectes reste incomplète, ce qui limite sa gamme d’applications en génomique fonctionnelle des insectes.
Nous présentons ici un protocole pour éliminer un gène fonctionnel chez H. armigera à l’aide du système CRISPR/Cas9. Un protocole détaillé étape par étape est fourni, y compris la conception et la préparation d’amorces spécifiques aux gènes pour la production d’ARNg, la collecte d’embryons, la microinjection, l’élevage d’insectes et l’identification des mutants. Ce protocole sert de référence précieuse pour manipuler tout gène fonctionnel chez H. armigera et peut être étendu à d’autres espèces de lépidoptères.

Le ver du coton, Helicoverpa armigera, est l’un des ravageurs les plus destructeurs au monde. Une combinaison de génétique moléculaire, de physiologie, de génomique fonctionnelle et d’études comportementales a fait de H. armigera une espèce modèle chez le Lepidoptera Noctuidae. Pour étudier les fonctions in vivo et les interactions entre différents gènes, la technologie d’édition du génome des courtes répétitions palindromiques régulièrement espacées (CRISPR) / associée à la protéine 9 (Cas9) est une méthode pratique et efficace utilisée pour effectuer des études génomiques fonctionnelles. Dans cette étude, nous fournissons une méthode systématique étape par étape pour compléter l’élimination du gène chez H. armigera en utilisant le système CRISPR / Cas9. La conception et la synthèse de l’ARN guide (ARNg) sont décrites en détail. Ensuite, les étapes suivantes consistant en la conception d’amorces spécifiques à un gène pour la création d’ARN guide (ARNg), la collecte d’embryons, la micro-injection, l’élevage d’insectes et la détection de mutants sont résumées. Enfin, des conseils de dépannage et des notes sont fournis pour améliorer l’efficacité de l’édition de gènes. Notre méthode servira de référence pour l’application de l’édition du génome CRISPR/Cas9 chez H. armigera ainsi que chez d’autres papillons lépidoptères.

Ce protocole fournit des étapes détaillées pour obtenir des lignées knock-out de gènes de H. armigera à l’aide de la technologie CRISPR/Cas9. Les résultats représentatifs obtenus par ce protocole sont résumés pour la sélection de l’ADNg, la collecte et l’injection d’embryons, l’élevage d’insectes et la détection de mutants.

Le ver du coton est l’un des ravageurs les plus destructeurs au monde. L’édition du génome avec la technologie CRISPR/Cas9 permet de poursuivre les recherches sur la fonction des gènes et l’interaction entre les différents gènes de cet organisme. Les deux principaux avantages de cette technique sont qu’elle a une grande spécificité et qu’elle est héréditaire.Afin de choisir les cibles sgRNA, rendez-vous sur le site web en ligne de CRISPR pour rechercher les sites guides possibles et les exons proches des cinq régions principales non traduites du gène. Entrez la séquence d’exons dans la zone de texte et sélectionnez le génome cible dans Helicoverpa armigera. Choisissez l’option PAM pour 20 BPNGG et laissez les autres paramètres sur les paramètres par défaut, selon les manuels d’utilisation du site Web.Comparez les séquences de guidage prédites à partir du logiciel et choisissez une séquence de guide de 20 paires de bases contenant une ou deux guanines près de la région non traduite des cinq principales avec la plus grande efficacité prédite et le moins d’incohérences pour améliorer l’efficacité de l’édition et réduire l’édition hors cible. Pour la préparation des embryons, sélectionnez 50 individus en bonne santé parmi des mâles de trois jours et des femelles de deux jours, et mélangez-les dans une cage en filet propre. Placez un morceau de coton humide contenant une solution de sucre à 10% dans la cage.Couvrez les cages en filet avec de la gaze et fixez la gaze avec un élastique. Vaporisez de l’eau sur la gaze pour la garder humide. Coupez un chiffon noir dans une taille appropriée et remplacez la gaze humide par ce chiffon noir pour permettre une position libre des ovules pendant 30 minutes.Collez du ruban adhésif double face sur une lame de microscope. À l’aide de pinces, collez les patchs avec des œufs en rangée sur la surface du ruban adhésif double face. Appuyez sur la marge de chaque patch pour vous assurer qu’ils adhèrent fermement à la bande.Recueillir 50 à 100 œufs par lame de microscope. Avant la microinjection, gardez la lame du microscope sur la glace pour retarder le développement des embryons. Préparez l’aiguille en tirant un verre capillaire à l’aide d’un micro-pipette comme décrit dans le manuscrit du texte.Broyer la pointe de l’aiguille à l’aide d’un micro broyeur jusqu’à une pointe à arêtes vives. Préparer des solutions injectables en ajoutant deux microlitres de protéine Cas9 commercialisée et d’ARNng à de l’eau sans RNace dans un tube PCR pour obtenir un mélange de volume de 10 microlitres. Bien mélanger en pipetant et mettre sur la glace.Définissez les paramètres du micro-injecteur électronique. Chargez deux microlitres du mélange dans une aiguille à l’aide d’une pointe de pipette de microchargeur, en évacuant autant d’air résiduel que possible dans la pointe de l’aiguille. Connectez l’aiguille d’injection à un micromanipulateur et assurez une connexion étroite entre les deux parties.Placez une lame dans une boîte de Pétri et mettez-la sur la scène du microscope. Insérez soigneusement la pointe de l’aiguille dans l’hémisphère supérieur d’un embryon à un angle de 45 degrés. Appuyez sur la pédale pour délivrer le mélange dans l’embryon, ce qui entraîne une légère expansion de l’embryon.Retirez immédiatement l’aiguille de l’embryon et déplacez la boîte de Pétri d’une main jusqu’à ce que l’embryon suivant soit à proximité de l’aiguille. Injectez au moins 300 embryons en utilisant la même procédure pour assurer une quantité suffisante d’éclosion. Couvrir le couvercle des boîtes de Petri après l’injection.Lorsque la couleur de surface des embryons s’est assombrie, mettez un régime artificiel dans la boîte de Pétri autour de la lame du microscope. Préparez des plaques de culture de 24 puits et remplissez chaque puits à 1/3 de sa capacité volumétrique avec un régime artificiel. Choisissez les larves d’éclosion à l’aide d’un petit pinceau et transférez-les dans la plaque de culture de 24 puits, de sorte qu’un puits ait une larve.Lorsque les larves atteignent le stade du troisième stade, transférez-les dans un nouvel éthrae ductile en verre. Transférez les adultes mâles G-zero nouvellement enfermés et les adultes femelles de type sauvage dans une cage en filet fraîche à un rapport égal. Fournissez-leur une solution de sucre à 10% déposée dans des boules de coton.Insectes d’élevage utilisant des méthodes de routine jusqu’à la nymphose de G-one. Utilisez une pince pour retirer soigneusement une patte arrière et placez chaque jambe dans un tube Lysing Matrix. Homogénéiser la patte arrière à l’aide d’un homogénéisateur tissulaire.Extraire l’ADN génomique de l’échantillon homogénéisé à l’aide d’un kit d’extraction d’ADNg commercial conformément aux instructions du fabricant. Amplifier le segment de gène à l’aide d’amorces de génotypage et des conditions de réaction pcriques du manuscrit textuel. Confirmez le génotype avec un service de séquençage de gènes.Mettez les individus G-one du même génotype dans une cage en filet, croisez eux-mêmes les descendants G-one et continuez à dépister en utilisant les mêmes méthodes. Le site cible du gène d’intérêt était situé dans son deuxième exon. Ce site a été hautement conservé et le fragment de bande cible de l’ARNg synthétisé a été confirmé par électrophorèse sur gel d’agarose.La détection mutante d’une seule cible d’ARNg a été réalisée en séquençant les produits pcR à partir d’unités parentales G-one. L’efficacité de l’utilisation de paires d’ARNg sans chevauchement sur différents exons a été démontrée car la grande délétion des mutants a été facilement distinguée des casseroles de type sauvage. Après avoir obtenu un certain nombre d’individus mutants, des expériences électrophysiologiques ou comportementales peuvent être effectuées pour clarifier la fonction des gènes et l’interaction entre différents gènes.
