アクチンアダプタータンパク質を可視化し、共局化するために、免疫蛍光技術が用いられ得る。残念ながら、光の回折障壁は、従来の蛍光顕微鏡の分解能を200〜350nm7に制限する。新規顕微鏡法は、例えば、刺激放出枯渇(STED)8、光活性化局在化顕微鏡(PALM)9、確率的光学再構成顕微鏡(STORMまたはdSTORM)、または個々の分子リターン(GSDIM)9、10、11の後に、約10nmまでの分解能を可能にする。しかし、これらの超分解能技術は、非常に高価な顕微鏡、十分な訓練を受けた人員を必要とし、したがって、多くの研究室では利用できません。

この拡張顕微鏡プロトコルにより、研究者は従来の顕微鏡でナノスケールの分解能を持つ糸球体タンパク質を可視化することができます。拡張顕微鏡の主な利点は、大規模な研究コミュニティにアクセスできることです。まず、ゲル化チャンバー用スペーサーを作ります。ダイヤモンドナイフを使用して、ナンバーワンとナンバー1.5ガラスカバーを5ミリメートルストライプにカットします。ガラススライドを染色チャンバーに入れ、2.5センチメートルの正方形を形成するように、1.5カバースリップストライプをガラススライドに置きます。二重蒸留水の液滴を正方形の角にピペットし、ガラスカバースリップストライプを互いに接着し、ガラススライドに貼り付けます。1.5個のカバースリップが安定して取り付けられるように、約20分待ちます。各番号1.5カバースリップストライプに水の滴をピペット。ナンバー1.5カバースリップに4つのナンバーワンのガラスカバースリップストライプを置きます。ゲル化チャンバ蓋の場合は、カバーガラスをパラフィンフィルムで包み、フィルムの折り目や汚れを避けます。アンカー処理のために、免疫染色細胞からPBSを取り出し、ガラスカバースリップに直接1ウェルあたり250マイクロリットルのアンカーバッファを追加します。プレートを室温で3時間インキュベートします。箱を使って、基板を暗闇の中に保管してください。インキュベーション後、アンカーバッファーを取り外し、カバースリップを1.5ミリリットルのPBSで1回洗います。アクチン繊維を染色するには、EXM適合ファロイジン溶液を解凍し、室温で45分間インキュベートします。一方、攪拌装置を用いて二重蒸留水にアクリレートナトリウムを溶解し、氷上でモノマー溶液を調製する。細胞からファロイジンを取り出し、室温で1.5ミリリットルのPBSで2回洗浄します。最後の洗浄後に井戸内にPBSの1.5ミリリットルを残します。鉗子とカニューレを使用して、カバーガラススリップを6つのウェルプレートから持ち上げ、ゲル化チャンバーに入れます。ゲル化液と渦液にAPSを短時間追加します。ピペット200マイクロリットルのゲル化液を試料に入れ、慎重にチャンバーを閉じ、ゲル内の気泡を避ける。染色チャンバーに水を加え、染色チャンバーを37°Cで少なくとも1時間インキュベートし、ゲルを重合させる。インキュベーターから染色チャンバーを取り出します。ゲル化チャンバーの蓋を開けるには、蓋とスペーサの間にカミソリの刃を導入し、蓋を慎重に取り外します。カミソリの刃でスペーサーを取り外し、余分なゲルをすべて切り落とします。ペイントブラシを使用して、ゲルの端を切り離します。スライドにゲルを入れ、ガラスをカバーしてPBSで満たされた皿に入れます。その後、取り外したカバーガラスをゲルから軽く振って取り外します。スライドをゲルの下に置き、スライドにゲルを取り付けます。スライドにゲルを付けて、カミソリの刃を使用してゲルを小さく分割します。ゲルの1枚をガラス底の6つのウェルプレートの井戸にそっと押し込み、ペイントブラシで広げます。少量のPBSでゲルを保湿するためにペイントブラシを使用してください。反転顕微鏡を使用して、拡大前画像の低い数値開口を持つ画像を撮影します。消化液を作るために消化バッファーで1ミリリットルあたり4単位にプロテイナーゼKを希釈.消化液を500マイクロリットルずつ各ウェルに加え、溶液内に浸漬します。蓋を閉じて室温で一晩消化させ、サンプルを暗闇の中に保ちます。ピペットで消化液を取り出し、廃棄します。二重蒸留水を1ミリリットル加え、浸漬したゲルを室温で10分間インキュベートします。水を取り出し、新鮮な二重蒸留水を1ミリリットル加えます。膨張の高原に達するまで10分ごとに水を交換し続け、ゲルが光学的に透明になる。ゲルから水を取り出し、すぐに顕微鏡検査を開始します。反転顕微鏡を使用して、低倍率で空気の目的を使用して、膨張前の状態にある細胞を見つけます。解像度を向上させるために、40 X と 63 X の目標に切り替えます。興味のある波長で興奮し、カメラを介して画像を撮ります。この EXM プロトコルは、最大 4 倍の拡張を可能にします。拡大因子を決定するには、細胞の展開前後の画像に不可欠です。不十分なアンカーと均質化は、細胞の歪みや破裂につながる可能性があります。破裂した細胞の代表的な例をここに示す。この方法は、ポドシンやネフリンなどのF-アクチンおよびアクチンアダプタータンパク質の共局在化を調べるのに使用できます。ポドシンは緑色、青のアクチン、赤のネフリンで描かれています。白い領域は、共局在を示します。蛍光シグナルの強度と豊富さは、EXMを成功させるための鍵です。ACXを介した蛍光色素の効率的なアンカーは、このプロトコルにとって非常に重要です。私たちの手で可溶化されたACXは3〜4ヶ月まで良いです。
