呼吸器ウイルスは、おそらく深刻な医療と経済的負担を引き起こす最も広範な病原体の1つです。新興流行株(H1N1、H5N1、H3N2、MERS、COVID-19など)の定期的な世界的な流行から、毎年のインフルエンザの季節性株まで、ウイルスは公衆衛生にとって絶え間ない脅威です。ワクチンは、これらの世界的な公衆衛生上の課題に対する対応の主な部分を占めていますが、これらの対策が単に応答性が高いことに注意するのは冷静です1,2。さらに、新しい感染株の出現とそのワクチンの開発の成功との間の遅れは避けられず3、ウイルスの拡散を抑制するために利用可能な措置が非常に限られている期間につながる。

インフルエンザに感染した上皮細胞は、先天性のT細胞を急速に活性化する因子を産生する12、13、14。この知見に基づいて、この非接触気液界面(ALI)共培養モデルは、感染初期の鼻上皮層とPBMCとの間の初期の化学的相互作用(感染上皮層によって放出される可溶性因子によって媒介される)を模倣することを目的としている。鼻上皮層(膜挿入物上で培養)とPBMC(下のチャンバー内)との間の物理的分離および上皮の完全性は、ウイルスによるPBMCの直接感染を防ぎ、PBMCに対する上皮由来可溶性因子の影響の詳細な研究を可能にする。したがって、同定された因子は、インフルエンザ感染から保護し得る適切な先天性T細胞集団を誘導する際のそれらの治療可能性についてさらに調査することができる。したがって、この論文は、上皮由来可溶性因子からの先天的T細胞活性化の研究のための共培養を確立する方法を詳述した。
