L’analyse de propagation cellulaire décrite permet le suivi continu des changements morphologiques(p. ex., taille et forme des cellules) et des mouvements des bords cellulaires(c.-à-d. vitesse de saillie et fréquence de rétractation), qui sont des caractéristiques manquantes dans la plupart des protocoles depropagation cellulaire 19,24. Les saillies lamellipodial sont des structures cytosquelettiques proéminentes formées à l’avant d’une cellule migrante. Dans la lamellipodia, la polymérisation de l’actine à l’aide du complexe Arp2/3 et des formins crée un maillage d’actine ram ramifère à croissance rapide qui pousse contre la membraneplasmatique 1,2. La force de poussée générée par le maillage actin propulse physiquement la cellule versl’avant 1,3,4,5. L’épuisement du complexe Arp2/3 ou la perturbation des voies de signalisation essentielles aux saillies lamellipodiales nuisent souvent à la migration cellulaire6, 7. Bien que la migration des cellules lamellipodia-déficientes ait égalementété rapportée 8,9,l’importance de la lamellipodia dans la migration cellulaire est évidente car l’épuisement de cette structure saillante perturbe la capacité de la cellule à se déplacer par des microenvironnements biologiquescomplexes 6,10.
Un obstacle majeur à la compréhension de la régulation de la lamellipodia dans les cellules migration est la variabilité naturelle de la cinétique de saillie lamellipodial, la taille et la forme11,12,13,14. En outre, des études récentes ont démontré que la lamellipodia présentent des comportements complexes saillants, y compris fluctuants, périodiques et accélérant les saillies14,15. Par rapport à la lamellipodie très variable des cellules migratives6,16, lamellipodie formée pendant la propagation cellulaire sont plusuniformes 12. Depuis l’activité saillante de la propagation et la migration des cellules est entraînée par des assemblages macromoléculaires identiques, qui comprennent un réseau d’actine ram ramée, faisceaux d’actomyosine contractile, et à base d’intégrine adhérencescellulaires matrice 17,18, cellules de propagation ont été largement utilisés comme un modèle pour étudier la régulation de la dynamique lamellipodia.
La propagation cellulaire est un processus mécanochimique dynamique où une cellule en suspension adhère d’abord à un substrat par des adhérences à base d’intégrine17,19,20, puis se propage en étendant les saillies à base d’actine21,22,23. Pendant la phase de propagation, la lamellipodia émanant du corps cellulaire dépasse isotropiquement et constamment avec peu ou pas de rétractation oud’étalon 12. Les protocoles de propagation cellulaire les plus couramment utilisés sont les analyses de points de terminaison, où les cellules de propagation sont fixées à divers moments aprèsle placage 19,24. Ces analyses, bien que rapides et simples, sont limitées dans leur puissance diagnostique pour détecter des changements dans les dispositifs dynamiques de lamellipodia. Pour déterminer les mécanismes moléculaires qui contrôlent la dynamique de la lamellipodia, le groupe sheetz a été le pionnier de l’utilisation de l’analyse quantitative des cellules vivantes de propagation et a découvert de nombreuses propriétés fondamentales des saillies de bord cellulaire11,12,22. Ces études ont démontré que l’essai de propagation des cellules vivantes est une technique robuste et puissante dans la boîte à outils d’un laboratoire de biologie cellulaire. Malgré cela, un protocole détaillé et un outil de calcul open-source pour un test de propagation des cellules vivantes ne sont actuellement pas disponibles pour la communauté de biologie cellulaire. À cette fin, notre protocole décrit les procédures d’imagerie des cellules de propagation en direct et fournit un outil automatisé d’analyse d’image. Pour valider cette méthode, nous avons utilisé l’inhibition arp2/3 comme traitement expérimental et avons montré que l’inhibition de la fonction du complexe Arp2/3 n’arrêtait pas la propagation cellulaire, mais causait une réduction significative de la vitesse de saillie cellulaire, ainsi que la stabilité des saillies du bord cellulaire, donnant lieu à des bords de cellules déchiquetés. Ces données démontrent que la combinaison de l’imagerie à cellules vivantes et de l’analyse automatisée de l’image est un outil utile pour analyser la dynamique des bords cellulaires et identifier les composants moléculaires qui régulent la lamellipodie.

La propagation cellulaire est un processus dynamique dans lequel une cellule suspendue dans le corps se fixe à un substrat et s’aplatit d’une forme arrondie à une forme mince et étalée. Après l’attachement cellule-substrat, la cellule forme une mince feuille de lamellipodie émanant du corps cellulaire. Dans la lamellipodie, les monomères globulaires d’actine (G-actin) polymérisent dans un maillage filamenteux dense d’actine (F-actin) qui pousse contre la membrane plasmatique, fournissant ainsi les forces mécaniques nécessaires à la propagation de la cellule. Notamment, les acteurs moléculaires qui contrôlent la polymérisation de l’actine dans la lamellipodie sont essentiels pour de nombreux autres processus cellulaires, tels que la migration cellulaire et l’endocytose. 
Depuis que les cellules qui se propagent forment une lamellipodie continue qui s’étend sur toute la périphérie cellulaire et se dilatent constamment vers l’extérieur, les analyses de propagation cellulaire sont devenues un outil efficace pour évaluer la cinétique des saillies lamellipodial. Bien que plusieurs implémentations techniques de l’analyse de propagation cellulaire aient été développées, il manque actuellement une description détaillée du flux de travail, qui comprendrait à la fois un protocole étape par étape et des outils informatiques pour l’analyse des données. Ici, nous décrivons les procédures expérimentales de l’analyse de propagation cellulaire et présentons un outil open-source pour l’analyse quantitative et impartiale de la dynamique du bord cellulaire lors de la propagation. Lorsqu’il est combiné avec des manipulations pharmacologiques et / ou des techniques de silencing des gènes, ce protocole est approprié à un écran à grande échelle des acteurs moléculaires régulant les saillies lamellipodial.

Le protocole ci-dessus décrit les procédures expérimentales pour l’imagerie des cellules vivantes des cellules de propagation et un outil de calcul pour l’analyse quantitative de la dynamique de propagation cellulaire. Le protocole décrit dans cette vidéo définira les procédures expérimentales requises pour l’imagerie grandeur nature des cellules de propagation et l’utilisation d’un outil informatique qui quantifie la dynamique de propagation des cellules d’une manière impartiale et automatisée. L’actif de propagation cellulaire permet le suivi continu du mouvement du bord cellulaire et des changements morphologiques lors de la propagation cellulaire, ce qui est une caractéristique qui manque dans la plupart des analyses de propagation cellulaire existantes. Après l’incubation, vous devrez d’abord ajouter 10 millilitres de médias de culture cellulaire dans un tube de centrifugeuse. Ensuite, ajoutez rapidement 10 millilitres de médias dans le plat afin d’étancher la trypsine.Pour diluer les cellules qui seront ensemencées sur le bordereau de couverture, pipette un millilitre du contenu du plat dans le tube de centrifugeuse. Du tube, pipette environ 500 à 1000 microlitres de cellules diluées dans le plat avec le bordereau de couverture. Assurez-vous que le glissement de couverture est à une confluence de 10% ou 50.000 cellules par millilitre et ajuster le volume de cellules diluées au besoin.Le but de ces cellules est d’établir un plan de concentration lors de l’acquisition d’images. Avec les cellules restantes dans le plat, passer un cinquième des cellules dans un petit plat par traitement. Ce seront les cellules qui seront analysées pour la dynamique de propagation.Placer les plats de passage et le couvercle glisser le plat dans un incubateur pendant huit à 24 heures. Pour tester l’importance de l’Arp2/3 pour la propagation cellulaire, ajoutez d’abord cinq millilitres de médias de culture cellulaire dans un contrôle et un tube de centrifugeuse de traitement. Ajouter ensuite 20 millilitres de dmem qui manque de phénol rouge dans chacun des deux plus grands tubes de centrifugeuse.Pipette soit le médicament CK-666 qui est un inhibiteur du complexe Arp2/3 ou le traitement contrôlé comme DMSO dans les petits et grands tubes. Pour commencer le traitement pharmacologique des cellules, enlever les plats de passage qui couvaient pendant la nuit. Aspirez les médias de culture cellulaire de tous les plats et lavez la vaisselle avec du PBS chaud.Prenez les petits tubes contenant soit CK-666 ou contrôlez les supports complétés et ajoutez le contenu du tube pertinent dans chacun des plats de passage. Étiquetez chacun des plats avec le traitement médicamenteux correct, puis placez les plats dans l’incubateur pendant une heure supplémentaire Après une heure d’incubation, aspirez le médicament complété par les médias de chacun des plats. Ensuite, ajoutez du PBS chaud à tous les plats afin d’enlever complètement tous les supports rouges de phénol restants.Ajouter 230 microlitres de trypsine EDTA et placer les plats dans un incubateur pendant une minute. Retirer les plats trypsinisés de l’incubateur et procéder à ajouter cinq millilitres de la drogue complétée dmem qui manque de phénol rouge dans un tube désigné comme tube B.Puis ajouter un autre cinq millilitres du même média dans le plat pertinent pour étancher la trypsine. Pipette les médias de haut en bas à plusieurs reprises afin de détacher toutes les cellules du plat.Transférer tout le contenu du plat dans un autre tube qui a déjà été désigné comme tube A.In afin de préparer une dilution cellulaire qui est appropriée pour l’imagerie, transférer un millilitre de cellules du tube A dans le tube B.Répétez toutes les étapes de dilution pour chaque traitement. Placez tous les tubes dans l’incubateur pendant 45 minutes pour permettre aux cellules de se remettre de la trypsinisation. Assurez-vous que toutes les parties d’une chambre magnétique cellulaire pouvant accueillir un glissement de couvercle de 22 par 22 millimètres ont été soigneusement nettoyées avant utilisation.Retirer le plat avec le couvercle de l’incubateur. Aspirez les supports de culture cellulaire et lavez le bordereau de couverture avec du PBS chaud. Retirer le bordereau de couverture à l’aide d’une paire de forceps et déposer délicatement le couvercle sur la plaque inférieure de la chambre magnétique.Ensuite, prenez le joint en silicone et placez-le sur le dessus de la feuille de couverture. Fixez le corps principal de la chambre magnétique sur la plaque inférieure. Ajouter un millilitre du dmem sans rouge phénol correspondant au traitement pertinent à la chambre magnétique.Prenez un tissu sans peluche et tamponnez soigneusement l’enceinte entre le corps principal et la plaque inférieure afin de vérifier les fuites. Pour compléter la chambre magnétique, abaissez le couvercle transparent sur le corps principal. Enfin essuyer le fond de la feuille de couverture avec un tissu pulvérisé avec de l’eau et un autre pulvérisé avec 70% d’éthanol.Préchauffer l’incubateur supérieur d’étape d’un microscope confocal à 37 degrés. Placez la chambre magnétique sur l’objectif. Avant d’acquérir une dynamique de propagation, concentrez-vous sur les cellules déjà polarisées du canal fluorescent vert.Cela garantit que les cellules de propagation seront au point une fois qu’elles se fixent à la feuille de couverture. Retirez le couvercle transparent de la chambre magnétique et pipette 500 microlitres du tube B qui a été retiré de l’incubateur. Remettre le couvercle transparent sur le dessus.Pour identifier les cellules idéales pour l’analyse de propagation cellulaire, recherchez des halos verts qui représentent des cellules qui n’ont pas encore été attachées au glissement de couverture ou qui en sont aux premiers stades de l’attachement. Acquérez des images et enregistrez les fichiers. Après avoir terminé l’acquisition d’images de diffusion cellulaire, commencez par exécuter un Python IDE compatible tel que Spyder.Pour l’analyse computationnelle de la dynamique de propagation cellulaire, localisez et ouvrez le fichier GUI de diffusion cellulaire fourni dans les paquets de scripts pertinents. Appuyez sur le bouton d’exécuteur qui ouvrira le panneau d’analyse GUI en arrière-plan. L’interface graphique abrite tous les paramètres nécessaires à l’analyse.Pour analyser la zone cellulaire et la circularité, entrez le fichier d’acquisition et les paramètres nécessaires dans l’onglet zone de propagation cellulaire. Des ajustements devront être effectués en fonction du type de cellule et des paramètres d’acquisition d’image. Après avoir appuyé sur exécuter, le script va créer une circularité cellulaire et zone versus parcelle de temps pour toutes les cellules de propagation.Pour les données kymographiques, appuyez sur le générateur de kymograph et l’onglet analyse. Cette analyse nécessite que les fichiers d’entrée soient recadrés piles d’images à cellule unique. En outre, les kymographes générés automatiquement fournissent une résolution temporelle essentielle à la compréhension de la dynamique des saillies.Les cellules de contrôle dépassent constamment avec peu ou pas de rétractations. En revanche, l’inhibition arp2/3 perturbe la stabilité des saillies comme l’indique l’augmentation des événements de rétractation tout au long de la propagation. Ces vidéos devraient vous fournir la compréhension de la façon de bien semer les cellules, effectuer des traitements pharmacologiques et préparer l’imagerie et les outils informatiques nécessaires à la quantification de la dynamique de propagation cellulaire.Ce protocole peut être combiné avec l’imagerie fluorescente de cytosquelette et les analyses de migration pour identifier les joueurs moléculaires régissant des saillies cellulaires. Live Cell Imaging of F-actin Dynamics via Fluorescent Speckle Microscopy (FSM)

In this protocol we describe the use of Fluorescent Speckle Microscopy (FSM) to capture high-resolution images of actin dynamics in PtK1 cells. A unique advantage of FSM is its ability to capture the movement and turnover kinetics (assembly/disassembly) of the F-actin network within living cells. This technique is particularly useful in deriving quantitative measurements of F-actin dynamics when paired with computer vision software (qFSM).  We describe  the selection, microinjection and visualization of fluorescent actin probes in living cells. Importantly, similar procedures are applicable to visualizing other macomolecular assemblies. FSM has been demonstrated for microtubules, intermediate filaments, and adhesion complexes.

Live-cell Imaging and Quantitative Analysis of Embryonic Epithelial Cells in Xenopus laevis

Embryonic epithelial cells serve as an ideal model to study morphogenesis where multi-cellular tissues undergo changes in their geometry, such as changes in cell surface area and cell height, and where cells undergo mitosis and migrate. Furthermore, epithelial cells can also regulate morphogenetic movements in adjacent tissues1. A traditional method to study epithelial cells and tissues involve chemical fixation and histological methods to determine cell morphology or localization of particular proteins of interest. These approaches continue to be useful and provide "snapshots" of cell shapes and tissue architecture, however, much remains to be understood about how cells acquire specific shapes, how various proteins move or localize to specific positions, and what paths cells follow toward their final differentiated fate. High resolution live imaging complements traditional methods and also allows more direct investigation into the dynamic cellular processes involved in the formation, maintenance, and morphogenesis of multicellular epithelial sheets. Here we demonstrate experimental methods from the isolation of animal cap tissues from Xenopus laevis embryos to confocal imaging of epithelial cells and simple measurement approaches that together can augment molecular and cellular studies of epithelial morphogenesis.

Determining Cell Number During Cell Culture using the Scepter Cell Counter

Counting cells is often a necessary but tedious step for in vitro cell culture. Consistent cell concentrations ensure experimental reproducibility and accuracy. Cell counts are important for monitoring cell health and proliferation rate, assessing immortalization or transformation, seeding cells for subsequent experiments, transfection or infection, and preparing for cell-based assays. It is important that cell counts be accurate, consistent, and fast, particularly for quantitative measurements of cellular responses.
Despite this need for speed and accuracy in cell counting, 71% of 400 researchers surveyed1 who count cells using a hemocytometer. While hemocytometry is inexpensive, it is laborious and subject to user bias and misuse, which results in inaccurate counts. Hemocytometers are made of special optical glass on which cell suspensions are loaded in specified volumes and counted under a microscope. Sources of errors in hemocytometry include: uneven cell distribution in the sample, too many or too few cells in the sample, subjective decisions as to whether a given cell falls within the defined counting area, contamination of the hemocytometer, user-to-user variation, and variation of hemocytometer filling rate2.
To alleviate the tedium associated with manual counting, 29% of researchers count cells using automated cell counting devices; these include vision-based counters, systems that detect cells using the Coulter principle, or flow cytometry1. For most researchers, the main barrier to using an automated system is the price associated with these large benchtop instruments1.
The Scepter cell counter is an automated handheld device that offers the automation and accuracy of Coulter counting at a relatively low cost. The system employs the Coulter principle of impedance-based particle detection3 in a miniaturized format using a combination of analog and digital hardware for sensing, signal processing, data storage, and graphical display. The disposable tip is engineered with a microfabricated, cell- sensing zone that enables discrimination by cell size and cell volume at sub-micron and sub-picoliter resolution. Enhanced with precision liquid-handling channels and electronics, the Scepter cell counter reports cell population statistics graphically displayed as a histogram.

The Scepter Cell Counter is a handheld automated device that can be used to count cells, monitor cell diameter and volume, and be used to check the health and quality of cellular populations from one culture to the next.

Quantitative Live Cell Fluorescence-microscopy Analysis of Fission Yeast

Several microscopy techniques are available today that can detect a specific protein within the cell. During the last decade live cell imaging using fluorochromes like Green Fluorescent Protein (GFP) directly attached to the protein of interest has become increasingly popular 1. Using GFP and similar fluorochromes the subcellular localisations and movements of proteins can be detected in a fluorescent microscope. Moreover, also the subnuclear localisation of a certain region of a chromosome can be studied using this technique. GFP is fused to the Lac Repressor protein (LacR) and ectopically expressed in the cell where tandem repeats of the lacO sequence has been inserted into the region of interest on the chromosome2. The LacR-GFP will bind to the lacO repeats and that area of the genome will be visible as a green dot in the fluorescence microscope. Yeast is especially suited for this type of manipulation since homologous recombination is very efficient and thereby enables targeted integration of the lacO repeats and engineered fusion proteins with GFP 3. Here we describe a quantitative method for live cell analysis of fission yeast. Additional protocols for live cell analysis of fission yeast can be found, for example on how to make a movie of the meiotic chromosomal behaviour 4. In this particular experiment we focus on subnuclear organisation and how it is affected during gene induction. We have labelled a gene cluster, named Chr1, by the introduction of lacO binding sites in the vicinity of the genes. The gene cluster is enriched for genes that are induced early during nitrogen starvation of fission yeast 5. In the strain the nuclear membrane (NM) is labelled by the attachment of mCherry to the NM protein Cut11 giving rise to a red fluorescent signal. The Spindle Pole body (SPB) compound Sid4 is fused to Red Fluorescent Protein (Sid4-mRFP) 6. In vegetatively growing yeast cells the centromeres are always attached to the SPB that is embedded in the NM 7. The SPB is identified as a large round structure in the NM. By imaging before and 20 minutes after depletion of the nitrogen source we can determine the distance between the gene cluster (GFP) and the NM/SPB. The mean or median distances before and after nitrogen depletion are compared and we can thus quantify whether or not there is a shift in subcellular localisation of the gene cluster after nitrogen depletion.

The fission yeast, Schizosaccharomyces pombe, is a good model system to study basic cellular processes. Here we describe a method to perform quantitative live cell analysis of fission yeast. In this particular experiment we focus on organisation of the genome within the cell nucleus, but the method can also be used to study cytosolic factors.

A Quantitative Fitness Analysis Workflow

Quantitative Fitness Analysis (QFA) is an experimental and computational workflow for comparing fitnesses of microbial cultures grown in parallel1,2,3,4. QFA can be applied to focused observations of single cultures but is most useful for genome-wide genetic interaction or drug screens investigating up to thousands of independent cultures. The central experimental method is the inoculation of independent, dilute liquid microbial cultures onto solid agar plates which are incubated and regularly photographed. Photographs from each time-point are analyzed, producing quantitative cell density estimates, which are used to construct growth curves, allowing quantitative fitness measures to be derived. Culture fitnesses can be compared to quantify and rank genetic interaction strengths or drug sensitivities. The effect on culture fitness of any treatments added into substrate agar (e.g. small molecules, antibiotics or nutrients) or applied to plates externally (e.g. UV irradiation, temperature) can be quantified by QFA.
The QFA workflow produces growth rate estimates analogous to those obtained by spectrophotometric measurement of parallel liquid cultures in 96-well or 200-well plate readers. Importantly, QFA has significantly higher throughput compared with such methods. QFA cultures grow on a solid agar surface and are therefore well aerated during growth without the need for stirring or shaking.
QFA throughput is not as high as that of some Synthetic Genetic Array (SGA) screening methods5,6. However, since QFA cultures are heavily diluted before being inoculated onto agar, QFA can capture more complete growth curves, including exponential and saturation phases3. For example, growth curve observations allow culture doubling times to be estimated directly with high precision, as discussed previously1.
Here we present a specific QFA protocol applied to thousands of S. cerevisiae cultures which are automatically handled by robots during inoculation, incubation and imaging. Any of these automated steps can be replaced by an equivalent, manual procedure, with an associated reduction in throughput, and we also present a lower throughput manual protocol. The same QFA software tools can be applied to images captured in either workflow.
We have extensive experience applying QFA to cultures of the budding yeast S. cerevisiae but we expect that QFA will prove equally useful for examining cultures of the fission yeast S. pombe and bacterial cultures.

Quantitative Fitness Analysis (QFA) is a complementary series of experimental and computational methods for estimating microbial culture fitnesses. QFA estimates the effect of genetic mutations, drugs or other applied treatments on microbe growth. Experiments scaling from focussed analysis of single cultures to thousands of parallel cultures can be designed.

Surface Spreading and Immunostaining of Yeast Chromosomes

The small size of nuclei of the budding yeast Saccharomyces cerevisiae limits the utility of light microscopy for analysis of the subnuclear distribution of chromatin-bound proteins. Surface spreading of yeast nuclei results in expansion of chromatin without loss of bound proteins. A method for surface spreading balances fixation of DNA bound proteins with detergent treatment. The method demonstrated is slightly modified from that described by Josef Loidl and Franz Klein1,2. The method has been used to characterize the localization of many chromatin-bound proteins at various stages of the mitotic cell cycle, but is especially useful for the study of meiotic chromosome structures such as meiotic recombinosomes and the synaptonemal complex. We also describe a modification that does not require use of Lipsol, a proprietary detergent, which was called for in the original procedure, but no longer commercially available. An immunostaining protocol that is compatible with the chromosome spreading method is also described.

A method for surface-spreading chromosomes from budding yeast is presented. This method is derived from a method previously described by Loidl and Klein. In addition, we demonstrate a procedure for immunostaining of spread chromosomes.

High-resolution Imaging and Analysis of Individual Astral Microtubule Dynamics in Budding Yeast

Dynamic microtubules are fundamental to many cellular processes, and accurate measurements of microtubule dynamics can provide insight into how cells regulate these processes and how genetic mutations impact regulation. The quantification of microtubule dynamics in metazoan models has a number of associated challenges, including a high microtubule density and limitations on genetic manipulations. In contrast, the budding yeast model offers advantages that overcome these challenges. This protocol describes a method to measure the dynamics of single microtubules in living yeast cells. Cells expressing fluorescently tagged tubulin are adhered to assembled slide chambers, allowing for stable time-lapse image acquisition. Live Cell Fluorescence Microscopy to Observe Essential Processes During Microbial Cell Growth

Core cellular processes such as DNA replication and segregation, protein synthesis, cell wall biosynthesis, and cell division rely on the function of proteins which are essential for bacterial survival. A series of target-specific dyes can be used as probes to better understand these processes. Staining with lipophilic dyes enables the observation of membrane structure, visualization of lipid microdomains, and detection of membrane blebs. Use of fluorescent-d-amino acids (FDAAs) to probe the sites of peptidoglycan biosynthesis can indicate potential defects in cell wall biogenesis or cell growth patterning. Finally, nucleic acid stains can indicate possible defects in DNA replication or chromosome segregation. Cyanine DNA stains label living cells and are suitable for time-lapse microscopy enabling real-time observations of nucleoid morphology during cell growth. Protocols for cell labeling can be applied to protein depletion mutants to identify defects in membrane structure, cell wall biogenesis, or chromosome segregation. Furthermore, time-lapse microscopy can be used to monitor morphological changes as an essential protein is removed and can provide additional insights into protein function. For example, the depletion of essential cell division proteins results in filamentation or branching, whereas the depletion of cell growth proteins may cause cells to become shorter or rounder. Here, protocols for cell growth, target-specific labeling, and time-lapse microscopy are provided for the bacterial plant pathogen Agrobacterium tumefaciens. Together, target-specific dyes and time-lapse microscopy enable characterization of essential processes in A. tumefaciens. Finally, the protocols provided can be readily modified to probe essential processes in other bacteria.

Lens-free Video Microscopy for the Dynamic and Quantitative Analysis of Adherent Cell Culture

Here, we demonstrate that lens-free video microscopy enables us to simultaneously capture the kinetics of thousands of cells directly inside the incubator and that it is possible to monitor and quantify single cells along several cell cycles. We describe the full protocol used to monitor and quantify a HeLa cell culture for 2.7 days. First, cell culture acquisition is performed with a lens-free video microscope, and then the data is analyzed following a four-step process: multi-wavelength holographic reconstruction, cell-tracking, cell segmentation and cell division detection algorithms. As a result, we show that it is possible to gather a dataset featuring more than 10,000 cell cycle tracks and more than 2 x 106 cell morphological measurements.

Micromanipulation Techniques Allowing Analysis of Morphogenetic Dynamics and Turnover of Cytoskeletal Regulators

Examining the spatiotemporal dynamics of proteins can reveal their functional importance in various contexts. In this article, it is discussed how fluorescent recovery after photobleaching (FRAP) and photoactivation techniques can be used to study the spatiotemporal dynamics of proteins in subcellular locations. We also show how these techniques enable straightforward determination of various parameters linked to actin cytoskeletal regulation and cell motility. Moreover, the microinjection of cells is additionally described as an alternative treatment (potentially preceding or complementing the aforementioned photomanipulation techniques) to trigger instantaneous effects of translocated proteins on cell morphology and function. Micromanipulation such as protein injection or local application of plasma membrane-permeable drugs or cytoskeletal inhibitors can serve as powerful tool to record immediate consequences of a given treatment on cell behavior at the single cell and subcellular level. This is exemplified here by immediate induction of lamellipodial cell edge protrusion by the injection of recombinant Rac1 protein, as established a quarter-century ago. In addition, we provide a protocol for determining the turnover of enhanced green fluorescent protein (EGFP)-VASP, an actin filament polymerase prominently accumulating at lamellipodial tips of B16-F1 cells, employing FRAP and including associated data analysis and curve fitting. We also present guidelines for estimating the rates of lamellipodial actin network polymerization, as exemplified by cells expressing EGFP-tagged &#946;-actin. Finally, instructions are given for how to investigate the rates of actin monomer mobility within the cell cytoplasm, followed by actin incorporation at sites of rapid filament assembly, such as the tips of protruding lamellipodia, using photoactivation approaches. None of these protocols is&#160;restricted to components or regulators of the actin cytoskeleton, but can easily be extended to explore in analogous fashion the spatiotemporal dynamics and function of proteins in various different subcellular structures or functional contexts.

We describe how micro- and photomanipulation techniques such as FRAP and photoactivation enable the determination of motility parameters and the spatiotemporal dynamics of proteins within migrating cells. Experimental readouts include subcellular dynamics and turnover of motility regulators or of the underlying actin cytoskeleton.
