Njure tubuloid kulturer kan etableras från friska njurvävnad och från urin möjliggör generering av representativa modeller för att studera njursjukdom och fysiologi. Detta protokoll möjliggör generering av friska njure tubuloider från vävnad och urin, den senare är ett icke-invasivt och patientvänligt sätt att erhålla material för generering av kulturer. Tubuloidkulturer kan användas för att studera njurfysiologi och för sjukdomsmodellering, såsom smittsamma, maligna och ärftliga njursjukdomar.
För att generera humana njure tubuloider från vävnad, placera njurvävnadsprovet i en milliliter avancerad DMEM plus plus medium i en 10 centimeter Petri-maträtt och använd skalpeller för att hacka vävnaden i bitar av ungefär en kubik millimeterstorlek. Använd tång och skalpeller för att överföra vävnadsbitarna till ett 15 milliliter rör och tvätta skålen med fem milliliter färskt medium. Använd en steril 10 milliliter pipett för att överföra mediet till röret.
Sedimentera vävnadsbitarna genom centrifugering och återanvänd pelleten i tre till fyra milliliter kollagenlösning. Inkubera vävnaderna på en horisontell skakapparat vid 37 grader Celsius och 250 varv per minut i 45 till 60 minuter med kraftig skakning var 15: e minut. När suspensionen är homogen och de flesta av vävnadsbitarna har smält, fyll röret med medium och blanda fem till tio gånger genom inversion.
Centrifugera suspensionen. Om pelleten är röd, tillsätt en milliliter lysbuffert för röda blodkroppar i röret för en fem minuters inkubation vid rumstemperatur. I slutet av inkubationen, tvätta röret med färskt medium.
Om vävnadsbitar fortfarande är synliga, återanvänd pelleten med fem milliliter färskt medium och filtrera suspensionen genom en 100 mikron cellsil i ett 50 milliliter rör. Överför den filtrerade fjädringen till ett nytt 15 milliliterrör för centrifugering och använd en P1000-pipett inställd på en känd volym för att mäta volymen på cellpelleten. Återanvänd försiktigt pelleten utan att skapa luftbubblor och inkubera cellerna i en minut på is.
I slutet av inkubationen, återanvänd pelleten i en 70 till 75% volym källarmembranextrakt och tallrik 15 mikroliter droppar av den resulterande suspensionen i en 37 graders uppvärmd multi-well cell kulturplatta. När alla droppar har pläterats, inkubera plattan upp och ner vid 37 grader Celsius i 15 till 20 minuter, tillsätt sedan lämplig volym på 37 grader Celsius värmt odlingsmedium och inspektera kulturerna under ett ljusfältmikroskop. För att generera humana njure tubuloider från urin, dela urinprovet lika mellan 50 milliliter rör och tvätta proverna två gånger med 10 till 20 milliliter färskt tvättmedium per tvätt.
Efter den andra tvätten, återanvänd pelletsen i sina kvarvarande supernatanter och slå samman proverna i ett enda 15 milliliterrör för en tredje centrifugation. Mät cellpelletvolymen som demonstrerad och återanvänd försiktigt pelleten utan att skapa luftbubblor. Efter en minuts inkubation i is, återanvänd pelleten i en 70 till 75% volym av källarmembranextrakt och plätera 15 mikroliter droppar av den resulterande suspensionen i 37 grader Celsius uppvärmda flerbrunnscelliga odlingsplattor som demonstreras, inkubera sedan dropparna upp och ner i 15 till 20 minuter innan du lägger till färskt medium och tittar på kulturerna med lätt mikroskopi.
Efter en till två veckor av båda typerna av kultur, använd en P1000-pipett för att störa tubuloiddroppar i varje brunn med spetsen för att skrapa brunnarnas bottnar för att samla de bifogade cellerna. Dra brunnsinnehållet i ett enda 15 milliliter rör som innehåller 10 milliliter avancerad DMEM plus plus medium och samla tubuloiderna genom centrifugering. Baserat på pelletens storlek, tillsätt trypsinersättningsmedel kompletterat med 10 mikromolar Y27632 för en fem minuters inkubation vid 37 grader Celsius.
I slutet av inkubationen, använd en 1 000 mikroliter pipettspets med en 10 mikroliter pipettspets placerad över spetsen för att pipett tubuloidfjädringen 20 till 30 gånger. Kontrollera under mikroskopet om det finns några intakta tubuloider kvar i röret. Det är mindre än 10% av intakta tubuloider närvarande, fyller röret med färsk avancerad DMEM plus plus plus och samlar tubuloiderna genom centrifugering så att pelletsvolymen kan mätas som visat.
Återanvänd pelleten i en 70 till 75% volym källarmembranextrakt och platt 15 mikroliterdroppar i en 37 grader Celsius uppvärmd multi-well cellkulturplatta. Efter en 15 till 20-minuters upp och ner inkubation vid 37 grader Celsius, lägg till förvärmt kulturmedium till dropparna och inspektera kulturen genom lätt mikroskopi. Tubuloider som genereras från njurvävnadsprover uppträder vanligtvis inom sju dagar efter odlingsanläggningen och måste passeras inom en till två veckor efter plätering.
I urin kulturer, kompakta tubuloid strukturer och vidhäftande celler visas cirka 14 till 21 dagar efter plätering och första passaging förekommer i allmänhet mellan tre och fyra veckor. Förekomsten av cystic epitelial tubuloider ökar med varje passage. Den framgångsrika generationen av mänskliga njure tubuloid kulturer kan bedömas av immunohistochemical färgning för markörer uttryckt i tubular njure epitel, såsom parade box gen 8 protein.
En korrekt tidpunkt för enzymatisk matsmältning av vävnad material och snabb bearbetning av urin prover är avgörande steg för det framgångsrika resultatet av dessa protokoll. Den höga effektiviteten av etablering av njure tubuloid kulturer kan bana väg för patientspecifika tester av droginducerad nefrotoxicitet, en vanlig biverkning av många kemoterapeutiska medel.
Mänskliga njure tubuloid kulturer representerar en värdefull in vitro modell för att studera njure fysiologi och sjukdom. Tubuloider kan etableras från njurvävnad (frisk och sjuk) samt urin, den senare representerar en lättåtkomlig och mindre invasiv källa till forskningsmaterial.
