Het bovenstaande protocol schetst de procedures voor het isoleren van recycling-endosomen uit gekweekte cellen door ultracentrifugatie. De betrouwbaarheid van deze methode is aangetoond door de laatste publicatie22, waaruit blijkt dat recycling-endosomen met succes worden geïsoleerd uit andere organellen (figuur 1), zoals het Golgi-apparaat en mitochondriën. Er moet aandacht worden besteed aan enkele kritische stappen voor het verkrijgen van een goed scheidingsresul...

Endosomale handel is een essentieel fysiologisch proces dat verschillende biologische gebeurtenissenimpliceert 1, bijvoorbeeld het transport van signaalreceptoren, ionkanalen en adhesiemoleculen. Eiwitten gelokaliseerd bij het plasmamembraan worden geïnternaliseerd door endocytose2. De geïnternaliseerde eiwitten worden vervolgens gesorteerd op het vroege endosoom3. Sommige eiwitten zijn gericht op lysosomen voor afbraak4. Een aanzienlijke hoeveelheid eiwitten wordt echter teruggevoerd naar het celoppervlak door snelle recycling en langzame recyclingprocessen. Bij snelle recycling verlaten eiwitten de vroege endosomen en keren ze direct terug naar het plasmamembraan. Omgekeerd worden eiwitten bij langzame recycling eerst gesorteerd naar het endocytische recyclingcompartiment en vervolgens teruggevoerd naar het plasmamembraan. Verschillende ladingeiwitten, bijvoorbeeld clathrin, retromeercomplex, retrievercomplex en Wiskott-Aldrich-syndroomeiwit en SCAR Homologue (WASH) -complex, nemen deel aan dergelijke membraanrecyclingprocessen4,5,6,7,8,9. De balans van de endocytose en recyclinggebeurtenis is cruciaal voor celoverleving en draagt bij aan verschillende cellulaire gebeurtenissen10, bijvoorbeeld celadhesie, celmigratie, celpolariteit en signaaltransductie.
ARF6, een kleine GTPase, is een gerapporteerde regulator van endocytische handel7,11,12. Van belang, verschillende onderzoeksgroepen hebben het belang van ARF6 in endocytische recycling13,14,15,16,17geïllustreerd. De studie heeft tot doel de relatie tussen ARF6-gemedieerde neurietuitgroei en endocytische recycling te onderzoeken. Het vorige rapport suggereert dat de activering van ARF6 stroomopwaarts is naar Rac1-activiteit door in te werken op ELMO1-dedicator van cytokinese 1 (DOCK180) complex18. Hoe ARF6 ELMO1-DOCK180 gemedieerde Rac1-signalering activeert, blijft echter onduidelijk. Dichtheidsgradiënt ultracentrifugatie werd gebruikt om de rol van ARF6-gemedieerde endocytische recycling in een dergelijk proces te onderzoeken. Door dat te gebruiken, werd de recycling endosoomhoudende fractie verkregen uit cellysaten19. De fractie werd onderworpen aan Western blotting voor analyse van het eiwitgehalte. De immunoblotresultaten toonden aan dat onder de aanwezigheid van FE65, een met de hersenen verrijkt adaptoreiwit, actieve ARF6 het niveau van ELMO1 in de recycling endosoombevattende fractie aanzienlijk verhoogde. Het volgende protocol omvat de procedures voor (1) transfectie van zoogdiercellen; (2) het voorbereiden van de monsters en de kolommen met dichtheidsgradiënt; en (3) het verkrijgen van de recyclingdosome-bevattende fractie.

density gradient ultracentrifugation for investigating endocytic recycling in mammalian cells

Endosomale handel is een essentieel cellulair proces dat een breed scala aan biologische gebeurtenissen reguleert. Eiwitten worden geïnternaliseerd uit het plasmamembraan en vervolgens getransporteerd naar de vroege endosomen. De geïnternaliseerde eiwitten kunnen worden overgebracht naar het lysosoom voor afbraak of worden gerecycled naar het plasmamembraan. Een robuuste endocytische recyclingroute is nodig om de verwijdering van membraanmaterialen van endocytose in evenwicht te brengen. Van verschillende eiwitten wordt gemeld dat ze de route reguleren, waaronder ADP-ribosylatiefactor 6 (ARF6). Dichtheidsgradiënt ultracentrifugatie is een klassieke methode voor celfractionering. Na de centrifugatie worden organellen aan hun isopreienoppervlak gesedimenteerd. De fracties worden verzameld en gebruikt voor andere downstream toepassingen. Hier wordt een protocol beschreven om een recycling endosoombevattende fractie te verkrijgen uit getransfecteerde zoogdiercellen met behulp van dichtheidsgradiënt ultracentrifugatie. De geïsoleerde fracties werden onderworpen aan standaard Western blotting voor het analyseren van hun eiwitgehalte. Door deze methode te gebruiken, identificeerden we dat de plasmamembraantargeting van engulfment en celmotiliteit 1 (ELMO1), een Ras-gerelateerd C3 botulinetoxinesubstraat 1 (Rac1) guanine nucleotide-uitwisselingsfactor, door ARF6-gemedieerde endocytische recycling is.

Na fractionering van de niet-getransfecteerde HEK293-cellen door middel van ultracentrifugatie van de dichtheidsgradiënt, werden 12 fracties verzameld vanaf de bovenkant van de gradiënt. De geoogste fracties werden verdund met de verdunningsbuffer in een verhouding van 1:1 en onderworpen aan een tweede ronde van centrifugatie. Dit protocol helpt bij het isoleren van de recycling-endosomen die fractie bevatten uit getransfecteerde zoogdiercellen om het onderzoek naar eiwithandel via endocytische handel te vergemakkelijken. Deze techniek is gemakkelijk te hanteren en toepasbaar op zowel weefsel- als celmonsters. De procedure wordt gedemonstreerd door Miss Zhai Yu Qi, een promovendus van Dr.Lao's Laboratory.Om te beginnen, plaat 2 keer 10 tot de 6 cellen in een 100 millimeter kweekschaal. Transfecteer de cellen de volgende dag met Lipofectamine volgens de instructies van de fabrikant. Om de cellen te oogsten, gooit u het kweekmedium 48 uur na transfectie weg en wast u de cellen tweemaal met ijskoude PBS.Voeg vervolgens een milliliter ijskoude PBS aangevuld met 0,5X proteaseremmer cocktail in 0,5X fosfataseremmer cocktail toe aan elk gerecht. Verzamel vervolgens met behulp van een celschraper de cellen en breng de celsuspensie over naar een centrifugebuis van 15 milliliter. de cellen door centrifugeren met behulp van een zwenkbakrotor.Gooi het supernatant weg en resuspend de celkorrel voorzichtig in vijf milliliter homogenisatiebuffer. Verzamel de cellen opnieuw door centrifugering en resuspend de celkorrel in één milliliter homogenisatiebuffer. Homogeniseer de cellen met een Dounce Homogenisator met 15 tot 20 slagen.Breng vervolgens het homogonaat over in een centrifugatiebuis van twee milliliter. Voeg 700 microliter homogenisatiebuffer toe aan het homogonaat. Draai vervolgens het verdunde homogonaat en verzamel 1,5 milliliter van het supernatant.Draai het verzamelde supernatant opnieuw, verzamel vervolgens 1,4 milliliter van het supernatant en label het als post-nucleair supernatant. Om de kolom met dichtheidsgradiënt voor te bereiden, brengt u 1,2 milliliter van het postnucleaire supernatant over naar een ultracentrifugebuis. Voeg vervolgens een milliliter 62% sucrose-oplossing toe aan het monster en meng goed door zachtjes te pipetteren.Voeg vervolgens voorzichtig 3,3 milliliter 35% sucrose-oplossing toe aan het monster, gevolgd door 2,2 milliliter 25% sucrose-oplossing bovenop de 35% sucrose-oplossing. Vul ten slotte de ultracentrifugebuis aan de bovenkant met homogenisatiebuffer. Centrifugeer de kolom met dichtheidsgradiënt en verzamel voorzichtig 12 fracties van elk 1 milliliter, te beginnen vanaf de bovenkant van de gradiënt.Verdun vervolgens alle fracties met één milliliter verdunningsbuffer en centrifugeer de verdunde monsters. Na het centrifugeren van het supernatant, aspirateer het en voeg 50 microliter 1X monsterbuffer toe om de fracties te oogsten. De recycling-endosoommarker Rab11 werd gedetecteerd in fractie zeven.Andere subcellulaire markers, waaronder beta COP, COX IV, GAPDH, EEA1, Rab 7 en Lamp1 werden ook onderzocht. Een positief EEA1-signaal werd ook gedetecteerd in fractie zeven. De gemeten bandintensiteiten van GAPDH, Cox IV en Rab11 in zowel Fractie zeven als het postnucleaire supernatant worden hier weergegeven.Na transecting HEC293-cellen met ELMO1, ELMO1 ARF6Q67L of ELMO1 ARF6Q67L FE65 werden geen veranderingen gevonden in ELMO1-distributie met ARF6Q67L en FE65-overexpressie. Het ELMO1-niveau in fractie zeven werd vergeleken tussen verschillende transfecties en bleek verhoogd te zijn na cotransfectie van ARF6Q67L. Een verdere toename werd waargenomen wanneer zowel ARF6Q67L als FE65 gelijktijdig werden uitgedrukt.Omgekeerd verminderde het uitschakelen van FE65 de ARF6-gemedieerde Elmo1-verrijking in fractie zeven. De verkregen fractie kan worden gebruikt voor latere analyse. Bijvoorbeeld western blotting om veranderingen in het eiwitniveau in de fractie te visualiseren.
