Ici, nous avons présenté un protocole pour l’implantation de fenêtres crâniennes chroniques pour l’imagerie in vivo d’astrocytes corticaux et de neurones chez des souris éveillées et retenues sur une cage domestique soulevée par air. Des exemples spécifiques ont été fournis de l’application de la fenêtre crânienne pour l’imagerie des astrocytes qui expriment les GECI et les structures synaptiques neuronales. La capacité d’effectuer une microscopie à fluorescence multiphotonique in vivo dans le cortex de souris est primordiale pour l’étude de la signalisation cellulaire et de la structure 1,2,3,4,5,6,7,8,9, de la pathologie de la maladie 10,11 et du développement cellulaire12,13 . Avec l’implantation de fenêtres crâniennes chroniques, l’imagerie longitudinale est possible, permettant une imagerie répétée des zones corticales pendant des jours, des semaines oudes mois 13,14 chez des animaux vivants. La microscopie multiphotonique est idéale pour l’imagerie in vivo et répétée en raison de l’amélioration du sondage en profondeur et de la réduction des photodommages associés au laser infrarouge utilisé. Cela permet d’étudier la dynamique moléculaire et cellulaire de cellules spécifiques dans diverses régions corticales.
La microscopie multiphotonique a été utilisée pour l’imagerie in vivo de cellules neuronales et gliales chez des souris 15,16,17,18,19,20. Diverses stratégies peuvent être mises en œuvre pour étiqueter des types de cellules et des domaines d’intérêt particuliers. Une approche courante consiste à stimuler l’expression de protéines fluorescentes génétiquement codées d’une manière spécifique à la cellule en utilisant le système de recombinaison Cre-Lox. Cela peut être effectué avec des souris génétiquement modifiées, par exemple en croisant une souris tdTomato « floxed » (Ai14) avec une souris exprimant la Cre-recombinase sous un promoteur d’intérêt21. Alternativement, le marquage spécifique aux cellules et au site peut être réalisé avec des injections virales. Ici, un virus codant pour la Cre recombinase sous un promoteur spécifique à la cellule et un virus codant pour un gène floxé d’intérêt sont injectés dans une région définie. Les types de cellules appropriés recevant les deux vecteurs viraux exprimeront alors le(s) gène(s) souhaité(s). Ces gènes peuvent être des marqueurs structurels, tels que tdTomato, pour visualiser les changements dans la morphologie cellulaire22 ou des indicateurs de calcium génétiquement codés (GECI), tels que GCaMP et / ou RCaMP, pour examiner la dynamiquedu calcium 23. Les méthodes de recombinaison génétique peuvent être appliquées individuellement ou en combinaison pour marquer un ou plusieurs types de cellules. Une troisième approche, ne nécessitant pas de souris transgéniques ou de constructions virales (qui ont une capacité d’emballage limitée), consiste à électroporation in utero des constructions d’ADN24. Selon le moment de l’électroporation, différents types de cellules peuvent être ciblés 25,26,27.
Lors de l’imagerie multiphotonique, les souris peuvent être imagées alors qu’elles sont éveillées ou anesthésiées. L’imagerie des souris éveillées peut être réalisée en sécurisant la souris via une plaque de tête attachée28. Cette approche est rendue moins stressante en permettant un mouvement relativement libre de l’animal à l’aide de méthodes, telles que des balles en polystyrène flottant librement et soutenues par air29, des tapis roulants flottantlibrement 1 ou un système de cage domestique soulevé par air où les souris sont fixées par une plaque de tête attachée et autorisées à se déplacer dans une chambre ouverte30. Pour chacune de ces conditions d’imagerie, il sera d’abord nécessaire d’habituer les souris à la configuration d’imagerie. Cet article décrit la procédure d’accoutumance et d’imagerie à l’aide d’un système de cage domestique soulevé par air.
Ce protocole décrit l’implantation d’une fenêtre crânienne chronique pour l’imagerie longitudinale in vivo dans le cortex. Ici, nous utiliserons des souris qui expriment conditionnellement GCaMP6f dans les astrocytes pour surveiller la dynamique de signalisation du calcium. En outre, cet article décrit la procédure pour les injections virales en utilisant tdTomato comme étiquette pour les neurones. Cela permet de déterminer les changements dans la structure synaptique neuronale et / ou la disponibilité en tant que marqueur structurel permettant l’imagerie répétée du même astrocyte. Tout au long du protocole, des étapes cruciales seront mises en évidence pour assurer la meilleure qualité possible des images obtenues à partir de la microscopie multiphotonique.

Pour bien comprendre la physiologie cellulaire des neurones et de la glie chez les animaux qui se comportent, il est nécessaire de visualiser leur morphologie et d’enregistrer leur activité in vivo chez les souris qui se comportent. Cet article décrit une méthode d’implantation d’une fenêtre crânienne chronique pour permettre l’imagerie longitudinale des cellules cérébrales chez des souris éveillées et retenues par la tête. En combinaison avec des stratégies génétiques et des injections virales, il est possible de marquer des cellules et des régions d’intérêt spécifiques avec des marqueurs structurels ou physiologiques. Ce protocole montre comment combiner des injections virales pour marquer les neurones à proximité des astrocytes exprimant GCaMP6 dans le cortex pour l’imagerie simultanée des deux cellules à travers une fenêtre crânienne. L’imagerie multiphotonique des mêmes cellules peut être effectuée pendant des jours, des semaines ou des mois chez des animaux éveillés et se comportant. Cette approche fournit aux chercheurs une méthode pour visualiser la dynamique cellulaire en temps réel et peut être appliquée pour répondre à un certain nombre de questions en neurosciences.

La qualité de la fenêtre crânienne peut être évaluée par la netteté des structures neuronales. Dans une bonne fenêtre, les épines dendritiques sont clairement visibles (Figure 1). Le protocole décrit comment effectuer des injections virales et implanter une fenêtre crânienne pour l’imagerie des cellules cérébrales chez des souris éveillées et retenues par la tête. L’avantage de cette méthode est que la structure et l’activité des neurones et des astrocytes peuvent être imagées à plusieurs reprises sur plusieurs séances. Cette approche peut être utilisée pour déterminer comment les cellules neurales sont modifiées dans des modèles de troubles neurodéveloppementaux ou neurodégénératifs, ou si la plasticité dépendante de l’expérience est altérée.Commencez par fixer une souris anesthésiée à un cadre stéréotaxique à l’aide de la pince à museau et des barres d’oreille. À l’aide d’outils chirurgicaux stériles, coupez et retirez la peau des sutures frontales antérieures au bregma à postérieurement au lambda. À l’aide d’une lame chirurgicale stérile en acier au carbone de taille 11, grattez doucement tout le tissu conjonctif du crâne.À l’aide d’une perceuse dentaire, percez progressivement l’os du crâne le long du contour de l’ouverture en cercles concentriques pour faciliter l’amincissement uniforme de l’os. Après chaque passage concentrique, ajoutez une goutte ou deux de solution saline à la zone forée et laissez la solution saline reposer pendant au moins 10 secondes avant de reprendre le forage. Appuyez doucement sur la zone centrale de l’os du crâne avec une pince fine pour vérifier que le crâne est correctement aminci et bouge.Assurez-vous que le système vasculaire sous-jacent où le forage a eu lieu est intact et que l’os n’est pas fissuré. Insérez soigneusement une lame pointue miniature à 15 degrés dans l’os aminci et coupez et utilisez la mousse de gel imbibée de solution saline pour arrêter tout saignement qui pourrait survenir. À l’aide de pinces, soulevez et retirez soigneusement l’os, en prenant soin de ne pas endommager la dure-mère.Pour l’injection, remplissez une pipette en verre biseauté stérile avec une pointe de 20 micromètres avec le mélange de virus. Ensuite, abaissez la pipette pour toucher la surface du cerveau et continuez à baisser pendant 200 à 300 micromètres supplémentaires pour les injections de couche 2/3. À l’aide d’un système de distribution de micro-injection intracellulaire, injectez de la pression dans le système 12 à 15 fois en deux minutes.Pour l’implantation de la fenêtre crânienne, placez le verre de couverture sur l’ouverture dans le crâne. Utilisez des pinces pour vous assurer que le verre est au ras de l’ouverture. Pour sceller les bords de la fenêtre en verre au crâne, appliquez un gel adhésif cyanoacrylate autour du périmètre de la surface du verre.Sur le gel adhésif, appliquez une couche de colle. Ajoutez ensuite une couche de liquide de ciment dentaire. Lorsque le ciment dentaire durcit, appliquez une fine couche de colle autour de l’ouverture centrale de la plaque de tête de type hélicoptère et placez la plaque de tête de type hélicoptère de la taille appropriée sur le verre de couverture.Laissez sécher la colle. Ajouter la poudre de ciment dentaire dans un tube de microfuge de 1,5 millilitre jusqu’à la marque de 0,1 millilitre. Mélangez sept à huit gouttes d’adhésif instantané à durcissement rapide dans la poudre et aspirez le mélange résultant dans une seringue d’un millilitre avec une aiguille de calibre 19 qui a été coupée pour créer une plus grande ouverture.Injecter le mélange de poudre à travers les trous latéraux de la barre d’hélicoptère jusqu’à ce qu’il s’infiltre de chaque côté. Appliquez le mélange adhésif de ciment dentaire sur le reste du crâne exposé pour fixer la plaque de tête au crâne. Enveloppez la souris dans un chiffon et fixez-la via sa plaque de tête au bras de fixation de la tête de la cage domestique transportée par avion, puis laissez la cage domestique exposée à la lumière.Après un temps d’accoutumance de 15 minutes, retirez la souris de la cage de la maison. En utilisant le mode grand champ du microscope, sélectionnez deux à trois positions de vascularisation facilement identifiable. Enregistrez les images des vaisseaux sanguins et enregistrez les coordonnées x et y qui apparaissent sur le contrôleur du moteur.Imagez les structures synaptiques des neurones et l’activité de GCaMP6f dans les astrocytes. Dans l’analyse représentative, la qualité de la fenêtre crânienne a été évaluée avec une imagerie répétée des dendrites et des astrocytes exprimant GCaMP6f au fil des jours. Dans une bonne fenêtre, les structures neuronales semblaient nettes avec des épines dendritiques clairement visibles.Deux nouvelles épines sont apparues le deuxième jour de l’imagerie. Une seule colonne vertébrale a persisté et était visible le cinquième jour. L’activité calcique a été étudiée dans les astrocytes exprimant GCaMP6f à différents moments.Un événement mondial englobant toute la cellule a été observé lors d’un combat de locomotion. Les deux étapes critiques de ce protocole sont que l’os doit être correctement aminci avant le retrait de l’os et le placement approprié du verre de couverture sur l’ouverture. L’imagerie peut être combinée à l’analyse comportementale, soit pendant l’imagerie, soit après le comportement, comme l’apprentissage d’une nouvelle habileté motrice, pour déterminer comment la structure et la fonction neuronales et astrocytaires sont modulées par l’apprentissage.
