Her har vi presentert en protokoll for implantasjon av kroniske kranialvinduer for in vivo-avbildning av kortikale astrocytter og nevroner i våken, hodebehersket mus på et luftløftet hjemmebur. Evnen til å utføre in vivo multifoton fluorescensmikroskopi i musenes cortex er avgjørende for studiet av cellulær signalering og struktur 1,2,3,4,5,6,7,8,9, sykdomspatologi 10,11 og cellulær utvikling12,13 . Ved implantasjon av kroniske kranialvinduer er langsgående avbildning mulig, noe som muliggjør gjentatt avbildning av kortikale områder i dager, uker eller måneder13,14 hos levende dyr. Multifotonmikroskopi er ideell for in vivo, gjentatt bildebehandling på grunn av forbedret dybdeprobing og redusert fotodamage forbundet med den infrarøde laseren som brukes. Dette gjør det mulig å studere molekylær og cellulær dynamikk av bestemte celler i ulike kortikale regioner.
Multifotonmikroskopi har blitt brukt til in vivo-avbildning av nevronale og glialceller hos mus 15,16,17,18,19,20. Ulike strategier kan implementeres for å merke bestemte celletyper og interesseområder. En vanlig tilnærming er å drive uttrykket av genetisk kodede fluorescerende proteiner på en cellespesifikk måte ved hjelp av Cre-Lox-rekombinasjonssystemet. Dette kan utføres med genmodifiserte mus, for eksempel ved å krysse tdTomato "floxed" mus (Ai14) med en mus som uttrykker Cre-recombinase under en promotor av interesse21. Alternativt kan celle- og stedsspesifikk merking oppnås med virale injeksjoner. Her injiseres en viruskoding Cre-rekombininase under en cellespesifikk promotor og et virus som koder et floxed gen av interesse, inn i et definert område. Passende celletyper som mottar begge virale vektorer vil da uttrykke de ønskede genene. For hver av disse bildeforholdene vil det først være nødvendig å habituate musene til bildeoppsettet. Dette dokumentet beskriver habituation og bildebehandling prosedyre ved hjelp av en luftløftet hjem bur system.
Denne protokollen beskriver implantasjonen av et kronisk kranialvindu for langsgående in vivo-avbildning i cortex. Her vil vi bruke mus som betinget uttrykker GCaMP6f i astrocytter for å overvåke kalsiumsignaldynamikk. Videre beskriver dette papiret prosedyren for virale injeksjoner ved hjelp av tdTomato som etikett for nevroner. Dette gjør det mulig å bestemme endringer i nevronal synaptisk struktur og/eller tilgjengeligheten som en strukturell markør som muliggjør gjentatt avbildning av samme astrocytt. Gjennom hele protokollen vil viktige trinn bli fremhevet for å sikre best mulig kvalitet på bilder hentet fra multifotonmikroskopi.

Protokollen beskriver hvordan man utfører virale injeksjoner og implanterer et kranialvindu for avbildning av hjerneceller i våkne, hodebeherskede mus. Fordelen med denne metoden er at struktur og aktivitet av nevroner og astrocytter kan avbildes gjentatte ganger over flere økter. Denne tilnærmingen kan brukes til å bestemme hvordan nevrale celler endres i modeller av nevrodevelopmentale eller nevrodegenerative lidelser, eller om erfaringsavhengig plastisitet er svekket.Begynn med å sikre en bedøvet mus til en stereotaxisk ramme ved hjelp av snuteklemmen og ørestengene. Bruk sterile kirurgiske verktøy, kutt og fjern huden fra frontal suturer fremre til bregma å bakre til lambda. Bruk et sterilt kirurgisk blad i karbonstål i størrelse 11 til å skrape forsiktig alt bindevevet fra skallen.Ved hjelp av en tannbor, bor gradvis skallebenet langs omrisset av åpningen i konsentriske sirkler for å lette jevn tynning av beinet. Etter hvert konsentriske pass legger du til en dråpe eller to saltvann i det borede området, og lar saltvannslinjen sitte i minst 10 sekunder før du fortsetter boringen. Skyv forsiktig på det sentrale området av skallebenet med fine tang for å kontrollere at skallen er tilstrekkelig tynnet og beveger seg.Påse at den underliggende vaskulaturen der boringen forekom er intakt, og at beinet ikke er sprukket. Sett forsiktig et miniatyr 15-graders spissblad inn i det tynne benet og kutt og bruk gelskummet gjennomvåt i saltvann for å stoppe eventuelle blødninger som kan oppstå. Bruk tang, løft forsiktig og fjern beinet, pass på at dura mater ikke skader.Til injeksjonen, fyll en steril skrå glasspipette med en 20-mikrometer spiss med virusblandingen. Senk deretter pipetten for å berøre overflaten av hjernen, og fortsett å senke i ytterligere 200 til 300 mikrometer for lag 2/3 injeksjoner. Når tannsement herdes, påfør et tynt lag lim rundt den sentrale åpningen av helikoptertypen hodeplate, og plasser helikoptertypehodeplaten av riktig størrelse over dekkglasset.La limet tørke. Tilsett tannsementpulver i et mikrofunksjonsrør på 1,5 milliliter opp til 0,1 millilitermerket. Bland syv til åtte dråper hurtigherdende øyeblikkelig lim inn i pulveret, og trekk den resulterende blandingen inn i en sprøyte med en 19 gauge nål som er kuttet for å skape en større åpning.Injiser pulverblandingen gjennom sidehullene på helikopterstangen til den siver fra hver side. Vikle musen i klut, og fest den via hodeplaten til hodefikseringsarmen til det luftløftede hjemmeburet, og la deretter hjemmeburet bli utsatt for lys.Etter en habituation tid på 15 minutter, fjern musen fra hjemmeburet. Bruk mikroskopets brede feltmodus til å velge to til tre posisjoner for lett identifiserbar vaskulatur. Lagre bildene av blodårene, og registrer x- og y-koordinatene som vises på motorkontrolleren.Bilde synaptiske strukturer av nevroner, og GCaMP6f aktivitet i astrocytter. I den representative analysen ble kvaliteten på kranialvinduet evaluert med gjentatt avbildning av dendritter og GCaMP6f-uttrykkende astrocytter over dager. I et godt vindu virket nevronstrukturene skarpe med tydelig synlige dendrittiske spines.To nye spines dukket opp på dag to av avbildning. Bare en ryggrad vedvarte og var synlig på dag fem. Kalsiumaktivitet ble studert i astrocytter som uttrykker GCaMP6f på forskjellige tidspunkter.En global hendelse som omfattet hele cellen ble sett under en bevegelseskamp. De to kritiske trinnene i denne protokollen er at beinet må tynnes tilstrekkelig før fjerning av beinet, og riktig plassering av dekkglasset over åpningen. Avbildning kan kombineres med atferdsanalyse, enten under avbildning eller post-atferd, for eksempel å lære en ny motorisk ferdighet, for å bestemme hvordan nevronal og astrocyttstruktur og funksjon moduleres ved læring.
