Denne protokol om differentiering og polarisering AF THP-1 monocytlignende celler inden for 14 dage giver en metode til at opnå makrofager med en særskilt M2-lignende fænotype på grund af lang behandling inkubation af celler med passende hvileperioder mellem trin.

Tumor-associerede makrofager (TAM) og deres rolle i kronisk inflammation, udbrud af kræft, og tumor udvikling er vigtige mål i nyere forskning1,2. Perifere blodmonocytter, der rekrutteres til vævsmikromiljøet af den udviklende tumor, differentieres til makrofager og kan polariseres i to hovedundertyper afmakrofager 3. Den klassisk aktiverede makrofag repræsenterer den primært proinflammatoriske M1-lignende fænotype, og den alternativt aktiverede M2-lignende undertype viser overvejende antiinflammatoriske egenskaber4. Makrofager kan skifte dynamisk mellem disse to vigtigste fænotyper afhængigt af deres cellulære metabolisme, med mellemliggende undertyper, der har både inflammatoriske og antiinflammatoriske egenskaber5. TAM repræsenterer en heterogen population af begge fænotyper. En tumorfremmende funktion og dårlig prognose i forskellige typer kræftformer er dog især forbundet med M2-lignendemakrofager 6,7,8.
De funktionelle profiler af makrofager og deres interaktion med andre celler i tumormikromiljøet er komplekse og udfordrende at fange i et konstant skiftende miljø under løbende tumorudvikling. Cellelinjer kan give en homogen cellepopulation med stabil levedygtighed i kulturen, hvilket kan lette processen med at demonstrere definerede cellulære og intercellulære mekanismer. Den monocyt-lignende THP-1 celle linje er en legitim model system for primære menneskelige monocytter9. Denne spontant udødeliggjorte cellelinje er opnået fra det perifere blod fra et etårigt spædbarn med akut monocytisk leukæmi9,10. Differentiering og polarisering af BP-1-celler er blevet rapporteret af flere undersøgelser og er blevet udført på flere forskellige måder11,12,13,14. Aktivering og dermed polarisering af makrofager i en M1-lignende fænotype efterfølges af en kompenserende antiinflammatorisk rebound-mekanisme, der fremmer en M2-lignende fænotype gennem cytokiner produceret af inflammatoriske makrofager, såsom interleukin 6 (IL-6) eller itaconate15,16. Dette kan tjene som en pause mekanisme til at dæmpe en overskridelse inflammatorisk respons efter celle aktivering17. Processen med at differentiere og polarisere monocytter og THP-1 monocyt-lignende celler i en antiinflammatorisk M2-lignende fænotype ledsages også af proinflammatoriske stimuli, der skal overvindes. Denne protokol demonstrerer en metode til at differentiere og polarisere THP-1 monocyt-lignende celler i en M2-lignende fænotype af makrofager inden for 14 dage.

macrophage differentiation and polarization into an m2-like phenotype using a human monocyte-like thp-1 leukemia cell line

Tumor-associerede makrofager (TAM) kan skifte deres udtryk og cytokin profil i henhold til eksterne stimuli. Denne bemærkelsesværdige plasticitet gør det muligt for TAM at tilpasse sig løbende ændringer inden for tumor mikromiljø. Makrofager kan have enten primært pro-inflammatoriske (M1-lignende) eller antiinflammatoriske (M2-lignende) attributter og kan løbende skifte mellem disse to hovedstater. M2-lignende makrofager inden for tumormiljøet er forbundet med kræft progression og dårlig prognose i flere typer af kræft. Mange forskellige metoder til at fremkalde differentiering og polarisering af THP-1 celler bruges til at undersøge cellulære og intercellulære mekanismer og virkningerne af TAM inden for mikromiljøet af tumorer. I øjeblikket er der ingen etableret model for M2-lignende makrofag polarisering ved hjælp af THP-1 celle linje, og resultaterne af udtryk og cytokin profiler af makrofager på grund af visse in vitro stimuli varierer mellem undersøgelser. Denne protokol tjener som detaljeret vejledning til at differentiere THP-1 monocyt-lignende celler i M0 makrofager og yderligere polarisere celler i en M2-lignende fænotype inden for 14 dage. Vi demonstrerer de morfologiske ændringer af THP-1 monocyt-lignende celler, differentierede makrofager og polariserede M2-lignende makrofager ved hjælp af lysmikroskopi. Denne model er grundlaget for cellelinjemodeller, der undersøger de antiinflammatoriske virkninger af TAM og deres interaktioner med andre cellepopulationer af tumormikromiljøet.

Denne protokol er en guide til at differentiere og polarisere THP-1 monocyt-lignende celler i M2-lignende makrofager inden for 14 dage. I øjeblikket er der ingen etableret model for antiinflammatorisk THP-1 polarisering.Denne protokol bruger passende hvileperioder og en langvarig polariseringsproces til at oprette en særskilt M2-lignende makrofagsundertype. Lad derefter cellerne hvile i 48 timer i inkubatoren.Gentag hele processen efter 48 timer og lad cellerne hvile i yderligere 48 timer i inkubatoren. Fjern vækstmediet og udskift det med en milliliter frisk vækstmedium. Lad derefter cellerne hvile i 48 timer i inkubatoren.Fjern det varme cellemedie fra hver brønd og udskift det med en milliliter blanding af iskold PBS uden calcium og magnesium og 5%FBS. Derefter skal cellepladen straks placeres på is i 45 minutter. Efter 45 minutter på is skrabes cellerne af ved hjælp af minicelleskrabere.Overfør forsigtigt de fritliggende makrofager til kold PBS og 5% FBS i et 15 milliliterrør, der holdes på is. Makrofagerne, der er afledt af polariseringsmetoden, viser udtrykket af CD14 samt CD11b-markører. M2-lignende makrofager er reproducerbar skabt, og er blevet karakteriseret ved hjælp af QR-TPCR, ELISA, og flow cytometri.Dette er grundlaget for at undersøge antiinflammatoriske mekanismer i kræftudvikling eller vævs remodeling.
