腫瘍関連マクロファージ(TAM)と慢性炎症におけるそれらの役割、癌の発症、および腫瘍の発達は、最近の研究1,2において重要な標的である。現像腫瘍の組織微小環境にリクルートされた末梢血単球はマクロファージに分化し、マクロファージ3の2つの主要なサブタイプに分極することができる。古典的に活性化されたマクロファージは、主に炎症促進性M1様表現型を表し、代わりに活性化されたM2様サブタイプは主に抗炎症特性4を示す。マクロファージは、細胞代謝に応じてこれら2つの主要なフェノタイプ間で動的に切り替えることができ、中間サブタイプは炎症性および抗炎症属性の両方を有する5。TAMは、両方の表現型の不均一な集団を表す。異なるタイプの癌における腫瘍促進機能と予後不良は、しかし、特にM2様マクロファージ6、7、8と関連している。

抗炎症腫瘍関連マクロファージは、癌の進行および予後不良に関連している。このプロトコルは、THP-1単球様細胞を14日以内にM2様マクロファージに分化し偏光するためのガイドです。現在、抗炎症性THP-1分極化のための確立されたモデルはありません。このプロトコルは、適切な休息期間と長期の偏光プロセスを使用して、異なるM2様マクロファージサブタイプを作成します。腫瘍の発達は、例えば大腸癌および組織リモデリングにおいて、抗炎症マクロファージによって制御される。その機能を調査する研究の中で、明確なM2様サブタイプの標準化された作成は、再現可能な所見を保証する。タイマーを 150 秒間設定します。液体窒素からTHP-1細胞株を含む冷凍バイアルを取り出し、きれいな水浴で37°Cで速やかにバイアルを解凍し、バイアルが水浴に入れるとすぐにタイマーを開始します。キャップを緩めて解凍プロセスに伴う圧力を解放し、チューブ開口部が水に接触しないようにして汚染を避けます。約4ミリメートルの大きさのアイスチップがバイアル内に残るまで細胞懸濁液を解凍し、すぐに次のステップに進みます。細胞懸濁液の液相を、9ミリリットルの温成長培地を含む15ミリリットルチューブに移します。その後、この温かい培地セル懸濁液の1ミリリットルをTHP-1バイアルに移し、残りのアイスチップを溶かすために15ミリリットルのチューブに戻し、バイアルを洗い流して細胞が残らないようにします。1ミリリットルのピペットで上下にピペットして、サスペンションを軽く混ぜます。サンプルの約10マイクロリットルを取り除き、トリパンブルーを使用して細胞の生存率をカウントします。透明な細胞質を有する細胞は生存可能であり、青色の細胞質は細胞死を示す。カウントしながら、摂氏37度で7分間、200倍Gで暖かい細胞サスペンションを回転させます。上清を完全に除去し、増殖培地中の細胞を再懸濁して、1ミリリットル当たり500,000細胞の細胞密度を達成する。懸濁液を軽く混ぜ、それぞれT75細胞培養フラスコに22ミリリットルを移す。フラスコを5%の二酸化炭素濃度で摂氏37度のインキュベーターに直立して保管し、3~4日ごとに成長培地を交換します。細胞懸濁液を準備し、1ミリリットル当たり300,000の密度で24ウェル細胞培養プレートに播種します。培地を軽く混ぜ、50ミリリットルのチューブに26ミリリットルのアリコートを用意します。24ウェルプレートの各ウェルに細胞含有培地の1ミリリットルを移す。転送の間に上下にピペットを使用して、メディアを穏やかに混ぜます。PMAの新しく調製された作業溶液を氷の上に保管し、井戸あたり100ナノグラムのPMAを追加します。72時間それ以上の処理をせずにインキュベーターにプレートをインキュベートします。72時間後、成長培地を取り出し、1ミリリットルの新鮮な成長培地に置き換え、ピペットチップでウェルの底部に触れないようにします。細胞をインキュベーターでさらに96時間休ませます。成長培地を取り除き、新鮮な成長培地の1ミリリットルに置き換えます.インターロイキン4と20ナノグラムのインターロイキン13ウェルあたり20ナノグラムを追加します。その後、細胞をインキュベーターで48時間休ませます。48時間後に全プロセスを繰り返し、インキュベーターでさらに48時間細胞を休ませます。成長培地を取り除き、新鮮な成長培地の1ミリリットルに置き換えます.その後、細胞をインキュベーターで48時間休ませます。各ウェルから温かい細胞媒体を取り出し、カルシウムとマグネシウムと5%FBSなしで氷冷PBSの1ミリリットルの混合物に置き換えます。その後、すぐに45分間氷の上にセルプレートを置きます。氷の上で45分後、ミニセルスクレーパーを使用して細胞を削り取ります。取り外したマクロファージを冷たいPBSに、氷の上に置いた15ミリリットルのチューブに5%FBSをそっと移します。偏光法に由来するマクロファージは、CD14の発現と同様にCD11bマーカーを示す。単球およびマクロファージマーカーCD14及びCD11bをそれぞれ70.9%および74.7%の細胞で発現した。細胞は、細胞の0.2%でM1様マーカーCD80の低表面レベルを示し、62.6%の細胞でM2様マーカーCD206の高い表面レベルを示した。THP-1細胞は、ミディアムではバイアルの底に沈む傾向があり、機械的ストレスを通じて活性化しやすいため、細胞の穏やかな混合と取り扱いが重要です。M2様マクロファージは再現的に作成され、QR-TPCR、ELISA、およびフローサイトメトリーを用いて特徴付けられています。これは、がんの発生や組織のリモデリングにおける抗炎症メカニズムを調査するための基礎です。
