Denne protokollen om differensiering og polarisering av THP-1 monocyttlignende celler innen 14 dager gir en metode for å oppnå makrofager med en distinkt M2-lignende fenotype på grunn av lang behandlingsinkubasjon av celler med tilstrekkelig hvileperioder mellom trinnene.
Enkelte trinn er avgjørende for denne protokollen. Doblingstiden for THP-1 monocytter er ca. 26 timer. Celler kan deles med en celletetthet på 9 x 105/ml og bør frøs med en tetthet på 3 x 105/ml ...

Tumorrelaterte makrofager (TAM) og deres rolle i kronisk betennelse, utbruddet av kreft og tumorutvikling er viktige mål i nyere forskning1,2. Perifere blodmonocytter som rekrutteres til vevsmikromiljøet til den utviklende svulsten, skiller seg ut i makrofager og kan polariseres til to hovedundertyper av makrofager3. Den klassisk aktiverte makrofagen representerer den hovedsakelig proinflammatoriske M1-lignende fenotypen, og den alternativt aktiverte M2-lignende undertypen viser overveiende antiinflammatoriske egenskaper4. Makrofager kan bytte dynamisk mellom disse to viktigste fenotypene avhengig av deres cellulære metabolisme, med mellomliggende undertyper som har både inflammatoriske og antiinflammatoriske attributter5. TAM representerer en heterogen populasjon av begge fenotypene. En tumorfremmende funksjon og dårlig prognose i ulike typer kreft er imidlertid spesielt forbundet med M2-lignende makrofager6,7,8.
De funksjonelle profilene til makrofager og deres interaksjon med andre celler i tumormikromiljøet er komplekse og utfordrende å fange i et kontinuerlig skiftende miljø under pågående tumorutvikling. Cellelinjer kan gi en homogen cellepopulasjon med stabil levedyktighet i kulturen, noe som kan lette prosessen med å demonstrere definerte cellulære og intercellulære mekanismer. Den monocyttlignende THP-1-cellelinjen er et legitimt modellsystem for primære menneskelige monocytter9. Denne spontant udødeliggjorte cellelinjen er hentet fra perifert blod fra et ett år gammelt spedbarn med akutt monocytisk leukemi9,10. Differensiering og polarisering av THP-1 celler har blitt rapportert av flere studier og har blitt utført på flere forskjellige måter11,12,13,14. Aktivering og derfor polarisering av makrofager i en M1-lignende fenotype etterfølges av en kompenserende antiinflammatorisk rebound-mekanisme, som fremmer en M2-lignende fenotype gjennom cytokiner produsert av inflammatoriske makrofager, som interleukin 6 (IL-6) eller itakonat15,16. Dette kan fungere som en pausemekanisme for å dempe en overskytende inflammatorisk respons etter celleaktivering17. Prosessen med å differensiere og polarisere monocytter og THP-1 monocyttlignende celler i en antiinflammatorisk M2-lignende fenotype er i seg selv også ledsaget av proinflammatoriske stimuli som må overvinnes. En inflammatorisk cytokinrespons kan skyldes mekanisk stress18, for eksempel endring av medier for å refeede cellene, eller legge til kjemiske forbindelser for å skille THP-1-celler, for eksempel phorbol 12-myristate 13-acetat (PMA), og indusere produksjon av tumornekrosefaktor α (TNFα), interleukin 1β (IL-1β) eller IL-66 19. Denne endrede cytokinuttrykksprofilen som svar på PMA kan påvirke og forhindre etterfølgende makrofagpolarisering20. Tilstrekkelige hvileperioder, som rapportert før etter PMA-behandling, tillater disse inflammatoriske responsene å redusere og lette cellepolarisering i en distinkt M2-lignende fenotype21.
Denne protokollen demonstrerer en metode for å differensiere og polarisere THP-1 monocyttlignende celler til en M2-lignende fenotype makrofager innen 14 dager.

macrophage differentiation and polarization into an m2-like phenotype using a human monocyte-like thp-1 leukemia cell line

Tumorrelaterte makrofager (TAM) kan bytte uttrykk og cytokinprofil i henhold til ytre stimuli. Denne bemerkelsesverdige plastisiteten gjør det mulig for TAM å tilpasse seg pågående endringer i tumormikromiljøet. Makrofager kan enten primært ha proinflammatoriske (M1-lignende) eller antiinflammatoriske (M2-lignende) attributter og kan kontinuerlig bytte mellom disse to hovedtilstandene. M2-lignende makrofager i tumormiljøet er forbundet med kreftprogresjon og dårlig prognose i flere typer kreft. Mange forskjellige metoder for å indusere differensiering og polarisering av THP-1-celler brukes til å undersøke cellulære og intercellulære mekanismer og effekten av TAM i mikromiljøet av svulster. For tiden er det ingen etablert modell for M2-lignende makrofag polarisering ved hjelp av THP-1 cellelinjen, og resultatene av uttrykk og cytokinprofiler av makrofager på grunn av visse in vitro stimuli varierer mellom studier. Denne protokollen fungerer som detaljert veiledning for å skille THP-1 monocyttlignende celler i M0 makrofager og å polarisere celler ytterligere til en M2-lignende fenotype innen 14 dager. Vi demonstrerer de morfologiske endringene i THP-1 monocyttlignende celler, differensierte makrofager og polariserte M2-lignende makrofager ved hjelp av lett mikroskopi. Denne modellen er grunnlaget for cellelinjemodeller som undersøker de antiinflammatoriske effektene av TAM og deres interaksjoner med andre cellepopulasjoner av tumormikromiljøet.

Antiinflammatorisk tumor assosiert makrofager er knyttet til progresjon og dårlig prognose i kreft. Denne protokollen er en veiledning for å differensiere og polarisere THP-1 monocyttlignende celler til M2-lignende makrofager innen 14 dager. For tiden er det ingen etablert modell for antiinflammatorisk THP-1 polarisering.Denne protokollen bruker tilstrekkelige hvileperioder og en langvarig polariseringsprosess for å skape en distinkt M2-lignende makrofagundertype. Tumorutvikling, for eksempel i tykktarmskreft og vevsoppussing, styres av antiinflammatoriske makrofager. Blant studier som undersøker deres funksjon, sikrer standardisert opprettelse av en distinkt M2-lignende undertype reproduserbare funn.Begynn med å stille inn en tidtaker i 150 sekunder. Fjern det frosne hetteglasset som inneholder THP-1-cellelinjen fra det flytende nitrogenet og tine hetteglasset umiddelbart ved 37 grader Celsius i et rent vannbad, og start timeren så snart hetteglasset settes i vannbadet. Etter 45 minutter på is, skrap av cellene ved hjelp av minicelleskrapere.Overfør forsiktig de frittliggende makrofagene til kald PBS og 5% FBS i et 15 milliliterrør holdt på is. Makrofagene som er avledet fra polariseringsmetoden, viser uttrykket for CD14, i tillegg til CD11b-markører. Monocytt- og makrofagmarkørene CD14 og CD11b ble uttrykt i henholdsvis 70,9 % og 74,7 %av cellene.Celler viste lave overflatenivåer av M1-lignende markør CD80 i 0,2% av cellene, og høye overflatenivåer av M2-lignende markør CD206 i 62,6% av cellene. Siden THP-1-celler i medium har en tendens til å synke til bunnen av et hetteglass og er enkle å aktivere gjennom mekanisk stress, er skånsom blanding og håndtering av cellene viktig. M2-lignende makrofager er reprodusert opprettet, og har blitt karakterisert ved hjelp av QR-TPCR, ELISA og strømningscytometri.Dette er grunnlaget for å undersøke antiinflammatoriske mekanismer i kreftutvikling eller vevsoppussing.
