導電性ポリマーは、界面を改善するために生体電子デバイスに広く使用されている有機材料です。従来のポリマーと同様に、導電性ポリマーは合成が容易であり、加工中に柔軟性がある1。導電性ポリマーは、化学的および電気化学的方法を使用して合成することができる。しかし、電気化学合成アプローチは特に有利である。これは主に、薄膜を形成する能力、同時ドーピングを可能にする能力、導電性ポリマー中の分子の捕捉、および最も重要なことに、合成プロセス1の単純さによる。加えて、導電性ポリマーは、均一で繊維状で、でこぼこしたナノ構造を形成し、電極表面に強固に接着し、電極2の活性表面積を増加させる。

有機溶媒で作られたPEDOTの薄層で金の微小電極表面を改質することにより、より高い表面積を得て、センサの感度を高めることができます。微小電極手順は、様々な媒体中の抗酸化物質の迅速な分析である。これは、飲料の監視から入院患者の状態の即時評価まで、さまざまな状況に適用できます。適切なポテンショスタットを使用して、目的の電気化学的手法としてサイクリックボルタンメトリーを実行します。ポテンショスタットとそれに接続されているコンピュータの電源を入れます。コンピュータと計測器間の通信をテストするには、ソフトウェアを起動し、セットアップメニューでハードウェアテストコマンドを選択します。ポテンショスタットからいくつかの音を聞いた後、ハードウェアテストの結果が別のウィンドウに表示されます。[OK] をクリックし、実験の実行を続行します。ハードウェアテストコマンドをクリックすると、リンク失敗エラーが表示されることがあります。接続とポートの設定を確認します。接続をテストした後、セットアップメニューをクリックし、テクニックを選択し、オープニングウィンドウからサイクリックボルタンメトリー方法を選択します。次に、セットアップメニューに戻り、パラメータをクリックして適切な実験パラメータを入力します。例えば、裸の金微小電極上の有機電解質中で0.1モルのEDOT電気重合を実行するには、初期電位を負0.3ボルト、最終電位を負0.3ボルト、高電位を1.2ボルト、セグメント数を8に設定し、スキャンレートを毎秒100ミリボルトに設定し、方向を正に設定します。適切なサイクリックボルタンメトリーパラメータを入力した後、ガラス電気化学セルに作用極、参照電極、白金線対極を含む3つの電極セットアップを準備します。これらの清潔で乾燥した電極をスタンドに取り付けられた電極ホルダーの穴に通し、ホルダーを電気化学セルの上に置き、電極をターゲット溶液またはサンプルに挿入します。電極表面に気泡がないことを確認します。気泡がある場合は、電極を取り外し、再び脱イオン水ですすぎ、ティッシュで撫で乾かします。次に、電極をスタンドホルダーと溶液に戻します。参照電極の周囲に気泡がある場合は、先端を軽く叩いてください。スキャン開始後に対極の周囲に気泡がある場合は、対極を清掃してください。サイクリックボルタンメトリースキャンでノイズが多くなった場合は、電極表面を清掃し、システムの接続、ワイヤ、およびクリップを確認します。参照、動作、および対極の 3 つのワイヤ接続がすべて正しく接続されていることを確認したら、[実行] をクリックして実験を開始します。金微小電極を前処理するには、円形および8字型の手の動きを当ててガラス研磨板の上に置いたアルミナ研磨パッド上のアルミナスラリーで30秒間研磨し、次いで微小電極を脱イオン水ですすいでください。無水エタノールの15ミリリットルと超音波処理の2分間を含むガラスの卑劣な中にそれを挿入します。次に、微小電極をエタノールと水ですすぎ、脱イオン水で再び4分間超音波処理して、電極表面から余分なアルミナを除去します。最後に、0.5モルの硫酸を負の0.4ボルトから正の1.6ボルトの電位の間の20セグメントの間、毎秒50ミリボルトのスキャン速度で循環させることによって、追加の不純物を除去します。電極を硫酸で洗浄するたびに、一貫した陽極電位および陰極電位での酸化金の形成と還元による2つの明確なピークがあることを確認します。有機溶液中で0.1モルEDOTを調製するために、まず調製した0.1モル過塩素酸リチウム溶液の1ミリリットルを電気化学セルに移す。次いで、マイクロピペットを用いて、10.68マイクロリットルのEDOTモノマーを電気化学セルに加える。裸の金の微小電極表面でEDOTを電気重合するには、すべての電極セットアップを溶液に挿入し、サイクリックボルタンメトリーを実行します。次いで走査型電子顕微鏡を用いて、この修飾電極の表面を特徴付ける。電極を脱イオン水ですすぎ、組織で乾燥させます。次に、このPEDOT修飾金微小電極をセンシング目的で使用するには、電極を0.1モルの過塩素酸ナトリウム溶液に浸漬し、サイクリックボルタンメトリースキャンを実行することによって、その表面を水溶液に順応させる。次いで、脱イオン水ですすぎた後、この有機的にPEDOT修飾および馴化マイクロ電極をリン酸緩衝液に浸漬し、サイクリックボルタンメトリーを実行してバックグラウンドスキャンを生成する。最後に、電極を緩衝液から取り出し、すすぎずに、直ちに尿酸またはミルク溶液に挿入して、サイクリックボルタンメトリースキャンを実行します。次に、アセトニトリル水溶液中で0.01モルのEDOTを調製するために、マイクロピペットを使用して、ガラスバイアル中のアセトニトリル1ミリリットルに10.68マイクロリットルのEDOTを加え、次いでバイアルに9ミリリットルの脱イオン水を加えて、10ミリリットルの0.01モルEDOT溶液を調製した。調製したEDOT溶液に110ミリグラムの過塩素酸リチウム粉末を加え、0.1モルの過塩素酸リチウム溶液を得て、穏やかに混合する。調製した溶液を電気化学セルに移す。アセトニトリル水溶液に電極を挿入し、サイクリックボルタンメトリーを実行して電極表面に0.01モルのEDOTを電気重合させ、次いで走査型電子顕微鏡によってこの修飾電極の表面を特徴付ける。有機溶液中で合成されたPEDOTセンサーを用いた通常の新鮮な牛乳中の尿酸含有量の分析は、0.35ボルトで28.4ナノアンペアの陽極ピーク電流をもたらし、これは82.7マイクロモルの濃度に相当する。0.65ボルトの通常の牛乳のサイクリックボルタンメトリースキャンにおける2番目の大きな酸化ピークは、システイン、トリプトファン、チロシンなどの電気活性アミノ酸を含む酸化可能な化合物に関連しています。キャラメルおよびホワイトチョコレートミルクサンプルについて得られたサイクリックボルタンメトリースキャンは、尿酸について0.36ボルトの明確なピークを示し、フレーバーミルクの成分の1つであるバニリン酸の存在に関連し得る0.5ボルトの追加のピークを示す。ベルギーのチョコレートミルクサンプルのサイクリックボルタンメトリーは、0.26ボルトのカテキン酸化ピークと0.22ボルトのカテキン還元ピークを示しています。カテキンピークの尾部に鋭いピークとして現れる0.3ボルトのピーク電流は、尿酸酸化によるものである。コロンビアのエスプレッソミルクサンプルのサイクリックボルタンメトリーは、コーヒー中の主要なフェノール系抗酸化物質、すなわちクロロゲン酸およびカフェ酸のために、それぞれ0.35ボルトおよび0.23ボルトで広い陽極および陰極ピーク電流を示す。電極の洗浄と摂動は、得られた電流信号に影響を与える可能性のあるこの実験の最も重要な部分です。抗酸化化合物のより詳細な分析が必要であり、LCMSなどのクロマトグラフィー方法に目を向けることができます。ただし、これは時間がかかり、すべてのサンプルに必要というわけではありません。
