このプロトコルは、大人のゼブラフィッシュにおけるスケーラブルな鈍い力外傷性脳損傷を誘発する、シンプルで迅速かつ再現可能な方法を提供する。この鈍い力外傷性脳損傷を受ける成体ゼブラフィッシュは、鈍い力TBIに苦しむヒトで観察された特性の多くを示す。この手順のデモンストレーションは、私の研究室の博士課程の学生であるジェームズ・ヘンティグ氏とケイリー・クローゲッシー氏です。ペトリ皿に粘土を詰めることから始めます。次に、指または鉗子の背面を使用して、追加のモデリング粘土を備えた上げプラットフォームを作成します。かみそりの刃を使用して、上げられたプラットフォームを縦に2つのほぼ等しい半分に分割します。それは成魚の長さを収容するように、チャネルに2つの半分を形成します。追加の粘土を使用して、頭を露出した魚の体の2/3を確保するために壁を構築します。損傷時に頭部の回転や反動を避けるために、壁に垂直に露出した頭部領域の小さい支持を形成する。チャネルの側面がドロップされた重みを妨げないようにします。ミニホールパンチを使用して、22ゲージのスチールフラッシュから3ミリメートルのスチールディスクを作成します。麻酔魚を無反応にして、側に体を固定するように、その後ろ側を上にしてチャネル内の粘土型に尾ピンチに応答しないように置きます。3ミリメートル22ゲージのスチールディスクをヘッドの上に置き、目的の衝撃点を中心にします。頭が片側に傾くのを避けるために、魚を垂直に整列させ、不均一な衝撃を引き起こす可能性があります。標準的なリングスタンドとアームクランプを使用してスチールまたはプラスチックチューブを固定し、まっすぐにして、チューブの底部がゼブラフィッシュの頭より1.5センチメートル上になるようにします。チューブを見下ろし、鋼板の上に位置合わせされていることを確認します。怪我の所望の重症度に基づいて適切なボールベアリングを選択します。所定の高さから鋼板にボールベアリングをチューブにドロップします。その後、負傷した魚を回復タンクに入れて監視します。ペトリ皿をモデリング粘土で満たし、解剖中に体を支えるために小さな空洞を作成します。裏側を上にして粘土の型に魚を置きます。2つの解剖ピンを配置し、1つは中線を通って体の途中、もう1つは頭の下部の約5ミリメートルの後ろに置きます。鈍い視神経を切断し、目を削除するために5デュモン鉗子のペアを使用してください。5つの鉗子の一方の端を右頭頂板の下に置きます。ロストラルの端に向かって動く意図的なはさみアクションを行い、右前頭プレートを取り外します。その後、魚を時計回りに90度回転させます。5鉗子の一端を左頭頂板の下に置き、同じシザーモーションを使用して左頭頂板と前頭板を取り除き、脳の後部全体を露出させます。5鉗子を使用して、嗅球が保存され、損傷しないように上顎をぶっきらぼうにトランセクトします。同じ鉗子を使用して、右のオペルクル、前耳、相互運用機能、およびサブパークルを取り除く。その後、カルバリウムの尾部端で筋肉をぶっきらぼうに切除して、脊髄を露出する。鉗子を使用して、脊髄をぶっきらぼうにトランセクトする。鉗子を脳の下に慎重に置き、脳を穏やかにカルバリウムから取り除きます。テキスト原稿の指示を使用して脳全体または関心のある領域を解剖し、脳を刺したり掻き取ったりしないように注意して、細かい鉗子を使って小さな重量ボートにすぐに脳を置きます。タール乾燥重量ボートに脳を移し、脳の濡れた重量を記録する。頭脳を向けて、後側を上に向けて計量ボートに平らに置きます。その後、脳と乾燥重量ボートを摂氏60度に設定したハイブリダイゼーションオーブンに8時間置きます。乾燥したら新しいタール付きの小さな計量ボートに脳を移すために、細かい鉗子を一緒につまみ、脳の腹側から始めて、上向きの動きですくう。濡れたスポンジの部分的な切開を行います。腹側を上にして開口部に一度に1匹の魚を置き、30ゲージの針を使用して約40マイクロリットルの10ミリモルEdUを魚の体に注入します。その後、システム水で満たされた保持タンクに魚を返します。前述のように脳を収集し、9つの部分の2ミリリットルの100%エタノールを含む9ミリリットルのガラスバイアルにグループとして配置し、37%ホルムアルデヒドを1つの部分にします。ロッカープラットフォーム上で摂氏4度で脳を固定します。クライオスタットチャックを使用して、ドライアイス上の所望の向きでTFMに脳を埋め込みます。血管損傷は、このモデルを介して成功した傷害を同定するための最も簡単で最も顕著な病理の1つであることがわかった。損傷時に使用される魚の株で変化する指標を識別する能力.野生型ABにおける血管損傷の同定は、色素沈着による軽度または中等度のTBIと損傷のない対照魚の区別が困難であった。傷害後、軽度のTBI魚は最小限の表面擦過傷を示し、中等度のTBI魚は限られた脳出血を示した。重度のTBI魚では、怪我の程度が明らかでした。対照的に,アルビノまたはキャスパー魚を使用する場合には血管損傷を容易に同定することができる。怪我による大脳の腫脹を浮腫を用いて評価した。対照的に、中等度のTBIと重度のTBIの両方が有意な浮腫、1 dpiおよび3 dpiを有していたが、中等度のTBIおよび重度のTBIの両方の流体含有量は、5dpiによって損傷のない制御に似たレベルに戻った。この鈍い力傷害は、神経軸にわたる堅牢な細胞増殖応答をもたらした。EdU標識の増加は、損傷していないコントロールと比較して前脳の心室および脳室下のゾーンで観察された。負傷した脳は、損傷していない魚の脳と比較して、骨周りグレイゾーン、光学テクタルローブ、および前視床下部の側面におけるEdU標識の増加を示した。重度のTBIに続いて、後脳の神経原性領域は、損傷していない脳と比較して細胞増殖の増加を示した。怪我の適切な誘導のために、魚が金型で固定され安定していることを確認し、チューブが中心外の衝撃を避けるためにまっすぐであることを確認するか、重量降下軌道を変更します。この手順は、繰り返し頭部外傷または傷害誘発回生研究に特に適用される回生モデルに適用される迅速かつ費用対効果の高い鈍い力傷害誘導方法を提供する。
