我々の研究では、SBEM法は、マウス海馬および組織性脳スライス4,5のサンプルにおける樹状脊椎およびPSDの構造的可塑性を調査するためにうまく採用されている。SBEMは、走査型電子顕微鏡室6、7、8、9の中にミニチュアの超ミクロトームを設置することに基づいています。サンプルブロックの上部が画像化され、その後、サンプルがウルトラミクロトームによって指定された深さで切断され、新しいブロック面が明らかになります。その結果、切断されたスライスが破片として失われている間、ブロック面の画像のみが残されます。SBEMが破壊的な技術と呼ばれる理由は、同じ場所を再びイメージすることはできません。しかし、破壊的なオンブロック法の利点は、データ品質とデータ分析3に大きな影響を与える可能性のある歪み問題やセクション損失に苦しんでいないということです。さらに、SBEMは、比較的大きな視野(>0.5mm×0.5mm)を高解像度3で画像化する可能性を与える。
SBEMを採用するには、画像の取得に使用される反射電子検出器により、サンプルを専用の非常にコントラストの高いプロトコルに従って調製する必要があります。ここでは、1980年代8,11年に開発された還元オスミウムチオカルボヒドラジド(rOTO)染色を用いて、Deerinck10(国立顕微鏡・イメージング研究センター(NCMIR)法)が開発した手順に基づいて、プロトコルに従ってサンプル調製を行う方法を示す。また、軽顕微鏡とSBEMによる免疫蛍光研究に同じ脳を用いることができる軽度の固定灌流を用いて、2段階固定法を導入する。
プロトコルでは、マウスの脳は主に軽度の固定剤で固定され、次いで半分に切断され、一方の半球は後固定され、免疫蛍光(IF)のために準備され、もう1つはEM研究のために(図1)である。

脳内の各ニューロンは、約7,000シナプスによって他の神経細胞と接続されています。哺乳類の脳内のシナプスは、主に樹状棘と呼ばれる膜の小さな突起に位置することを除いて。シナプスや樹状棘等の可塑性は、学習や記憶過程などの重要な脳機能を果たします。電子顕微鏡だけが樹状脊椎にポストナプティック入力があるかどうかを完全に洞察する必要があることに注意することが重要です。樹状脊椎イメージングに関する様々な技術が利用可能です。しかし、シリアルブロック顔走査電子顕微鏡は、高解像度で大量の組織を画像化するユニークな可能性を与えます。シリアルブロックベースの走査型電子顕微鏡技術では、重金属との強い対比を伴う特殊なサンプル調製プロトコルが必要です。灌流中の私の主な固定は、私たちが軽くした電子顕微鏡のために同じ脳を使用することができました。マウスを屠殺し、軽度の一次固定剤を浸透させた。脳は半分に切断され、1つの半球は免疫蛍光専用の固定剤、凍結保護剤で固定され、クライオスタットを使用してスライスされ、免疫蛍光試験のために処理された。他の半球が電子顕微鏡固定で後方固定された場所で、ビブラートメでスライスし、電子顕微鏡試験のために調製した。シリアルブロックベースの走査型電子顕微鏡研究のための脳スライスは対照的で、樹脂に平らに埋め込まれ、次に海馬のCA1領域をピンに取り付け、シリアルブロックベースの走査型電子顕微鏡で画像化した。黄色いボックスで強調表示されたプロトコルの一部は、ビデオで紹介されました。固定された組織を取り、100個のマイクロバイオティクススライスに切断し、それぞれ3分間冷たいリン酸緩衝液で5回洗浄します。4%オスミウム四酸化水素と3%カリウムフェロシアン化物の1対1の混合物を調製します。最終製品は茶色になります。この混合物にサンプルを浸します。その後、氷の上にサンプルを置きます。そして、この段階から、光からそれらを保護することが重要です。1時間穏やかな揺れでそれらをインキュベートします。その間、TCH溶液を準備する。10ミリリットルの二重蒸留水と0.1グラムのTCHを混ぜます。60°Cに設定したオーブンに1時間置きます。時々解決策を取り上げるためには重要です。準備ができたら、室温まで冷まします。次に、サンプルを2重蒸留水でそれぞれ3分間5回洗浄し、ろ過されたTCH溶液に浸します。スライスが黒く変わります。室温で20分間インキュベートします。サンプルを2倍蒸留水でそれぞれ3分間5回洗浄し、2%オスミウム四酸化2%を室温で30分間インキュベートします。再度、サンプルを2重蒸留水でそれぞれ3分間5回洗浄し、ろ過した1%またはアセテートアセテートアミルに入れます。一晩で摂氏4度でインキュベートします。翌日、D-アスパラギンスの準備から始めます。硝酸鉛をアスパラギン酸溶液と混合し、摂氏60度にあらかじめ温めます。摂氏60度に設定された水浴に混合物を置き、1つの水酸化ナトリウムモルでpHを5.5に調整します。鉛アスパルテでバイアルを閉じ、摂氏60度で30分間放置します。解決策は明確であるべきです。曇った場合は破棄する必要があり、新しいものを準備する必要があります。その間、ガス二重蒸留水でサンプルをそれぞれ3分間5回洗浄し、オーブンで摂氏60度で30分間保管します。サンプルを作りたての鉛アスパラギンス溶液に浸し、摂氏60度に設定したオーブンで20分間インキュベートします。エポキシ樹脂を準備します。材料の重量を量り、加速器DMP 30を追加する前に、少なくとも30分間樹脂をよく混ぜます。エタノールの段階的希釈でウイルスを準備します。その後、オーブンからサンプルを取り出し、それぞれ3分間5回二重蒸留水で再び洗います。その間、1対1の割合で100%エタノールと樹脂を混合し、50%の樹脂を得る。よく混ぜます。これに続いて、エタノールの各希釈でサンプルを5分間脱水する。サンプルは完全に乾燥してはならないことを覚えておいてください。サンプルを最初に50%樹脂に30分間浸潤し、次に100%樹脂で1時間浸潤し、次に再び100%のフレッシュで一晩浸潤する。翌日、サンプルを1時間新鮮な100%樹脂に入れ、フッ素樹脂埋め込みシートの間に埋め込みます。エタノールで脱脂した埋め込みシートを用意し、支持として使用されるガラススライドを脇に置きます。ガラススライドに埋め込みシートを置き、その上に樹脂を一滴置き、木製のスティックを使用して慎重にサンプルを転送します。2枚目で覆います。気泡を避けるようにしてください。組織は非常に壊れやすいので、穏やかにしてください。慎重に埋め込みシートピースの上に別のガラススライドを置き、少なくとも48時間摂氏70度でオーブンでサンプルを治します。層を分離し、カミソリの刃で埋め込まれたサンプルの一部をカットします。パラフィンに移します。これにより、静電気によるサンプルの損失の危険性を最小限に抑えます。エタノールで脱脂したアルミニウムピンを取ります。導電性エポキシをよく混ぜて、少量を使ってサンプルをピンに取り付けます。導電性エポキシを摂氏70度で10分間硬化させます。サンプルブロックの両側をダイヤモンドナイフでトリミングし、組織が露出するまでブロックの面を磨きます。導電性塗料を使用してピンにサンプルを接地し、それを治します。充電を最小限に抑えるために、サンプルは、金または金のパラジウムの薄い層でコーティングされた斑点を付ける必要があります。サンプルを用いたピンをシリアルブロックベースの走査型電子顕微鏡のチャンバーに入れます。サンプルをナイフに合わせて、チャンバーを閉じてパラメータを設定します。画像のスタックを収集します。記載した方法を用いて、マウス脳組織の画像が得られた。海馬1領域の大きな画像を撮影し、画像は地層放射体の中心に設定された。画像のスタックが取得され、樹状脊椎やシナプスなどの対象物がセグメント化されました。良質の組織形態は、はっきりと見える膜とシナプス小胞およびよく保存されたミトコンドリアで得られた。PSD 95抗体の免疫蛍光試験は、4%PFAを有する軽度の一次固定および伝統的な固定が同等の結果を与えるのを確認した。提示されたプロトコルは、シリアルブロックベースの走査電子顕微鏡、高品質の組織形態、および12の可視膜を使用して脳組織画像を取得し、10の正しいセグメンテーションと再構成を容易にします。私の主な固定化により、PSD 95免疫蛍光と電子顕微鏡イメージング樹状脊椎の解析に同じ脳を使用することが可能になりました。ここでは、PSE 9、500の芽を使用しています。ただし、もう一方も同様に使用できます。
