De herfstlegerworm (FAW), Spodoptera frugiperda, is inheems in tropische en subtropische gebieden van Amerika. Momenteel is dit een verwoestende insectenherbivoor in meer dan 100 landen wereldwijd1. FAW-larven voeden zich met meer dan 350 waardplanten, waaronder enkele belangrijke basisvoedselgewassen2. Het sterke migratievermogen van FAW-volwassenen draagt bij aan de recente snelle verspreiding van Amerika naar andere plaatsen1,2. Als gevolg hiervan bedreigt dit insect nu de voedselzekerheid internationaal. Het toepassen van nieuwe technologieën kan geavanceerde studies in FAW vergemakkelijken en nieuwe strategieën bieden om deze plaag te beheersen.
Insectenkiemlijntransformatie is gebruikt om de genfunctie te bestuderen en transgene insecten te genereren voor gebruik bij genetische controle3,4. Van de verschillende methoden die worden gebruikt om genetische transformatie bij insecten te bereiken, is de op piggyBac-elementen gebaseerde methode de meest gebruikte methode5. Vanwege de lage transpositiesnelheid is het echter nog steeds een uitdaging om transgenese uit te voeren bij niet-modelinsecten. Onlangs is een hyperactieve versie van piggyBac transposase (hyPBase) ontwikkeld6,7. Kiembaantransformatie werd onlangs bereikt in FAW8, het eerste rapport dat de hyPBase gebruikte bij lepidoptera-insecten. In dit rapport wordt gedetailleerde informatie beschreven over het gebruik van hyPBase mRNA bij het genereren van transgene FAW. De hier beschreven methode kan worden toegepast om de transformatie van andere lepidoptera-insecten te bereiken.

hyperactive piggybac transposase-mediated germline transformation in the fall armyworm, spodoptera frugiperda

Stabiele insertie van genetische lading in insectengenomen met behulp van transponeerbare elementen is een krachtig hulpmiddel voor functionele genomische studies en het ontwikkelen van genetische plaagbestrijdingsstrategieën. Het meest gebruikte transponeerbare element in insectentransformatie is piggyBacen piggyBac-gebaseerdekiembaantransformatie is met succes uitgevoerd in modelinsecten. Het is echter nog steeds een uitdaging om deze technologie toe te passen in niet-modelinsecten die landbouwongedierte bevatten. Dit artikel rapporteert over kiembaantransformatie van een wereldwijde landbouwplaag, de herfstlegerworm (FAW), Spodoptera frugiperda, met behulp van het hyperactieve varkenBac transposase (hyPBase).
In dit werk werd het hyPBase-mRNA geproduceerd en gebruikt in plaats van helpplasmide in embryonale micro-injecties. Deze verandering leidde tot de succesvolle generatie van transgene FAW. Verder worden ook de methoden voor het screenen van transgene dieren, pcr-gebaseerde snelle detectie van transgene insertie en thermische asymmetrische interlaced PCR (TAIL-PCR) -gebaseerde bepaling van de integratieplaats beschreven. Dit artikel presenteert dus een protocol om transgene FAW te produceren, wat piggyBac-gebaseerdetransgenese in FAW en andere lepidoptera-insecten zal vergemakkelijken.

Een construct voor de expressie van hyPBase-bevattende T7 promotor: polyhedrin-5'UTR: hyPBase: polyhedrin-3'UTR: poly (A) signaal werd gegenereerd (Figuur 1A) en versterkt als een ~2,2 kb PCR fragment om hyPBase mRNA in vitro te synthetiseren (Figuur 1B). Dit protocol toont de productie van transgene Fall Armyworm met behulp van piggyBac-gebaseerd transformatiesysteem. We hopen dat deze methoden ten goede zullen komen aan een betere transformatie, niet alleen in de Fall Armyworm, maar ook in lepidoptera-insecten. De hier beschreven methode maakt de kiembaantransformatie van lepidopteran plaaginsecten efficiënt.De procedure wordt gedemonstreerd door Dr. Xien Chen, een postdoctoraal fellow in mijn laboratorium. Begin met het plaatsen van 12 mannelijke poppen en 12 vrouwelijke poppen in een doos. Plaats een 10% sucrose-oplossing in de doos voor het voeden van volwassenen.Bedek de doos met een papieren handdoek. Stel op dag twee na eclosie de volwassenen 's nachts bloot aan intens licht. Breng op dag drie na de eclosie de volwassenen over naar een donkere plaats.Verzamel de embryo's binnen twee uur na overpositie. Voeg druppels water toe aan het stuk van de papieren handdoek met verse eieren die in een petrischaaltje op ijs worden bewaard. Breng de eieren voorzichtig over op een glazen schuif met een fijne borstel en lijn ze een voor een uit op een glazen dia.Houd de glazen glijden met de eieren uitgelijnd op ijs voordat ze micro-injectie. Injecteer een mengsel van de 200 nanogram per microliter piggyBac-gebaseerde EGFP-tot expressie brengende plasmide, 200 nanogram per microliter hyperactief varkenBac transposase mRNA en steriel gedestilleerd water in de eieren met behulp van de compensatiedruk van de geautomatiseerde micro-injector. Houd de geïnjecteerde eieren op 27 plus of min één graad Celsius, 60 plus of min 10% relatieve vochtigheid totdat ze uitkomen.Voed de uitgekomen larven op een kunstmatig dieet en breng elke larve over naar een klein kopje in het vroege wrongel- en sterstadium. Verzamel de poppen en plaats ongeveer 150 vrouwelijke en 150 mannelijke poppen in een kooi. Hang verschillende stukken keukenpapier van 30 centimeter bij 15 centimeter groot in de kooi om de eieren te verzamelen.Verzamel de papieren handdoeken met eieren dagelijks en bewaar de eieren in de juiste omstandigheden totdat ze uitkomen. Houd de pasgeboren larven vijf minuten op vier graden Celsius om ze te immobiliseren en breng ze vervolgens over op een ijskoude plaat. Screen de geïmmobiliseerde pasgeboren larven onder een fluorescentie stereomicroscoop.Selecteer de EGFP-positieve pasgeborenen, til ze op naar het popstadium, in ongeveer twee weken bij 27 plus of min één graad Celsius. Het hyperactieve piggyBac transposase mRNA dat in vitro werd gesynthetiseerd, werd gedetecteerd met behulp van een 1%Agarose-gel. De PDS-geïnjecteerde embryo's vertoonden geen EGFP-signaal, terwijl de EGFP-tot expressie brengende plasmide en hyperactieve piggyBac transposase mRNA-geïnjecteerde embryo's EGP-fluorescentie vertoonden, wat aangeeft dat de piggyBac-constructie succesvol was.De G1-larven verkregen uit eieren die het resultaat waren van een zelfkruising van G0-volwassenen vertoonden een sterk EGFP-signaal. Een fragment met de promotor en het EGFP-fragment werd met succes versterkt met behulp van het genomische DNA geïsoleerd uit de EGFP-positieve larven als sjabloon. Hetzelfde fragment werd echter niet gedetecteerd toen genomisch DNA werd geïsoleerd uit de larven van het wilde type als de sjabloon.De eieren voor micro-injectie moeten zo vers mogelijk zijn en moeten altijd op ijs worden gehouden voordat ze micro-injecties maken om hun ontwikkeling te vertragen. Deze techniek maakt de weg vrij voor onderzoekers om vragen te beantwoorden in de functionele genomica van plaaginsecten en genetische bestrijdingsmethoden voor deze wereldwijde plaag te ontwikkelen.
