La chenille légionnaire d’automne (FAW), Spodoptera frugiperda,est originaire des régions tropicales et subtropicales d’Amérique. Actuellement, il s’agit d’un insecte herbivore dévastateur dans plus de 100 pays à travers le monde1. Les larves de FAW se nourrissent de plus de 350 plantes hôtes, y compris d’importantes cultures vivrières de base2. La forte capacité de migration des adultes FAW contribue à sa récente propagation rapide des Amériques à d’autres endroits1,2. En conséquence, cet insecte menace maintenant la sécurité alimentaire à l’échelle internationale. L’application de nouvelles technologies peut faciliter les études avancées sur la FAW et fournir de nouvelles stratégies pour lutter contre ce ravageur.
La transformation de la lignée germinale des insectes a été utilisée pour étudier la fonction des gènes et générer des insectes transgéniques à utiliser dans le contrôle génétique3,4. Parmi les différentes méthodes utilisées pour réaliser la transformation génétique chez les insectes, la méthode à base d’éléments piggyBac est la méthode la plus utilisée5. Cependant, en raison du faible taux de transposition, il est encore difficile de procéder à la transgénèse chez les insectes non modèles. Récemment, une version hyperactive de la transposase piggyBac (hyPBase) a été développée6,7. La transformation de la lignée germinale a été réalisée dans FAW récemment8, qui est le premier rapport qui a utilisé l’hyPBase chez les insectes lépidoptères. Dans ce rapport, des informations détaillées sur l’utilisation de l’ARNm hyPBase dans la génération de FAW transgénique sont décrites. La méthode décrite ici pourrait être appliquée pour réaliser la transformation d’autres insectes lépidoptères.

L’insertion stable de cargaisons génétiques dans les génomes d’insectes à l’aide d’éléments transposables est un outil puissant pour les études génomiques fonctionnelles et l’élaboration de stratégies de lutte génétique contre les ravageurs. L’élément transposable le plus utilisé dans la transformation des insectes est piggyBac, et la transformation germinale à base de piggyBaca été menée avec succès chez des insectes modèles. Cependant, il est toujours difficile d’utiliser cette technologie dans des insectes non modèles qui incluent des ravageurs agricoles. Cet article rend compte de la transformation germinale d’un ravageur agricole mondial, la légionnaire d’automne (FAW), Spodoptera frugiperda,en utilisant la transposase hyperactive piggyBac (hyPBase).
Dans ce travail, l’ARNm hyPBase a été produit et utilisé à la place du plasmide auxiliaire dans les microinjections embryonnaires. Ce changement a conduit à la génération réussie de FAW transgénique. En outre, les méthodes de dépistage des animaux transgéniques, la détection rapide de l’insertion de transgènes par PCR et la détermination du site d’intégration par PCR entrelacée asymétrique thermique (TAIL-PCR) sont également décrites. Ainsi, cet article présente un protocole pour produire du FAW transgénique, qui facilitera la transgénèse à base de piggyBacchez le FAW et d’autres insectes lépidoptères.

Ce protocole montre la production de légionnaire d’automne transgénique à l’aide d’un système de transformation basé sur piggyBac. Nous espérons que ces méthodes bénéficieront à l’amélioration de la transformation, non seulement chez la chenille légionnaire d’automne, mais aussi chez les insectes lépidoptères. La méthode décrite ici rend efficace la transformation germinale des insectes nuisibles lépidoptères.La démonstration de la procédure sera faite par le Dr Xien Chen, boursier postdoctoral dans mon laboratoire. Commencez par placer 12 nymphes mâles et 12 nymphes femelles dans une boîte. Placez une solution de saccharose à 10% dans la boîte pour nourrir les adultes.Couvrez la boîte avec une serviette en papier. Le deuxième jour après l’éclosion, exposez les adultes à une lumière intense pendant la nuit. Le troisième jour après l’éclosion, transférez les adultes dans un endroit sombre.Prélever les embryons dans les deux heures suivant la surposition. Ajoutez des gouttes d’eau au morceau de l’essuie-tout avec des œufs frais conservés dans une boîte de Pétri sur de la glace. Transférez doucement les œufs sur une lame de verre avec une brosse fine et alignez-les un par un sur une lame en verre.Gardez les lames de verre avec les œufs alignés sur la glace avant la microinjection. Injecter un mélange de 200 nanogrammes par microlitre de plasmide exprimant l’EGFP à base de piggyBac, de 200 nanogrammes par microlitre d’ARNm hyperactif de la transposase piggyBac et d’eau distillée stérile dans les œufs en utilisant la pression de compensation du micro-injecteur automatisé. Conservez les œufs injectés à 27 plus ou moins un degré Celsius, 60 plus ou moins 10% d’humidité relative jusqu’à l’éclosion.Nourrissez les larves écloses avec un régime artificiel et transférez chaque larve dans une petite tasse au stade précoce du caillé et de l’étoile. Recueillez les nymphes et placez environ 150 nymphes femelles et 150 nymphes mâles dans une cage. Accrochez plusieurs morceaux d’essuie-tout de 30 centimètres sur 15 centimètres dans la cage pour ramasser les œufs.Ramassez les serviettes en papier avec les œufs tous les jours et conservez les œufs dans des conditions appropriées jusqu’à l’éclosion. Gardez les larves de nouveau-nés à quatre degrés Celsius pendant cinq minutes pour les immobiliser, puis transférez-les sur une plaque glacée. Dépister les larves de nouveau-nés immobilisés au stéréomicroscope à fluorescence.Sélectionnez les nouveau-nés EGFP positifs, élevez-les au stade nymphal, en environ deux semaines à 27 plus ou moins un degré Celsius. L’ARNm hyperactif de la transposase piggyBac synthétisé in vitro a été détecté à l’aide d’un gel d’agarose à 1 %. Les embryons injectés par PDS ne montraient pas de signal EGFP, tandis que les embryons injectés de plasmide exprimant l’EGFP et d’ARNm hyperactif de transposase piggyBac présentaient une fluorescence EGP, indiquant que la construction de piggyBac était réussie.Les larves de G1 obtenues à partir d’œufs résultant d’un auto-croisement d’adultes G0 ont montré un fort signal EGFP. Un fragment contenant le promoteur et le fragment EGFP a été amplifié avec succès en utilisant l’ADN génomique isolé des larves EGFP positives comme modèle. Cependant, le même fragment n’a pas été détecté lorsque l’ADN génomique a été isolé des larves de type sauvage comme modèle.Les œufs pour la microinjection doivent être aussi frais que possible et doivent toujours être conservés sur de la glace avant la microinjection pour ralentir leur développement. Cette technique ouvre la voie aux chercheurs pour répondre à des questions en génomique fonctionnelle des insectes nuisibles et développer des méthodes de contrôle génétique pour ce ravageur mondial.
