このプロトコルは、ピギーバクトベースの変換システムを用いたトランスジェニック秋ミミミズの生産を示す。私たちは、これらの方法が秋のミミズだけでなく、鱗翅目昆虫においても、変革の改善に利益をもたらすことを願っています。ここで説明する方法は、鱗翅目害虫の生殖系列の形質転換を効率的にする。この手順のデモンストレーションは、私の研究室の博士研究員であるXien Chen博士です。まず、12匹のオスの子犬と12匹の雌の子犬を箱に入れることから始めます。大人に餌を与える箱の中に10%スクロース溶液を入れます。箱をペーパータオルで覆います。2日目のポストエローションでは、一晩で大人を強烈な光にさらす。3日目のポストエローションでは、大人を暗い場所に移します。位置を越えてから2時間以内に胚を収集します。紙タオルに水滴を加え、新鮮な卵を氷の上のペトリ皿に入れておきます。細かいブラシで卵をグラススライドにそっと移し、グラススライドの上に一つずつ揃えます。マイクロインジェクションの前に氷の上に整列した卵でガラススライドを保ちます。プラスミドを発現するマイクロリットル当たり200ナノグラムのマイクロリットルピギーバベースのEGFP、マイクロリットル多動ピギーバツトランスポザーゼmRNAあたり200ナノグラム、および無菌蒸留水を自動マイクロインジェクターの補償圧力を使用して卵に注入する。注入した卵を27プラスマイナス1度、60プラスマイナス10%の相対湿度で孵化するまで保管します。人工的な食事で孵化した幼虫を養い、早いカードと星の段階で各幼虫を1つの小さなカップに移します。子犬を収集し、ケージに約150の女性と150の男性の子犬を配置します。卵を集めるためにケージに30センチメートル×15センチメートルの大きさのペーパータオルを数枚掛けます。毎日卵とペーパータオルを収集し、孵化するまで適切な状態で卵を保ちます。新生児の幼虫を5分間摂氏4度に保ち、氷の冷たいプレートに移します。蛍光ステレオ顕微鏡で固定化された新生児幼虫をスクリーニングします。EGFP陽性の新生児を選択し、27プラスマイナス1度の摂氏で約2週間で、プパルステージに上げます。インビトロで合成した多活性ピギーババトランスポザーゼmRNAを1%アガロースゲルを用いて検出した。PDS注入胚はEGFPシグナルを示さなかったのに対し、EGFP発現プラスミドおよび多動性ピグバ菌トランスポザーゼmRNA注入胚はEGP蛍光を示し、ピギーバクの構築が成功したことを示した。G0成人の自己交差から生じた卵から得られたG1幼虫は強いEGFPシグナルを示した。プロモーターとEGFPフラグメントを含む断片を、EGFP陽性幼虫から分離したゲノムDNAを鋳型としてうまく増幅した。しかし、ゲノムDNAが鋳型の野生型幼虫から単離された場合、同じ断片は検出されなかった。マイクロインジェクション用の卵は可能な限り新鮮でなければならず、マイクロインジェクションの前に常に氷の上に保管して開発を遅らせる必要があります。この技術は、研究者が害虫の機能性ゲノミクスの質問に答え、この世界的な害虫の遺伝的制御方法を開発する道を開きます。
