尽管这些定义明确和关键的作用，为宏脊柱侧体在维持免疫力和平衡，与其他更常见的研究模式的内分泌，很少的宏观皮诺索的内部（发光）工作是已知的。开发标准化的协议和工具来研究巨脊质素的发光生物化学，不仅有助于我们更好地了解它们独特的生物学，而且将提供见解，可用于新的治疗策略，包括药物输送20。这种方法手稿将侧重于最近开发的工具，在单一细胞层解剖巨细胞素的发光生物化学的各个方面。
荧光素可用于测量细胞器的特定生物化学，如果 i） 它们优先划分到兴趣隔间和/或 ii） 它们根据兴趣参数进行光谱变化。例如，在 pH 的情况下，荧光弱碱基，如青紫橙、新月紫罗兰和莱索跟踪染料优先积聚在酸性有机体中。因此，它们的相对强度是标记的细胞器是酸性的粗略迹象。其他pH响应荧光素，如荧光素、pHrodo和环丙烯，在与质子结合时会发生光谱变化（图1A-C）。因此，对pH敏感的荧光素的荧光发射的变化可以提供pH值的有用近似值。然而，单氟的使用存在一些缺点。例如，焦点平面的变化、光模糊和单个细胞体积的变化，在巨细胞体21中很常见，可诱发单个荧光素的荧光强度变化，而这不能轻易纠正为22。因此，单波长评估虽然有助于可视化酸性隔间，但纯粹是定性的。
更定量的方法是瞄准参数敏感的荧光，以及参考荧光素到感兴趣的细胞器。参考荧光对细胞内的生化变化（图1D-F）理想情况下不敏感，因此可用于纠正焦点平面、风琴体积的变化，以及在某种程度上光出血23。使用这种称为双荧光比的方法，可以通过生成参数敏感荧光的荧光排放与参考荧光的比例来实现校正。
在这里，协议将利用双氟比度成像原理来测量大皮诺索体内的pH值、氧化事件和蛋白质降解。在每种情况下，都会选择对兴趣参数和参考氟磷敏感的荧光素。为了将荧光素专门针对巨皮诺索，它们将共价耦合到70kDa dextran，优先地整合到大皮诺索24中。所有检测将在 Raw264.7 细胞中执行，但可以适应其他细胞类型。在可能的情况下，荧光比将根据参考曲线校准以获得绝对值。重要的是，所有测量将在活细胞中进行，以便对巨细胞体的亮度环境进行动态和定量评估。
在选择pH敏感荧光时，必须权衡一些考虑因素。第一个是荧光素的pKa，它表示探测器最敏感的pH值范围。如果假定在形成后不久，巨氨酸体的pH值将接近细胞外介质（+pH 7.2），并且它将通过与晚期内分泌体和溶酶体（+pH 5.0）的相互作用逐渐酸化，那么应选择具有pKa的探针，该探针在该范围内是敏感的（图2C）。氟氟素的pKa为6.4，在这个范围内具有最佳敏感性。它已被广泛用于测量其他类似的细胞器，如噬菌体，并将成为荧光在这份手稿22，25选择。作为氟的参考，将使用四甲基钠胺，这是麻木不仁的pH值（图1E）。其他氟，如普罗多和环丙烯可以替代荧光素，其中荧光素的光谱特性确实与其他实验变量匹配。一些建议参考氟为 pHrodo 和环孢子 5e 显示在图 1中。
第二个考虑因素是将这两种荧光素专门针对巨皮诺体的方法。70 kDa大小的Dextran，其水动力学半径约为7纳米，不粘在细胞上，并纳入巨无霸，但不是单体涂层坑或洞穴，因此标记巨无霸（图2A和图3A，B）16，24，26。在本协议中，荧光素标签的 70 kDa dextran 和四甲基钠 （TMR） 标记的 70 kDa dextran 将分别用作 pH 敏感和参考探头。
在先天免疫细胞中，巨脊细胞增多和噬菌体病是外源物质内化加工和随后向适应性免疫反应细胞呈现的两条主要途径。对噬菌体和巨皮诺体流明的氧化化学进行仔细和协调的控制，对于外源材料的上下文特定处理至关重要。也许在噬菌体中研究最充分的氧化事件调节器是NADPH氧化酶，这是一种大型多亚子综合体，在噬菌体28的流明内产生大量的活性氧物种（ROS）。事实上，其活性是适当抗原处理在噬菌体29，30的核心。然而，尚未探索大皮诺体膜上NADPH氧化酶的活性。
