以前，只有被动的中暑模型已经得到很好的建立。这种劳力性中暑模型没有主要限制，更接近于模仿人类病理生理学。不可能有效地研究人类中暑，因为它是一种危及生命的疾病。该临床前模型保留了人类劳力性中暑的临床特征。该技术有助于测试降温率对劳力性中暑恢复、药物摄入合并症以及中暑暴露的长期后果的影响。演示该程序的将是来自Clington博士实验室的高级博士生Jamal Alzahrani。麻醉小鼠后，使用眼部润滑剂，如兽医软膏，以保护动物的眼睛在手术过程中免受损伤或伤害。为了准备手术部位，用小动物理发器或市售脱毛剂剃除下腹部。给予第一剂皮下注射每公斤丁丙诺啡0.1毫克。用三洗聚维酮碘或氯己定擦洗该区域，然后冲洗70%异丙醇，然后将小鼠转移到手术区域，使用粘合剂悬垂物隔离小鼠上的手术部位。使用无菌技术的无菌器械，沿着距肋缘约0.5厘米的白线在中线上做一个约一厘米的切口，然后将皮肤与肌肉层分开，并在白线上做一个略小的切口，小心不要损伤肠道或内脏器官。一旦肌肉层打开，将无菌遥测计放入腹膜腔前的尾动脉和静脉以及消化器官的背侧，以使其自由漂浮。用无菌的5-O可吸收缝合线关闭腹部开口，并使用带有5-O脯氨酸缝合的简单中断缝合缝合皮肤。将鼠标放在干净的笼子里，笼子下面有一个依偎的微波炉加热垫。在麻醉恢复的第一个小时内，每15分钟监测一次小鼠，然后返回动物收容所。在劳力性中暑方案的前一天晚上，将小鼠置于室温下的环境室中以适应该室。使用数据采集系统收集连续的核心温度或TC平均值，间隔30秒。在劳力性中暑方案的早晨，确保小鼠处于或低于36至37.5摄氏度的日间温度的正常范围，然后再增加腔室温度，以确保小鼠在此期间没有发烧或经历过过度的压力。一旦小鼠稳定并在正常静息核心温度范围内，取出食物和水并称量动物。关闭腔室门，将腔室温度提高到37.5摄氏度和40%至50%相对湿度或所需的环境温度和湿度的目标。使用经过校准的温度和湿度监测器验证腔室温度和湿度。用遮光帘包围房间，以在协议期间将光线和干扰降至最低。在实验过程中，通过远程红外照明摄像机连续监控鼠标。将第二个摄像头对焦于靠近跑轮的温度和湿度监测器上。针对环境腔室设定点对控制器进行调整，以确保动物附近的温度读数准确。一旦腔室达到温度监视器上的第二个摄像头测量的目标温度，请快速打开腔室门并将鼠标放在强制运行轮中。以每分钟2.5米的速度启动强制运行轮协议，每10分钟将速度提高0.3米/分钟，直到鼠标达到41摄氏度的核心温度。一旦达到核心温度，让速度保持恒定，直到出现症状限制，其特征是明显的意识丧失，向后跌倒或昏厥，以及无法继续奔跑或抓住方向盘。确认鼠标在滚轮上有三次向后旋转而没有物理响应迹象的时间点。或者，按照本地 IACUC 规则确定人道端点，以确定何时停止协议。该终点略高于几乎所有小鼠的症状限制。要执行快速冷却协议，请在鼠标达到症状限制时停止滚轮，并立即从强制运行的滚轮上取下鼠标。称量鼠标并将其放回家笼中，以便在室温下恢复。在此期间，将培养箱门打开，并将培养箱设定点恢复到室温，以使培养箱快速冷却，从而产生大于99%的长期存活率。要进行更严重的劳力性中暑暴露，请在劳力性中暑方案期间将动物的笼子保持在37.5摄氏度的腔内。当动物达到症状限制时，让它们留在跑轮中，直到它们恢复远程摄像机观察到的意识，然后迅速将鼠标从跑轮中取出，直接将其返回到预热的笼子中，导致冷却曲线慢得多，基本上消除了劳累性中暑低温阶段。在此期间，从笼子中取下过滤器顶部，以改善与腔室的平衡。预冷却至室温的恢复笼可用于执行不太严重的替代程序，以导致抑制的低温期，存活率为100%对于严重的劳累性中暑方案，在恢复期间仔细监测小鼠并持续检查人道终点，包括恢复期间的正常运动，例如梳理， 正常呼吸和舔舐。在此期间监控核心温度。如果它们的核心温度在恢复阶段逆转方向，最终超过40摄氏度，小鼠不太可能恢复。在这种情况下，终止实验并继续评估小鼠的标准人道终点。在整个小鼠早期恢复的劳力性中暑方案期间，典型的体温调节曲线由四个不同的阶段组成，可以定义为腔室加热阶段，增量运动阶段，稳态运动阶段以及通过快速或慢速冷却方法恢复阶段。对于重度模型，恢复温度曲线表明，在整个两小时的恢复期内，核心温度保持在37摄氏度以上。在这个模型中，雌性小鼠对中暑的抵抗力更强，并且比雄性小鼠跑的距离长近两倍。两款样本在恢复到39.5摄氏度所需时间内没有观察到差异。然而，冷却到37.5摄氏度的环境温度的时间大大延长。最重要的是在手术后和劳累性中暑后始终仔细监测动物。这种方法有助于研究长达数月的中暑暴露的长期影响，并且还使我们能够测量中暑疾病中其他因素的影响，例如摄入非甾体抗炎药和表观遗传学。
