Bacteriële hechting is een proces waarbij bacteriën zich hechten aan andere cellen of oppervlakken. Succesvolle vestiging van infectie door bacteriële pathogenen vereist adhesie aan gastheercellen, kolonisatie van weefsels en in sommige gevallen invasie van gastheercellen1,2,3. Opkomende infectieziekten vormen grote bedreigingen voor de volksgezondheid, zoals blijkt uit de recente COVID-19-pandemie4,5,6. Belangrijk is dat nieuwe of opkomende pathogenen mogelijk niet gemakkelijk worden onderscheiden met behulp van genomische benaderingen, vooral in gevallen waarin de ziekteverwekker is ontworpen om detectie te ontwijken of geen genomische handtekeningen bevat die het als pathogeen identificeren. Daarom kan de identificatie van potentiële pathogenen met behulp van methoden die kenmerken van pathogeniciteit direct beoordelen, zoals bacteriële hechting aan gastheercellen, een cruciale rol spelen bij de identificatie van pathogenen.
Bacteriële hechting aan gastheercellen wordt al tientallen jaren gebruikt om mechanismen van bacteriële pathogenese te evalueren1,7. Microscopische beeldvorming8,9 en de opsomming van bacteriële kolonievormende eenheid (CFU)10,11,12,13 door post-infectie plating zijn twee goed ontwikkelde laboratoriummethoden voor het testen van microbiële hechting en / of infectie van gastheercellen14. Gezien de micrometerschaalgrootte van bacteriële cellen, vereist de inventarisatie van de aanhangende bacteriële cellen over het algemeen het gebruik van geavanceerde microscopietechnieken met hoge vergroting, evenals beeldvormingsbenaderingen met hoge resolutie, waaronder elektronenmicroscopie, expansiemicroscopie (ExM)15,16en driedimensionale beeldvorming17 . Als alternatief kan de opsomming van bacteriën die gebonden zijn aan of geïnternaliseerd zijn in gastheercellen worden uitgevoerd door de verdunningsreeks van geoogste bacteriën op vaste agar te platen en de resulterende CFU's10,12,13te tellen . Deze methode is bewerkelijk en omvat veel handmatige stappen, wat problemen oplevert bij het vaststellen van een gestandaardiseerde of geautomatiseerde procedure die vereist is voor analyses met een hoge doorvoer18,19. Daarom zou de ontwikkeling van nieuwe methoden voor het evalueren van gastheercelaanhechting de huidige beperkingen in het veld aanpakken.
Een dergelijke methode wordt hier beschreven die gebruik maakt van geautomatiseerde high throughput microscopie, gecombineerd met high throughput beeldverwerking en statistische analyse. Om de aanpak aan te tonen, werden experimenten met verschillende bacteriële pathogenen uitgevoerd, waaronder Pseudomonas aeruginosa, een opportunistische Gram-negatieve bacteriële ziekteverwekker van mensen, dieren en planten14,20, waarvan vaak wordt vastgesteld dat het de luchtwegen koloniseert van patiënten met verminderde afweerfuncties van de gastheer. Deze aanpak optimaliseerde het microscopische beeldvormingsproces beschreven in eerdere studies14,20. De beeldvormingsdetectie werd vereenvoudigd door fluorescentie-gelabelde gastheercellen en bacteriën om de nabijheid ervan snel te volgen, wat de microscopiebelasting drastisch verminderde om beelden met een hoge resolutie te krijgen voor het onderscheiden van bacteriën. Bovendien verving de geautomatiseerde statistische analyse van beelden bij het tellen van gastheercellen en bacteriën het hands-on experiment van bacteriële CFU-plating om de verhouding van het aantal adherente bacteriën per gastheercel te schatten. Om de compatibiliteit van deze methode te bevestigen, zijn ook meerdere bacteriestammen en gastheerceltypen getest, zoals Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus en Klebsiella pneumoniae, evenals menselijke navelstrengas endotheelcellen (HUVECs), en de resultaten ondersteunen de diversiteit en effectiviteit van de methode.

Identificatie van opkomende bacteriële pathogenen is van cruciaal belang voor de menselijke gezondheid en veiligheid. Bacteriële hechting aan gastheercellen is een essentiële stap in bacteriële infecties en vormt een kenmerk van potentiële bedreiging. Daarom kan het onderzoeken van de hechting van bacteriën aan gastheercellen worden gebruikt als onderdeel van bacteriële dreigingsanalyse. Een standaardmethode voor het opsommen van bacteriële hechting aan gastheercellen is om bacteriën samen met gastheercellen te incuberen, de aanhangende bacteriën te oogsten, de geoogste cellen op vaste media te plaatsen en vervolgens de resulterende kolonievormende eenheden (CFU) te tellen. Als alternatief kan bacteriële hechting aan gastheercellen worden geëvalueerd met behulp van immunofluorescentiemicroscopie-gebaseerde benaderingen. Conventionele strategieën voor het implementeren van deze benaderingen zijn echter tijdrovend en inefficiënt. Hier wordt een recent ontwikkelde geautomatiseerde fluorescentiemicroscopie-gebaseerde beeldvormingsmethode beschreven. In combinatie met high-throughput beeldverwerking en statistische analyse maakt de methode een snelle kwantificering mogelijk van bacteriën die zich hechten aan gastheercellen. Twee bacteriesoorten, Gram-negatieve Pseudomonas aeruginosa en Gram-positieve Listeria monocytogenes en bijbehorende negatieve controles, werden getest om het protocol aan te tonen. Deze methode biedt een snelle, high-throughput detectie van bacteriën die zich hechten aan gastheercellen. In vergelijking met de conventionele platingmethoden vermindert het de hands-on tijd die nodig is voor de kwantificering van de aanhangende bacteriën. Deze techniek is geautomatiseerd, tijdbesparend en zeer reproduceerbaar.Het is ook een veel toegepaste zowel op de meeste van de gastheer cellen. Deze methode, daar is heel eenvoudig. Voor de eerste proef moeten de omstandigheden zoals de veelheid van infectie en ook de co-incubatietijd van de gastheerbacteriën worden geoptimaliseerd.Oogst eerst de bacterieculturen in een exponentiële fase door centrifugering op 13.000 maal G gedurende twee minuten bij kamertemperatuur. Na het wassen van het cellenpallet met één milliliter PBS, resuspend het bacteriële palet in één milliliter PBS. Bepaal de concentraties van bacteriële suspensie door de optische dichtheid op 600 nanometer te meten.Voeg vervolgens voor het kleuren van de bacteriële suspensie twee microliter van de 500-voudig geconcentreerde bouillon groene of rode kleurstof toe in één milliliter bacteriële suspensie om de kleurstof één keer te verdunnen. Incubeer de cellen bij kamertemperatuur gedurende 30 minuten met zachte rotatie in het donker. Centrifugeer na 30 minuten de gekleurde bacteriën gedurende twee minuten op 13.000 keer G en resuspend de pellet in één milliliter PBS.Verzamel de gekleurde bacteriële cellen door centrifugering op 13.000 keer G gedurende twee minuten. Resuspend de pellet in één milliliter vers F12 K-medium en meet de optische dichtheid van elke cultuur op 600 nanometer. Verdun de culturen vervolgens tot de gewenste concentraties op basis van de veelheid van infectie en gastheercelconcentratie.Voor bacteriële hechtingstest, was de eerder gekweekte A549-cel monolagen drie keer door 100 microliter warme PBS aan elke put toe te voegen en pipetteer voorzichtig op en neer. Gooi pbs vervolgens weg. U kunt ook 10 seconden wachten nadat u PBS hebt toegevoegd en PBS vervolgens met vacuüm verwijderen.Om de kinetiek van bacteriële associatie te bepalen, bedekt u de cellen met 100 microliter gewenste concentraties bacteriële suspensie met verschillende veelheid van infectie. Spin de bacteriën en incubeer de geïnfecteerde A549-cellen bij 37 graden Celsius en 5% koolstofdioxide gedurende nog eens een uur. Verwijder de ongebonden bacteriën door de monolagen vijf keer te wassen met warme PBS zoals aangetoond.Fixeer vervolgens de cellen door 100 microliter 4% formaldehyde toe te voegen in alle putten van een 96-putplaat op het ijs. Was de plaat na 15 minuten drie keer met PBS om de fixatieoplossing te verwijderen. Beits in de kernen met 50 nanogram per milliliter DAPI-vlek gedurende 10 minuten bij kamertemperatuur.Na het wassen van de cellen met PBS, bedek de geïnfecteerde A549-cellen met 100 microliter PBS om droging te voorkomen en bewaar de plaat vervolgens op vier graden Celsius gedurende maximaal twee dagen in het donker of ga verder voor beeldvorming. Om de gegevensintegriteit te behouden, kiest u willekeurig en handmatig vijf locaties van elke put om de afbeeldingen vast te leggen met een vergroting van 20 keer. Leg de fluorescerende beelden van bacteriën vast onder GFP-kanalen, A549-cellen onder DAPI en gebruik voor de bacteriën gekleurd door de rode fluorescerende kleurstof PE Si vijfkanaals.Stel de parameters van de rollende bal in voor het afvlakken en meet automatisch de puntspreidingsfunctie van beelddeconvolutie op basis van het doel voor het bereiken van een betere resolutie. Meet de fluorescentie-intensiteit van de gastheercellen en bacteriën. Stel de zwakste fluorescentie-intensiteit van gastheercellen en bacteriën in als de drempels voor het aantal cellen en tel alle bacteriën die zich binnen 15 micrometer afstand van een gastheercel bevinden als de hechtingsbacteriën.Zodra de cellen zijn geteld uit alle geautomatiseerde afbeeldingen, analyseert u kritische uitlezingen, zoals het aantal gastheercellen, celgrootten en -vormen, het totale aantal bacteriën en het gemiddelde bacteriële account per gastheercel, wat de belangrijkste indicator is om bacteriële hechting te bepalen. Na een uur co-incubatie hechtte pseudomonas aeruginosa stam PAO1 zich op dosisafhankelijke wijze aan A549-cellen. Vergelijkbare resultaten werden ook waargenomen bij grampositieve bacteriën listeria monocytogenes en de negatieve controle bacillus subtilis.Een representatief beeld van gastheertrouw P aeruginosa segmentatie toont het gemak van het tellen van bacteriële cellen met behulp van dit protocol. De resultaten van de geautomatiseerde analyses omvatten bacteriële en gastheerceltellingen, de gemiddelde bacteriële tellingen per gastheercel en gastheercelgroottes en gebieden die de status van de gezondheid van gastheercellen, bacteriële hechtingsniveau en bacteriële cytotoxiciteit vertegenwoordigen. Naast A549-cellen werden ook HUVECs getest op therapietrouwtest.Serratia rubidaea en streptococcus agalactiae waren wel aanhangend aan HUVEC, maar niet aan A549-cellen, en dat toont de effectieve detectie en geschiktheid van meerdere gastheren aan om de interacties van de gastheerbacteriën met behulp van deze methode te bestuderen. Gastheercellen moeten ongeveer 90 tot 100% samenvloeiing bereiken om de ongewenste bacteriehechting aan de bodem van de putten te minimaliseren. Met deze techniek kunnen onderzoekers de nieuwe mechanismen verkennen, de interacties tussen gastheer en plaag.
