この方法は、細菌が宿主細胞に付着する迅速で高スループットの検出を提供する。従来のめっき法と比較して、接着性細菌の定量に必要な実地時間を短縮する。この技術は自動化され、時間を節約し、非常に再現可能です。また、ほとんどの宿主細胞に両方を広く適用した。この方法は、非常に簡単です。最初の試験では、感染の多重度、および宿主菌の共培養時間などの条件を最適化する必要があります。まず、室温で2分間Gの13,000倍の遠心分離によって指数相で細菌培養を収穫する。細胞パレットを1ミリリットルPBSで洗浄した後、細菌パレットを1ミリリットルPBSで再懸濁する。600ナノメートルで光学濃度を測定することにより、細菌懸濁液の濃度を決定します。次いで、細菌懸濁液を染色するために、500倍の濃縮ストックグリーンまたは赤色染色染料の2マイクロリットルを1ミリリットルの細菌懸濁液に加えて、染料を1倍に希釈する。暗い中で穏やかな回転で30分間室温で細胞をインキュベートします。30分後、染色した菌を13,000倍Gで2分間遠心し、PBSの1ミリリットルでペレットを再懸濁する。染色した菌体を13,000倍Gで2分間遠心分離して回収する。新鮮なF12K培地の1ミリリットルにペレットを再懸濁し、600ナノメートルで各培養物の光学密度を測定する。感染の多重度および宿主細胞濃度に基づいて所望の濃度に培養物を後で希釈する。細菌付着アッセイの場合は、以前培養したA549細胞モノ層を100マイクロリットルの温かいPBSをそれぞれに加えて3回洗浄し、ピペットを上下に軽くピペットします。その後、PBSを破棄します。あるいは、PBSを追加した後、10秒間待ってから、真空を使用してPBSを取り除きます。細菌の関連の運動学を決定するには、異なる多重度の感染を有する細菌懸濁液の所望の濃度の100マイクロリットルで細胞を重ね合わせる。細菌をスピンダウンし、感染したA549細胞を摂氏37度、5%の二酸化炭素でさらに1時間インキュベートします。実演したように、モノ層を暖かいPBSで5回洗浄して、非結合菌を除去する。次に、氷上の96ウェルプレートのすべてのウェルに4%ホルムアルデヒドの100マイクロリットルを加えることによって細胞を固定します。15分後、プレートをPBSで3回洗浄し、固定液を除去する。1ミリリットルのDAPI染色で核内に10分間染色する。PBSで細胞を洗浄した後、乾燥を避けるために100マイクロリットルのPBSで感染したA549細胞を覆い、暗闇の中で最大2日間摂氏4度でプレートを保存するか、イメージングに進みます。データの整合性を維持するには、ランダムに手動で各ウェルの5つの場所を選択し、20倍の倍率で画像をキャプチャします。GFPチャネル下の細菌の蛍光画像をキャプチャし、DAPI下のA549細胞、および赤色蛍光色素で染色された細菌についてはPE Si 5チャネルを使用します。平滑化のためのローリングボールのパラメータを設定し、より良い解像度を達成するための目的に基づいて、画像デコンボリューションの点広がり関数を自動測定します。宿主細胞および細菌の蛍光強度を測定する。宿主細胞および細菌の最も弱い蛍光強度を細胞数の閾値として設定し、すべての細菌を15マイクロメートル以内の宿主細胞に近接する細菌を付着菌として数える。すべての自動画像から細胞がカウントされたら、ホスト細胞数、細胞サイズと形状、総細菌数、および細菌の付着を決定するための最も重要な指標である宿主細胞あたりの平均細菌数などの重要な読み出しを分析します。1時間の共培養の後、緑膿菌株PAO1は用量依存的な方法でA549細胞に付着した。同様の結果は、グラム陽性菌リステリア単球遺伝子およびその陰性対照菌サブティリスの場合にも観察された。宿主の付着P緑膿性のセグメンテーションの代表的な画像は、このプロトコルを使用して細菌細胞を数える容易さを示している。自動化された分析の結果には、細菌および宿主細胞数、宿主細胞および宿主細胞サイズおよび宿主細胞の健康状態を表す領域、細菌の付着レベルおよび細菌細胞毒性の平均細菌数が含まれる。A549細胞とは別に、HUVECは付着アッセイについても試験した。セラチア・ルビダエアおよびレンサ球菌アガラクチオーはHUVECに付着したが、A549細胞には付着しておらず、この方法を用いて宿主菌の相互作用を研究する複数の宿主の有効な検出および適合性を示している。宿主細胞は、ウェルの底面に対する望ましくない細菌の付着を最小限に抑えるために、約90〜100%の合流に達するべきである。この技術は、研究者が新しいメカニズム、宿主と害虫の相互作用を探求することを可能にする。
