アデノ関連ウイルス(AAV)は、4.6kbの一本鎖DNAを含む非エンベロープヒトパルボウイルスである。AAVベクターは、遺伝子治療アプリケーション1、2、3、4に対する他のウイルスベクターに対していくつかの利点があります。AAV は自然にレプリケーションを行う必要がなっており、それにより、レプリケーションにはヘルパー ウイルスとホストマシンが必要になります。AAVは、感染した宿主3,5において疾患を引き起こせず、免疫原性が低い。AAVは静止細胞と積極的に分裂する細胞の両方に感染し、宿主細胞のゲノムに統合することなくエピソームとして持続する(AAVはめったに宿主ゲノムに統合しない)1,3。これらの特徴はAAVを遺伝子治療の適用のための望ましい用具にした。
AAV遺伝子導入ベクターを生成するために、治療用遺伝子を含むトランスジーンカセットは、2つの内部末端反復(ITR)の間でクローニングされ、これは典型的にはAAV血清型2に由来する。5' ITR から 3' ITR までの最大サイズは、トランスジーン配列を含めて 4.6 kb6 です。異なるキャプシドは、異なる細胞または組織のトロピズムを有することができる。したがって、カプシドは、AAVベクター7で標的とされることを意図した組織または細胞タイプに基づいて選択されるべきである。
組換えAAVベクターは、ヒト胚性腎臓細胞などの哺乳動物細胞株において一般的に産生されるが、HEK293はAAV遺伝子導入ベクターの一過性トランスフェクションによって、AAV rep-cap、およびヘルパーウイルスプラスミド2,3である。しかしながら、付着性HEK293細胞の一過性トランスフェクションによる大規模なAAV産生にはいくつかの制限がある。まず、多数のセルスタックまたはローラーボトルが必要です。第2に、高品質のプラスミドDNAとトランスフェクション試薬が必要であり、製造コストが増加します。最後に、付着性HEK293細胞を使用する場合、血清は最適な生産のために頻繁に必要とされ、下流処理1、2、3を複雑にする。AAV製造の代替方法は、昆虫細胞株、スポドプテラ・フルギペルダ(Sf9)細胞、および組換えオートグラフ・カリフォルニカ多重重体核多重体症ウイルス(AcMNPVまたはバクロウイルス)8、9、10と呼ばれる昆虫ウイルスを使用することを含む。Sf9細胞は、スケールアップが容易で、現在の良好な製造実践(cGMP)生産と大規模な製造に対応する無血清懸濁液培養で増殖し、プラスミドやトランスフェクション試薬を必要としません。また、Sf9-バキュロウイルス系を用いたAAV産生のコストは、HEK293細胞11へのプラスミドの一過性トランスフェクションを用いたコストよりも低い。
バキュロウイルス-Sf9細胞を用いた元のrAAV産生系は、3つのバキュロウイルスを用いた:遺伝子導入カセットを含む1つのバキュロウイルス、Rep遺伝子を含む第2のバキュロウイルス、および血清型特異的キャプシド遺伝子12、13を含む第3のバキュロウイルス。しかし、rep構築物を含むバキュロウイルスは、複数の通過時に遺伝的に不安定であったため、大規模なAAV産生のためのバキュロウイルスの増幅を妨げた。この問題を解決するために、2つのバキュロウイルス(TwoBac)を含む新しいrAAVベクターシステムが開発されました:AAV遺伝子導入カセットを含む1つのバキュロウイルスと、元のシステムよりも遺伝的に安定しており、3つの14の代わりにTwoBacを使用しているためにrAAVを生成するのに便利なAAVレップ遺伝子を含む別のバキュロウイルス、 15.OneBacシステムは、単一のバキュロウイルスにおいてAAV遺伝子導入カセットおよびRep-cap遺伝子を用い、TwoBacまたはThreeBac2、16、17を使用する代わりに1つのバキュロウイルスを使用するため、rAAVを産生する方が便利である。我々の研究では、TwoBacシステムは最適化のために使用された。

このプロトコルは、Baculoウイルス昆虫細胞培養システムを用いたAAV産生および精製の方法を開発したい研究者に有用である。バキュロウイルス昆虫細胞培養システムを用いたAAV生産は、スケールアップが容易で、現在の良好な製造慣行と互換性のある費用対効果の高い方法です。プロトコルの一部は、私の研究室の研究助手であるアラン・ハイザーによって実証されます。まず、Sf9細胞のバイアルを1本解凍し、すぐに15ミリリットルの昆虫細胞培養培地に播種します。125 RPMで軌道シェーカーインキュベーターでSf9細胞を成長させ、空気交換のために緩く取り付けられたキャップを持つ丸底フラスコで摂氏28度。200ミリリットルの培地を含む1リットルフラスコで細胞を伝播し、TIPS細胞産生に十分な細胞を得る。2つのフラスコで1ミリリットル当たり6番目のパワーセルに2倍10でSf9細胞の50ミリリットルを追加します。Sf9細胞の1フラスコに0.5ミリリットルのバキュロウイルス-AAV2-GFP、もう1フラスコのバキュロウイルス-AAV2-rep-capを0.5ミリリットルの細胞に感染させる。3日後、バキュロウイルス感染Sf9細胞の小さなアリコート(TIPS細胞とも呼ばれる)を収穫する。暗い環境下で30分間、100マイクロリットルのブロッキングバッファーに蛍光色素を含む0.5マイクログラムのマウス抗バキュロウイルスgp64抗体を有する未感染およびバキュロウイルス感染Sf9細胞の5番目のパワーの20倍に10回染色する。細胞をPBSで洗浄して抗体を除去した後、0.5ウシ血清アルブミンを含むPBSの300マイクロリットルで染色された細胞を再懸濁する。フローサイトメトリーを用いて細胞上でのバキュロウイルスgp64発現を解析する。細胞が80〜90%生存可能である場合、直径の増加に伴い、細胞を1ミリリットルの昆虫培養培地で1回10〜7番目のパワー細胞に収穫し、10%の胎児ウシ血清と10%のジメチルスルホキシドをマイナス80°Cの遅凍結容器に置き換える。翌日、TIPS細胞を含むチューブを液体窒素冷凍庫に移し、長期保存します。400ミリリットルの昆虫細胞培養液を含む2リットルフラスコで、2倍10〜6番目のパワーセルで2倍の10~6番目のパワーセルで、アデノ関連ウイルスまたはAAV産生に必要なシェーカーインキュベーターで培養し、ナイーブSf9細胞を増殖させる。3〜4日後、細胞密度は1ミリリットル当たり6番目のパワーセルに最大5〜6倍に増加する。軌道シェーカーインキュベーターのフラスコでのAAV生産の場合、ピペットまたは蠕動ポンプを使用して、20倍のSf9培養物を20〜6番目のパワーセルで200ミリリットルのフラスコに播種します。軌道シェーカーを125 RPM、摂氏28度に設定します。各バキュロウイルス-AAV-GFPとバキュロウイルス-AAV-rep-cap TIPS細胞の1バイアルを解凍します。20ミリリットルの昆虫培養培地で細胞を希釈し、トリパンブルーを用いて細胞の生存率カウントを行う。シェイカーフラスコまたはバイオリアクターで培養したナイーブSf9細胞に対して1〜10,000の比率で両方のTIPS細胞を接種する。2日目には、無菌的にSf9培養の1ミリリットルを収穫し、トリパンブルー染料で染色して細胞数、生存率、形態を分析する。3日目に、2日目に行われたステップを繰り返し、Sf9細胞を染色した後のバキュロウイルス感染の状態を確認します。5日目に、2日目に行ったステップを繰り返し、Sf9の生存率が50%程度に低下した場合に培養培地と細胞を収穫し、紡糸後に上清と細胞ペレットを集め、80°Cの負の80度で保存します。AAVプロデューサ細胞ペレットが解凍されたら、細胞のリシスバッファーを加え、激しく混合します。再懸濁したペレットを周囲温度で30分間インキュベートし、AAV粒子を放出する。遠心分離後、細胞のライセートを新しい容器に移します。解凍後に細胞ライセートと細胞培養上清を混ぜます。1ミリリットルのヌクレアーゼあたり20単位、塩化マグネシウム10ミリモルを細胞ライゼに加えてDNAとRNAを消化し、摂氏37度で2~4時間インキュベートします。0.8マイクロメートルと0.2マイクロメートルのポリエーテルサルホン二重ろ過システムをポンプを使用して、ライセートをフィルター処理します。明確化した細胞ライセートを摂氏4度で一晩保管するか、すぐにAAVを精製します。クロマトグラフィーマシンに10ミリリットルAVBセファローズカラムを取り付け、パイプラインをAAVサンプル、洗浄バッファ、溶出バッファーに挿入します。クロマトグラフィーマシンのフラクションコレクタースロットにチューブを挿入し、AAV粒子を含むカラムパススルー溶液、洗浄バッファー、溶出バッファーの画分を収集します。AVBセファローズカラムを、毎分5ミリリットルの流量でPBSの5カラム体積で平衡化する。サンプルポンプを搭載したクロマトグラフィーマシンを使用して、AAV粒子を含む濾過細胞溶解物をAVBセファローズ親和性カラムにロードします。1 分間に 3 ミリリットルの流量でサンプルを実行します。280ナノメートルの紫外線吸光度曲線がベースラインに戻り、安定するまで、1分間に3ミリリットルの流量でカラムを通してPBSを実行します。AVBセファローズカラムからAAV粒子を1分間に3ミリリットルの流量でクエン酸50ミリモルのクエン酸緩衝液pH3で溶出する。AAVを含む酸性溶出バッファーを中和するために、500 ミリモルトリス HCl pH 8.0 の 1/5 体積で AAV 上清をすぐに希釈します。これは酸性pH 3.0溶出緩衝液と起こりうるpH媒介分解を防ぐ。次に、AAV上清をPBSで10倍に希釈し、AAV粒子が生理学的緩衝液に入るようにする。各カラムランスルーサンプルの1ミリリットルを保存し、洗浄バッファー、および100マイクロリットルの溶出したAAV上清を標的細胞の感染によるAAVの存在を評価する。AAVを濃縮するために100キロの分子量カットオフを持つポリエーテルサルホン膜カートリッジを装備し、接線流ろ過システムを設定します。200ミリリットルのPBSを10分間実行し、接線流ろ過モジュールを平衡化します。サンプルが所望の容積に減少するまでポンプの流れを制御することによってAAVサンプルをロードしなさい。100ミリリットルのPBSで再狭窄をダイアフィルする。AAVサンプルを所望の容積に濃縮した後、AAVサンプルを回収し、0.2マイクロメータのポリエーテルスルホンフィルターを通してそれを濾過し、それをアリコートします。次に、AAVサンプルをマイナス80°Cで保存します。Sf9細胞のほとんどは、バキュロウイルス糖タンパク質gp64発現によって証明された感染の複数のラウンドのために、3〜4日でバキュロウイルスに感染した。細胞の大部分はまた、直径の増加を示す。細胞は、直径の増加を示しています, 細胞パシー効果, そして、細胞の約半分は、感染後5日間で死ぬ, AAV産生の完了の兆候である.AAV画分に相当する酸性緩衝液を用いて溶出している間に、タンパク質のピークが見られた。精製されたAAVサンプルは、SDS-PAGEと銀染色後の3つの異なるキャプシドタンパク質を示しています:VP1、VP2、VP3。最も重要なステップは、細胞のほとんどがバキュロウイルスに感染し、細胞死の最小数で、凍結保存の前にTIPS細胞を収穫することです。我々は、血清型2のAAVベクターの製造及び精製をモデルとしてここで説明した。ただし、プロトコルは、他の AAV 血清型にも適用できます。
