リボヌクレオプロテイン(RNP)顆粒は、均質な流体が2つの異なる液体相に分解する現象である液液相分離(LLPS)を介して膜のないパーティションを組み立てることによって、核および細胞質における無数の細胞活動を制御する。RNP顆粒成分として通常機能するRNA結合タンパク質の調節不変LLPSは、異常な相転移を促進し、タンパク質凝集をもたらす。このプロセスは、神経発達および神経変性疾患に関与している3,4,5.RNA結合タンパク質の異常なLLPSと疾患の病態との因果関係の正確な評価は、LLPSが効果的な治療標的として利用できるかどうかを決定する上で極めて重要である。RNA結合タンパク質のLLPSは、インビトロや単細胞モデルでは比較的容易に研究できますが、多細胞生物、特に脊椎動物では困難です。組織環境内の個々の細胞におけるこのようなLLPSを分析するための重要な要件は、疾患に脆弱な細胞タイプの目的でLLPSのイメージングおよび操作のためのプローブを安定的に発現させることである。
筋萎縮性側索硬化症(ALS)は、脳と脊髄の運動ニューロンが変性のために選択的かつ徐々に失われる、最終的に致命的な神経疾患である。現在までに、25以上の遺伝子の突然変異は、ALS症例全体の5%-10%を占めるALSの遺伝性(または家族性)形態と関連しており、これらのALSを引き起こす遺伝子の一部はhnRNPA1、TDP-43、FUS6,7などのRNPからなるRNA結合タンパク質をコードする。さらに、ALSの散発的な形態は、ALS症例全体の90%〜95%を占め、退化運動ニューロンに沈着したTDP-43の細胞質凝集によって特徴付けられる。これらのALS関連RNA結合タンパク質の主な特徴は、本質的に障害領域(IDR)または低複雑性ドメインであり、秩序ある三次元構造を欠き、LLPS7,8を駆動する多くの異なるタンパク質との弱いタンパク質タンパク質相互作用を媒介する。ALSを引き起こす突然変異がIDRでしばしば起こるという事実は、これらのALS関連タンパク質の異常なLLPSおよび相転移がALS病因の根源であるかもしれないという考えにつながっています。
近年、光光によるタンパク質相互作用の調節を可能にするクリプトクロム2ベースの光遺伝学的手法である光滴法が、IDRs11を用いたタンパク質の相転移を誘導するために開発された。この技術がTDP-43に正常に拡張されたので、TDP-43の病理学的相転移の基礎となるメカニズムとそれに関連する細胞毒性12、13、14、15を明らかにし始めた。このプロトコルでは、ALSに脆弱な細胞型、すなわち、運動ニューロン仕様用ホメオドメインタンパク質をコードするmnr2b/mnx2b遺伝子のBACを用いてゼブラフィッシュの脊髄運動ニューロンに光遺伝学的TDP-43を送達する遺伝的方法の概要を示す16,17。ゼブラフィッシュ幼虫の高い半透明性は、脊髄運動ニューロンにおけるその相転移を引き起こす光遺伝学的TDP-43の単純で非侵襲的な光刺激を可能にする。また、光刺激に対する光遺伝学的TDP-43の応答を特徴付けるために、自由に入手可能なフィジー/ImageJソフトウェアを用いたゼブラフィッシュ脊髄運動ニューロンのライブイメージングと画像解析の基本的なワークフローも提示する。これらの方法は、ALSに脆弱な細胞環境におけるTDP-43相転移の調査を可能にし、細胞および行動レベルでの病理学的結果を探求するのに役立つべきである。
