Het muis RVO-model biedt een manier om RVO-pathologie verder te begrijpen en potentiële therapieën te testen. Hoewel het muis RVO-model veel wordt gebruikt in het veld, is er behoefte aan een actueel gedetailleerd protocol van het model dat de variabiliteit aanpakt en de optimalisatie van het model beschrijft. Hier bieden we een gids met voorbeelden uit ervaring over wat kan worden gewijzigd om de meest consistente resultaten te krijgen voor een cohort van proefdieren en betrouwbare gegevens te verstrekken.

Retinale ader occlusie (RVO) is een veel voorkomende retinale vaatziekte die in 2015 wereldwijd ongeveer 28 miljoen mensen trof1. RVO leidt tot achteruitgang en verlies van het gezichtsvermogen bij werkende volwassenen en ouderen, wat neerkomt op een voortdurende gezichtsbedreigende ziekte die naar schatting in de loop van het nabije decennium zal toenemen. Enkele van de verschillende pathologieën van RVO omvatten hypoxisch-ischemisch letsel, retinaal oedeem, ontsteking en neuronaal verlies2. Momenteel is de eerste behandelingslijn voor deze aandoening door de toediening van vasculaire endotheliale groeifactor (VEGF) -remmers. Hoewel anti-VEGF-behandeling heeft geholpen bij het verbeteren van retinaal oedeem, worden veel patiënten nog steeds geconfronteerd met een achteruitgang van het gezichtsvermogen3. Om de pathofysiologie van deze ziekte verder te begrijpen en om potentiële nieuwe behandelingslijnen te testen, is het nodig om een functioneel en gedetailleerd RVO-muismodelprotocol voor verschillende muizenstammen samen te stellen.
Er zijn muismodellen ontwikkeld die hetzelfde laserapparaat implementeren dat wordt gebruikt bij menselijke patiënten, in combinatie met een beeldvormingssysteem dat is geschaald naar de juiste grootte voor een muis. Dit muismodel van RVO werd voor het eerst gerapporteerd in 20074 en verder vastgesteld door Ebneter en anderen 4,5. Uiteindelijk werd het model geoptimaliseerd door Fuma et al. om belangrijke klinische manifestaties van RVO te repliceren, zoals retinaal oedeem6. Sinds het model voor het eerst werd gerapporteerd, hebben veel studies het gebruikt met behulp van de toediening van een fotosensitizerkleurstof gevolgd door fotocoagulatie van belangrijke retinale aderen met een laser. De hoeveelheid en het type kleurstof die wordt toegediend, het laservermogen en de tijd van blootstelling variëren echter aanzienlijk tussen studies die deze methode hebben gebruikt. Deze verschillen kunnen vaak leiden tot een verhoogde variabiliteit in het model, waardoor het moeilijk te repliceren is. Tot op heden zijn er geen gepubliceerde studies met specifieke details over mogelijke wegen voor de optimalisatie ervan.
Dit rapport presenteert een gedetailleerde methodologie van het RVO-muismodel in de C57BL/6J-stam en een tamoxifen-induceerbare endotheel caspase-9 knockout (iEC Casp9KO) stam met een C57BL/6J achtergrond en van belang voor RVO pathologie als referentiestam voor een genetisch gemodificeerde muis. Een eerdere studie had aangetoond dat niet-apoptotische activering van endotheel caspase-9 retinaal oedeem aanwakkert en neuronale dood bevordert8. Ervaring met het gebruik van deze stam heeft geholpen bij het bepalen en inzichtelijk maken van mogelijke aanpassingen om het RVO-muismodel, dat toepasbaar kan zijn op andere genetisch gemodificeerde stammen, aan te passen.

Muismodellen van retinale ader occlusie (RVO) worden vaak gebruikt in de oogheelkunde om hypoxisch-ischemisch letsel in het neurale netvlies te bestuderen. In dit rapport wordt een gedetailleerde methode gegeven die wijst op kritieke stappen met aanbevelingen voor optimalisatie om consistent succesvolle occlusiepercentages te bereiken bij verschillende genetisch gemodificeerde muizenstammen. Het RVO-muismodel bestaat voornamelijk uit de intraveneuze toediening van een fotosensitizerkleurstof gevolgd door laserfotocoagulatie met behulp van een retinale beeldvormingsmicroscoop die is bevestigd aan een oogheelkundige geleide laser. Drie variabelen werden geïdentificeerd als determinanten van occlusieconsistentie. Door de wachttijd na toediening van rose bengalen aan te passen en de baseline en experimentele laseroutput in evenwicht te brengen, kan de variabiliteit tussen experimenten worden beperkt en een hoger slagingspercentage van occlusies worden bereikt. Deze methode kan worden gebruikt om retinale ziekten te bestuderen die worden gekenmerkt door retinaal oedeem en hypoxisch-ischemisch letsel. Bovendien, omdat dit model vasculair letsel induceert, kan het ook worden toegepast om de neurovasculatuur, neuronale dood en ontsteking te bestuderen.
