網膜静脈閉塞症(RVO)は、2015年に世界中で約2,800万人が罹患した一般的な網膜血管疾患です1。RVOは、働く高齢者と高齢者の視力低下と喪失につながり、近所の10年間で増加すると推定される進行中の視力を脅かす病気を表しています。RVOの明確な病状には、低酸素虚血性損傷、網膜浮腫、炎症、およびニューロン喪失が含まれます2。現在、この障害の治療の第一線は、血管内皮増殖因子(VEGF)阻害剤の投与によるものです。抗VEGF治療は網膜浮腫の改善に役立っていますが、多くの患者は依然として視力低下に直面しています3。この疾患の病態生理学をさらに理解し、潜在的な新しい治療ラインをテストするには、さまざまなマウス系統に対して機能的で詳細なRVOマウスモデルプロトコルを構成する必要があります。

本報告では、C57BL/6J系統のRVOマウスモデルと、C57BL/6Jをバックグラウンドとし、遺伝子改変マウスの基準株としてRVO病理に関連するタモキシフェン誘導性内皮カスパーゼ-9ノックアウト(iEC Casp9KO)株の詳細な方法論を提示する。以前の研究では、内皮カスパーゼ-9の非アポトーシス活性化が網膜浮腫を引き起こし、神経細胞死を促進することが示されていました8。この株を使用した経験は、他の遺伝子組み換え株に適用できるRVOマウスモデルを調整するための潜在的な改変を決定し、洞察を提供するのに役立ちました。
