使用急性脑切片培养物来建立该方案，因为它们（1）通过一些练习，易于产生;（2）提供一个可访问的系统来研究嵌入在准完全生理环境中的生长锥，但足够透明以允许高分辨率活细胞成像;（3）可与其使用无数转基因小鼠系扩展使用;（4）与IUE或EUE结合使用，提供几乎无限的潜力，以提供分子工具，以评估在功能状态的丧失/增益下体内生长锥和轴突的性能，以及荧光报告基因和细胞骨架探针。

该协议使得对组织结构切片内的神经元进行超分辨率成像成为可能。从历史上看，光散射是成像厚标本时面临的一个重大问题。在过去的二十年中，光学技术的广泛进步使厚标本的成像成为可能。在这里，使用长工作距离物镜来更好地可视化较小的结构，例如生长锥。不可避免的是，该协议不会捕获更详细的事件，例如逆行肌动蛋白流或微管动力学。长工作距离物镜需要较低的数值孔径（NA），以保留来自厚切片的信息。但是，也可以调整该协议以用于更短工作距离的目标。这需要将切片平稳地转移到玻璃底培养皿中，以保持结构完整性。然而，由于气体交换的损失，使用这种方法导致存活期缩短-〜15小时（数据未显示）。与 2D 培养物不同，3D 中的生长锥占据更大的体积，并且需要在 z 轴上进行运动伪影补偿。为了提高对详细事件进行成像的能力，必须利用现代共聚焦技术。因此，建议使用快速扫描的z轴堆叠电机，例如高灵敏度共聚焦显微镜33上可用的z-Galvo。
值得注意的是，该协议存在三个主要限制。首先，在 体内控制任何给定质粒的表达水平/表达细胞数量通常具有挑战性。这引入了所有切片之间的可变性，即使在保持相同的质粒浓度时也是如此。因此，在所用表达载体中调节元件的选择必须谨慎地预先确定。其次，使用膜插入物对详细事件进行成像目前尚不可行。这第二个限制可以通过上一段所建议的方法更新来克服。最后，生长锥具有高度光敏性，可以迅速变成光漂白。因此，频繁成像生长锥细胞，使用激光扫描显微镜短短5分钟，通常可以使生长锥塌陷。在这点上，光片显微镜产生的装置的新进展可以适应于脑切片34的长期成像。
这种类似的协议被设想为开辟新的研究途径，从而更好地了解生长锥体阅读和应对复杂的 体内 环境所需的条件，更重要的是，解开这种复杂相互作用的机制。

从电穿孔胚胎中解剖大脑是一个关键步骤，需要仔细进行以获得高质量的切片，从而保留天然的大脑结构。事先制备解剖器械和振动切片机并仔细乙醇灭菌（图3A，B）。接下来，仔细解剖电穿孔胚胎的头部并提取大脑。在这里，显示了从E15（图3C-F）和E12.5（图3G-J）处进行EUE的胚胎的代表性大脑解剖。将大脑立即包裹在琼脂糖基质中，切片，并放置在底部玻璃培养皿内的PTFE膜插入物上进行孵育（图3K-M）。
脑切片的健康状况是控制以确保可靠结果的重要点。每天对任何污染物进行目视检查。此外，一旦培养完成，脑切片被固定并进行免疫组化。在这里，4'，6-二脒-2-苯基吲哚（DAPI）用于控制整体细胞组织和维门汀染色以揭示神经胶质组织;特别是径向神经胶质细胞（RG）支架。通常，从IUE或EUE衍生的成功培养的脑切片显示出DAPI和具有顶端导向pial接触过程的某种有组织的RG阵列17 所揭示的正常细胞分布（分别为图4A，B）。偶尔，在培养的脑切片中观察到RG支架的显着紊乱，特别是在EUE电穿孔衍生的那些中（图4C）。具有极度无序RG支架的脑切片显示神经元迁移受损和轴突生长缺陷（未显示）。因此，控制RG支架是一种简单的培养后方法，可以对从可靠的脑切片获得的数据进行分类。
来自IUE或EUE的脑切片与Lyn-mNeonGreen表达质粒混合物导致类似的稀疏神经元标记。以表达Lyn-mNeonGreen及其生长锥体的动态行为为示例显示一个代表性的金字塔CPN（图5A 和 补充视频1， 左上角）。此外，使用表达肌动蛋白探针的质粒标记神经元，以 原位 分析轴突生长锥的肌动蛋白动力学（图5B 和 补充视频1， 左下角）。还使用双Cre / Dre荧光团表达质粒设计进行 原位 实验（图1C 和 补充视频1，右）。该质粒中的tRFP或ZsGreen荧光团可以分别由邻近神经元中的Dre或Cre重组酶特异性和单独激活（图5C）。该实验阵容允许并排分析来自具有相邻修饰神经元的控制神经元的生长锥（任何给定的功能损失或增益）。这规避了因使用不同切片来测试控制和实验条件而产生的可变性。
分析从记录的电影中生成的Kymographs，从中可以很容易地获得动态生长参数，例如随时间推移的突出活动和生长长度（图6A）。请注意，对延时时间分辨率进行简单调整，可以测量轴突伸长速度2小时（图6A）。此外，生长锥体积随时间的变化 - 一般生长锥动态活动的度量 - 可以很容易地获得，在这种情况下使用许可软件（图6B 和 图6E，F）。这可用于评估肌动蛋白跑步机的速度以及生长锥探索活动期间丝状体/薄片的平衡。
