Recycling-endosomen zijn een cohort van endocytische organellen en ze bemiddelen de recycling van lading naar het celoppervlak in alle celtypen21,22,23,24,25. In gespecialiseerde celtypen zoals melanocyten leiden deze organellen hun smokkelroute gedeeltelijk om naar de melanosomen voor hun biogenese3,16,26.<...

Biosynthese van melaninepigmenten vindt plaats in melanosomen, een melanocyt-specifiek lysosoom-gerelateerd organel (LRO) dat naast conventionele lysosomen bestaat. Het endocytische systeem speelt een sleutelrol in de biogenese van melanosomen, die nodig zijn voor huidskleur en fotoprotectie tegen ioniserende straling1,2,3. Tijdens dit proces worden de melaninesynthetiserende enzymen gesorteerd op vroege / sorteer-endosomen en vervolgens getransporteerd naar premature melanosomen via tubulaire of vesiculaire endosomen die recycling-endosomen (RE's) worden genoemd 4,5,6,7,7,8,9,10. De targeting en fusie van deze organellen reguleren de rijping van volledig functionele gepigmenteerde melanosomen7,11,12,13,14. Defecten in de vorming van deze organellen of ladingssortering naar deze organellen veroorzaken oculocutaan albinisme en andere klinische fenotypen, waargenomen bij Hermansky-Pudlak-syndroom15,16.
Hier beschrijven we een eenvoudige op microscopie gebaseerde techniek om de RE's te bestuderen en te analyseren. Bij deze methode hebben we gebruik gemaakt van een transmembraaneiwit, Qa-SNARE Syntaxin (STX)13, dat zich bevindt op het recyclen van endosomen17 en cycli tussen het sorteren van endosomen en melanosomen in melanocyten12,18. Verder zorgt het verwijderen van N-terminaal ongestructureerd regulerend domein (namelijk SynN of STX13Δ129) ervoor dat de SNARE vast komt te zitten in melanosomen, die de voorwaartse smokkelroute naar het melanosoom meet12. We hebben een bekende recycling endosomale marker Rab GTPase (Rab)11 gebruikt in onze studies14,19. Fluorescentiebeeldvorming van de eiwitten GFP-STX13WT, GFP-STX13Δ129, mCherry-Rab11 en TYRP1 in melanocyten van het wilde type, gevolgd door kwantificering van hun relatieve lokalisatie, zal de aard en dynamiek van RE's bieden naast hun targeting op melanosomen. Dit is dus een eenvoudige techniek die kan worden gebruikt om de dynamiek van RE's in melanocyten te visualiseren en te meten.

Recycling-endosomen (RE's) zijn tubulair-vesiculaire organellen die worden gegenereerd uit vroege / sorterende endosomen in alle celtypen. Deze organellen spelen een sleutelrol in de biogenese van melanosomen, een lysosoom-gerelateerd organel geproduceerd door melanocyten. RE's leveren de melanocyt-specifieke lading aan premature melanosomen tijdens hun vorming. Blokkering bij het genereren van RE's, waargenomen bij verschillende mutanten van het Hermansky-Pudlak-syndroom, resulteert in hypopigmentatie van huid, haar en ogen. Daarom is het bestuderen van de dynamiek (zie aantal en lengte) van RE's nuttig om de functie van deze organellen in normale en ziekteomstandigheden te begrijpen. In deze studie willen we de RE-dynamiek meten met behulp van een resident SNARE STX13.

De zwartgekleurde organellen die je in de cellen ziet, worden melanosomen genoemd.Melanosomen staan bekend als een klasse van organellen genaamd lysozome gerelateerde organellen of LRO. Recycling-endosomen zijn buisvormige vesiculaire organellen van vroege of sorteer-endosomen in alle celtypen. Deze organellen spelen een sleutelrol in de biogenese van melanosomen, een lysozome-gerelateerd organel geproduceerd door melanocyten.Recyclage-endosomen leveren de melanocyt-specifieke lading aan premature melanosomen tijdens hun vorming. de generatie van recycling-endosomen waargenomen in verschillende syndroom is cellen in hyperpigmentatie van huid, haar en oog. Daarom is het bestuderen van de dynamiek van recycling-endosomen nuttig om de functie van deze organellen in normale en ziekteomstandigheden te begrijpen.Deze studie heeft tot doel de dynamiek van recycling-endosomen te meten met behulp van arresterende SNARE-syntaxin-13. De eerste stap is het zaaien van muizenmelanocyten op voorbehandelde coverslips. Bekleed de glazen afdekplaten in petrischaal in keldermembraanmatrixmedium.En droog het 15 minuten in de weefselkweekkap. Was de coverslips eenmaal met 1X PBS voor gebruik. Zaai de cellen op het keldermembraan medium gecoate coverslips op 50 tot 60% competentie.Voeg altijd 200 nanomol PMA toe aan de vergulde celsuspensie in volledige RPMI-media. Na het zaaien van de cellen, incubeer de schaal met cellen in CO2-incubator gedurende 12 tot 24 uur. De tweede stap is de transfectie van cellen met syntaxin-13 plasmiden.Incubeer de uiteinden met DNA en transfectiereagens, meng gedurende vijf minuten en meng zonder herhaaldelijk pipetteren. Tik elke tien minuten met de hand op de buis gedurende 30 minuten bij kamertemperatuur. Was de cellen tijdens de incubatie twee keer met 1X PBS, één keer met OPTI-MEM en voeg vervolgens 1 mil OPTI-MEM toe aan de cellen.Voeg na 30 minuten incubatie het transfectiereagens DNA-mengsel op een druppelsgewijze manier toe aan de cellen door de schaal te bedekken. Incubeer de cellen bij 37 graden Celsius gedurende zes uur. Zuig het OPTI-MEM medium aan met transfectiereagens en voeg een compleet RPMI medium toe aangevuld met 200 nanomol PMA.Incubeer de cellen bij 37 graden Celsius gedurende 48 uur. De derde stap is fixatie van de cellen. Houd er rekening mee dat de volgende procedure buiten de weefselkweekkap wordt uitgevoerd.Na 48 uur transfectie, was de cellen twee keer met 1X PBS en fixeer de cellen vervolgens met 3% formaldehyde gedurende 30 minuten. Was de cellen na fixatie twee keer met 1X PBS en bewaar de coverslips in 1X PBS tot verder gebruik. Als alternatief kunnen cellen op glasplaten worden gemonteerd of bij vier graden Celsius worden opgeslagen.De vierde stap is immunostaining van de cellen. Bereid een vochtige kamer voor. Leg het gesneden stuk parafilm op een muisfilterpapier in een petrischaaltje.Dek af met aluminiumfolie. Bereid 25 microliter primaire antilichaamoplossing voor. Voeg antilichaam toe bij een verdunning van één tot 200.Voeg deze oplossing toe als druppel op de parafilm in de vochtige kamer. Til de deklip voorzichtig op met een tang. Keer het om op de druppel primaire antilichaamkleuringsoplossing en bedek vervolgens het deksel van de vochtige kamer.Incubeer bij kamertemperatuur gedurende 30 minuten. Bereid op dezelfde manier de secundaire antilichaamoplossing bij een verdunning van één tot 500 en plaats deze op de parafilm naast de dekplaat in de vochtige kamer. Voeg voor het kleuren van de kern DAPI met een verdunning van 120.000 tot 130.000 toe aan de oplossing.Pak met behulp van een tang voorzichtig de coverslip van de primaire antilichaamoplossing en dompel deze twee keer in 1X PBS. Tik op de coverslip op tissuepapier om de overtollige PBS op de coverslip te verwijderen. Plaats het op de secundaire antilichaamkleuringsoplossing in de vochtige kamer en stel het niet bloot aan het licht, vanwege de aanwezigheid van fluorescerend geverfde antilichamen in de oplossing.Pak na de incubatie voorzichtig de coverslip van de secundaire antilichaamoplossing en dompel deze vervolgens twee keer in 1X PBS. Tik verder op de coverslip op tissuepapier om de overtollige PBS op de coverslip te verwijderen. Plaats 12 microliter beeldreagens op een glasplaat en plaats de gebrandschilderde afdekkingslip voorzichtig op het montagereagens.Keer de glasplaat om op het vloeipapier en druk vervolgens zachtjes aan. Stel vervolgens de coverslip af met behulp van een fluorescentiemicroscoop en analyseer de afbeeldingen met ImageJ. De zesde stap is de kwantificering van de overlap tussen de recycling endosome gelokaliseerde eiwitten en melanosomen.Kwantificering van syntaxin-13 delta-129 colocalisatie met de melanosomen. Immunofluorescentiemicroscopie van syntaxin-13 in wildtype melanocyten van muizen toonde GFP-syntaxin-13 wild-type gelokaliseerd als vesiculaire en buisvormige structuren. En GFP-syntaxin-13 delta-129 lokaliseert ringachtige structuren naast het celoppervlak.Verder vertoonde intracellulaire ringachtige GFP-syntaxin-13 delta-129 colocalisatie met het melanosoomeiwit TYRP1 in heldere veldbeeldige melanosomen. Zoals eerder getoond, wordt een cohort van overexpressie GFP-syntaxin-13 wild type waargenomen in melanosomen. Om de relatieve lokalisatie van GFP-syntaxin-13 wild type en GFP-syntaxin-13 delta-129 naar melanosomen te meten, werd Fiji-software gebruikt en analyse uitgevoerd met JACOP-plug-in.De gemeten Mander's overlapcoëfficiënt, dat is MOC, tussen GFP-syntaxin-13 delta-129 met TYRP1 is ongeveer 1,5 keer hoger in vergelijking met GFP-syntaxin-13 wild type met TYRP1. Interessant is dat TYRP1 2,9 keer hogere MOC-waarden liet zien met GFP-syntaxin-13 delta-129 in vergelijking met GFP-syntaxin-13 wild type. Deze gegevens geven aan dat de lokalisatie van GFP-syntaxin-13 delta-129 melanosomen relatief hoger is in vergelijking met GFP-syntaxin-13 wild type bij een steady state.Kwantificering van syntaxin-13 wild type lokalisatie van de recycling endosomen. Immunofluorescentiemicroscopie van GFP-syntaxin-13 wildtype toonde colocalisatie met het bekende recycling endosomale eiwit Rab11. De MOC tussen GFP-syntaxin-13 wild type met mCherry-Rab11 is ongeveer 1,4 keer hoger in vergelijking met mCherry-Rab11 met GFP-syntaxin-13 wild type.Om het aantal en de lengte van GFP-syntaxin-13 wildtype positieve endosomale tubuli te meten, werd Fiji-software gebruikt zoals beschreven in de protocolsectie. mCherry-Rab11 wordt gebruikt als een positieve controle in de experimenten. Melanocyten getransfecteerd met GFP-syntaxin-13 wild type vertoonden een hoger aantal tubuli per cel in vergelijking met cellen die mCherry-Rab11 tot expressie brachten.De tubuli worden echter verminderd bij co-expressie van zowel GFP-syntaxin-13 wild type als mCherry-Rab11 in de cellen. De zevende stap is de kwantificering van het buisvormige aantal en de lengte van recycling-endosomen. De tubuli worden gereduceerd bij coexpressie van zowel GFP-syntaxin-13 wild type als mCherry-Rab11 in de cellen.Interessant is dat de gemiddelde tubuluslengte van zowel GFP-syntaxin-13 wild type als mCherry-Rab11 vergelijkbaar is met elkaar in cellen die individueel of samen tot expressie komen. Samen suggereren deze gegevens dat GFP-syntaxin-13 wild type lokaliseert naar recycling endosomen als vergelijkbaar met Rab11. Deze studie toonde aan dat de N-terminale levering van syntaxin-13 mutant, dat wil zeggen GFP-syntaxin-13 delta-129, lokaliseert naar melanosomen GFP-syntaxin-13 delta-129 kan mogelijk worden gebruikt als een verslaggever voor vesiculaire handel van recycling endosomen naar het celoppervlak en LRO.Daarentegen lokaliseert GFP-syntaxin-13 wild type naar recycling endosomen vergelijkbaar met Rab11. Deze studie illustreerde dat GFP-syntaxin-13 wild type ook kan worden gebruikt voor het markeren van recycling endososomen in melanocyten. GFP-syntaxin-13 wild type kan een betere recycling endosomale marker zijn dan Rab11, omdat Rab11 de endosomale dynamiek verandert.Alles bij elkaar werkt GFP-syntaxin-13 wild type als een potentiële recycling endosomale marker om de dynamiek bij steady state-omstandigheden te bestuderen.
