Mensen en muizen hebben verschillende DC-subsets, waaronder klassieke DC's (cDC's, inclusief cDC1's en cDC2's), plasmacytoïde DC's (pDC's) en monocyten-afgeleide DC's (MoDC's)9,10,11. Het is algemeen aanvaard dat cDC1's cytotoxische T-lymfocyten (CTL) -reacties op intracellulaire pathogenen en kanker reguleren, en cDC2's reguleren immuunresponsen op extracellulaire pathogenen, parasieten en allergenen12....

Dendritische cellen (DC's) zijn de krachtigste antigeen-presenterende cellen (APC's) voor het activeren van naïeve T-cellen en het induceren van specifieke cytotoxische T-lymfocyten (CTL) reacties tegen infectieziekten, allergieziekten en tumorcellen 1,2,3. DC's zijn de primaire schakel tussen aangeboren immuniteit en adaptieve immuniteit en spelen een essentiële rol in immunologische afweer en het behoud van immuuntolerantie. In de afgelopen 40 jaar hebben veel onderzoekers geprobeerd de subsets van DC's en hun functies in ontsteking en immuniteit te definiëren. Volgens die studies ontwikkelen DC's zich langs de myeloïde en lymfoïde afstammingslijnen van beenmergcellen. Tumorvaccins hebben de afgelopen jaren belangrijke mijlpalen bereikt en hebben een veelbelovende toekomst. Mechanisch moduleren tumorvaccins de immuunrespons en voorkomen tumorgroei door cytotoxische T-lymfocyten te activeren met behulp van tumorantigenen. Het vaccin op basis van DC's speelt een belangrijke rol in tumorimmunotherapie en is geïdentificeerd als een van de meest veelbelovende antitumortherapieën 1,4. Bovendien zijn DC's op grote schaal gebruikt bij het testen van nieuwe moleculair gerichte geneesmiddelen en immuuncheckpointremmers5.
Onderzoekers hebben dringend een groot aantal zeer zuivere DC's nodig om de rol van DC's verder te bestuderen. DC's zijn echter zeldzaam in verschillende weefsels en bloed, goed voor slechts 1% van de bloedcellen bij mens en dier. In vitro kweek van beenmerg dendritische cellen (BMDC) is een belangrijke methode voor het verkrijgen van grote hoeveelheden DC-cellen. Ondertussen is het Lutz-protocol voor het genereren van DC's uit beenmerg veel gebruikt door onderzoekers6. Hoewel het protocol effectief is bij het verkrijgen van DC-cellen, is het complex en duur, waarbij hoge concentraties cytokines en de lysis van rode bloedcellen worden toegevoegd.
In deze studie rapporteren we een methode voor het isoleren van bijna alle beenmergcellen uit het beenmerg van muizen (BM) en het induceren van differentiatie in BMDC na 7-9 dagen incubatie in vitro, met een lagere concentratie GM-CSF en IL-4. Deze procedure duurt slechts 10 minuten om bijna alle beenmergcellen te oogsten en in een volledig medium op te hangen. Kortom, we bieden in dit onderzoek een efficiënte en kosteneffectieve kweekmethode voor BMDC.

De vraag naar dendritische cellen (DC's) neemt geleidelijk toe naarmate het immunologisch onderzoek vordert. DC's zijn echter zeldzaam in alle weefsels. De traditionele methode voor het isoleren van DC's omvat voornamelijk het induceren van beenmerg (BM) differentiatie in DC's door het injecteren van grote doses (>10 ng / ml) granulocyt-macrofaag koloniestimulerende factor / interleukine-4 (GM-CSF / IL-4), waardoor de procedure complex en duur wordt. In dit protocol werden, met behulp van alle BM-cellen gekweekt in 10 ng / ml GM-CSF / IL-4 medium, na 3-4 halve cultuuruitwisselingen, tot 2,7 x 107 CD11c + cellen (DC's) per muis (twee dijbenen) geoogst met een zuiverheid van 80% -95%. Na 10 dagen in cultuur nam de expressie van CD11c, CD80 en MHC II toe, terwijl het aantal cellen afnam. Het aantal cellen piekte na 7 dagen kweek. Bovendien duurde deze methode slechts 10 minuten om alle beenmergcellen te oogsten en werd een groot aantal DC's verkregen na 1 week cultuur.

De 1 x 10 7-1,7 x 107 cellen werden geëxtraheerd uit twee dijbenen en werden opnieuw gesuspendeerd in 24 ml medium voordat ze in een 6-well plaat werden geplant (figuur 1A). Na 2 dagen werden niet-hechtende cellen verwijderd door het kweekmedium volledig te veranderen. Met de toename van tumorimmuniteitsonderzoek neemt de vraag naar DC-cellen geleidelijk toe.DC's zijn echter zeldzaam in alle weefsels. De traditionele methode om voornamelijk de differentiatie van beenmerg in DC's te induceren, is ingewikkeld en kostbaar. Twee.
