研究者は、DCの役割をさらに研究するために、多数の高純度DCを緊急に必要としています。しかし、DCは様々な組織や血液ではまれであり、ヒトや動物の血球のわずか1%しか占めていません。骨髄樹状細胞(BMDC)の 体外 培養は、大量のDC細胞を得るための重要な方法である。一方、骨髄からDCを生成するためのLutzプロトコルは、研究者によって広く使用されています6。このプロトコルはDC細胞を得るのに有効であるが、複雑で高価であり、高濃度のサイトカインの添加および赤血球の溶解を伴う。
本研究では、マウス骨髄(BM)からほぼすべての骨髄細胞を単離し、GM-CSFおよびIL-4の濃度を低くして、 インビトロでの7~9日後のBMDCへの分化誘導方法を報告する。この手順は、ほとんどすべての骨髄細胞を採取し、それらを完全な培地に懸濁するのに10分しかかかりません。簡単に言えば、我々はこの研究においてBMDCのための効率的かつ費用対効果の高い培養方法を提供する。
